The UK human anthrax vaccine consists of the alum-precipitated culture supernatant of Bacillus anthracis Sterne. In addition to protective antigen (PA), the key immunogen, the vaccine also contains a number of other bacteria- and media-derived proteins. These proteins may contribute to the transient side effects experienced by some individuals and could influence the development of the PA-specific immune response. Bacterial cell-wall components have been shown to be potent immunomodulators. B. anthracis expresses two S-layer proteins, EA1 and Sap, which have been demonstrated to be immunogenic in animal studies. These are also immunogenic in man so that convalescent and post-immunisation sera contain specific antibodies to Ea1, and to a lesser extent, to Sap. To determine if these proteins are capable of modifying the protective immune response to PA, A/J mice were immunised with equivalent amounts of recombinant PA and S-layer proteins in the presence of alhydrogel. IgG isotype profiles were determined and the animals were subsequently challenged with spores of B. anthracis STI. The results suggest that there was no significant shift in IgG isotype profile and that the presence of the S-layer proteins did not adversely affect the protective immune response induced by PA. 相似文献

Under competitive conditions, stem elongation in plants is thought to enhance fitness by increasing light interception. However, the onset of competition should vary with the species of competitor due to interspecific differences in timing of emergence and plant growth form. The fitness benefits of elongation may therefore depend on the timing of this plastic response. Phenotypic selection analyses and path analysis were used to evaluate selection acting on stem elongation at early and late life-history stages and the combination of germination timing and elongation in an annual plant. Velvetleaf (Abutilon theophrasti) were raised in one of three environments experienced by natural populations (cornfields; soybean fields; and disturbed, weedy sites). Due to the rapid growth rate and high density of plants in disturbed areas, selection to increase seedling-stage elongation was expected in weedy sites. Due to the wide spacing of crop plants, competition for light is initially low in cultivated fields, but intensifies as the season progresses. Selection for increased elongation at later nodes was expected in soybean fields because velvetleaf can often overtop soy and thereby increase leaf exposure. In contrast, selection against late elongation was expected in cornfields because velvetleaf are incapable of overtopping corn. Individuals that elongate would experience the carbon cost of allocating to structural tissue, but fail to experience a carbon return through increased light interception. The phenotypic selection analyses were consistent with these predictions and therefore support the role of stem elongation as an adaptation to interspecific competition. Selection also acted on the combination of germination timing and elongation. In the weedy environment, early emergence in conjunction with enhanced stem elongation conveyed the highest fitness. Reduced elongation was favored among individuals that emerged late, potentially because these individuals were unable to overtop neighbors. The results of this study demonstrate that the timing of stem elongation strongly affects competitive success. Environments that differ in the timing of competition for light select for elongation at different life-history stages, and this selection depends on the timing of emergence. 相似文献

A problem that frequently occurs in biological experiments with laboratory animals is that some subjects are less susceptible to the treatment than others. A mixture model has traditionally been proposed to describe the distribution of responses in treatment groups for such experiments. Using a mixture dose-response model, we derive an upper confidence limit on additional risk, defined as the excess risk over the background risk due to an added dose. Our focus will be on experiments with continuous responses for which risk is the probability of an adverse effect defined as an event that is extremely rare in controls. The asymptotic distribution of the likelihood ratio statistic is used to obtain the upper confidence limit on additional risk. The method can also be used to derive a benchmark dose corresponding to a specified level of increased risk. The EM algorithm is utilized to find the maximum likelihood estimates of model parameters and an extension of the algorithm is proposed to derive the estimates when the model is subject to a specified level of added risk. An example is used to demonstrate the results, and it is shown that by using the mixture model a more accurate measure of added risk is obtained. 相似文献

In plants, the ratio of red to far-red wavelengths (R:FR) reliably indicates neighbor proximity and influences stem elongation. Enhanced elongation increases light interception and fitness under crowded conditions. However, many environmental factors vary simultaneously such that responses to R:FR may be affected by abiotic conditions or maternal environmental conditions. This study examines the effects of temperature, photoperiod, and maternal environment on stem-elongation responses to R:FR. Four populations of Abutilon theophrasti (two from disturbed, weedy areas and two from cornfields) were used in factorial common-garden experiments of temperature × R:FR × population and photoperiod × R:FR × population. Seedling growth of greenhouse- and field-derived seed was compared to evaluate maternal effects. Maternal environment did not alter seedling elongation. Higher temperatures resulted in both a twofold increase in average elongation and increased responsiveness to R:FR. Significant three-way interactions in both experiments demonstrate that population responses to R:FR differ depending on temperature and photoperiod conditions. These results indicate that elongation responses to R:FR are more variable than previously realized. The observed variability in elongation also suggests that the outcome of competitive interactions in the natural environment will depend on ambient temperature, photoperiod length, and population origin. 相似文献

Interactive effects of low atmospheric CO2 and elevated temperature on growth, photosynthesis and respiration in Phaseolus vulgaris 总被引:2,自引:0,他引:2
For most of the past 250 000 years, atmospheric CO2 has been 30–50% lower than the current level of 360 μmol CO2 mol–1 air. Although the effects of CO2 on plant performance are well recognized, the effects of low CO2 in combination with abiotic stress remain poorly understood. In this study, a growth chamber experiment using a two-by-two factorial design of CO2 (380 μmol mol–1, 200 μmol mol–1) and temperature (25/20 °C day/night, 36/29 °C) was conducted to evaluate the interactive effects of CO2 and temperature variation on growth, tissue chemistry and leaf gas exchange of Phaseolus vulgaris. Relative to plants grown at 380 μmol mol–1 and 25/20 °C, whole plant biomass was 36% less at 380 μmol mol–1× 36/29 °C, and 37% less at 200 μmol mol–1× 25/20 °C. Most significantly, growth at 200 μmol mol–1× 36/29 °C resulted in 77% less biomass relative to plants grown at 380 μmol mol–1× 25/20 °C. The net CO2 assimilation rate of leaves grown in 200 μmol mol–1× 25/20 °C was 40% lower than in leaves from 380 μmol mol–1× 25/20 °C, but similar to leaves in 200 μmol mol–1× 36/29 °C. The leaves produced in low CO2 and high temperature respired at a rate that was double that of leaves from the 380μmol mol–1× 25/20 °C treatment. Despite this, there was little evidence that leaves at low CO2 and high temperature were carbohydrate deficient, because soluble sugars, starch and total non-structural carbohydrates of leaves from the 200μmol mol–1× 36/29 °C treatment were not significantly different in leaves from the 380μmol mol–1× 25/20 °C treatment. Similarly, there was no significant difference in percentage root carbon, leaf chlorophyll and leaf/root nitrogen between the low CO2× high temperature treatment and ambient CO2 controls. Decreased plant growth was correlated with neither leaf gas exchange nor tissue chemistry. Rather, leaf and root growth were the most affected responses, declining in equivalent proportions as total biomass production. Because of this close association, the mechanisms controlling leaf and root growth appear to have the greatest control over the response to heat stress and CO2 reduction in P. vulgaris. 相似文献

We present a design to quantify fitness consequences of jasmonate-induced responses in plants that are competing for limited resources with a conspecific. Under both high and low nitrogen supply rates, uninduced (control) Nicotiana attenuata plants growing next to a plant induced with 250 μg methyl jasmonate (MJ) yielded more seed capsules than control plants competing with another control plant. We conclude that there is a opportunity benefit for control plants growing next to an induced plant. Initially, MJ-induced plants grew more slowly, but by senescence they had produced the same number of seed capsules as control plants that had competed with another control plant. Replacement series showed that the fitness of MJ-induced plants is not influenced by the competitive status of their neighbour plant. We argue that competitive designs are useful tools for evaluating the phenotypic costs of ecologically important traits. 相似文献
