We sequenced 512 nucleotides in two variable regions of the N gene of 23 rabies isolates from the northeastern part of Thailand by direct sequencing of PCR-amplified products. The sequencing data revealed two new lineages in these rabies isolates. Based on the results of this study together with the findings of our earlier study, the rabies isolates in Thailand were divided into two genogroups, designated as T1 and T2, which were predominantly localized in the northern and northeastern areas, respectively. Each of these two genogroups consisted of four lineages. There was a correlation between these eight lineages and the geographical origins of the isolates. Two lineages belonging to the T2 genogroup from the northeastern area of Thailand were newly identified in this study. The isolates in both genogroups were also prevalent in the central area of Thailand. Each lineage in the T1 and T2 genogroups was found independently in dogs in the upper and lower southern areas of Thailand, respectively. These genetic data and the historical background suggest that rabies viruses belonging to the T2 genogroups were prevalent many years ago in the central and northeastern areas of Thailand and were later transferred to the lower southern parts of Thailand. 相似文献

Changes in plant species richness over the last century in the eastern Swiss Alps: elevational gradient, bedrock effects and migration rates 总被引:2,自引:0,他引:2
Areas of 2,800–3,000 m a.s.l. represent the alpine-nival ecotone in the Alps. This transition zone connecting the closed swards
of the alpine belt and the scattered vegetation of the nival belt may show particularly strong climate warming driven fluctuations
in plant species richness compared to the nival belt. To test this hypothesis, 12 summits within this range were investigated
in the canton of Grisons, Switzerland in 2004. Complete lists of vascular plant species consisting of 5–70 species were collected
on each summit and the elevation of the uppermost occurrence of each species was recorded. These data were compared to historical
records over 120 years in age. Within this time, vascular plant species richness increased by 11% per decade on summits in
the alpine-nival ecotone. Despite this considerable change, a comparison with nival summits did not support the hypothesis
that species richness increase at the alpine-nival ecotone is higher than in the nival belt. A general trend of upward migration
in the range of several metres per decade could be observed. Anemochorous species were more often found to be migrating than
zoochorous or autochorous species and migration was higher on calcareous than on siliceous bedrock. A comparison between the
summits with the adjacent slopes in our study revealed that changes in species number could be used as an indicator for climate-induced
changes—if at all—only for the narrow summit areas. 相似文献

选取缙云山针阔混交林、常绿阔叶林、楠竹林和灌木林作为研究对象,对4种林分的凋落物储量和营养元素释放量等进行观测,并应用室内模拟酸雨实验对4种林分凋落物进行淋溶模拟。结果表明:缙云山各林分现存凋落物厚度为1.4~4.5 cm,具有明显的分层结构;林分未分解U层、半分解S层和分解D层现存凋落物量分别为1.97~8.59、2.84~10.18和3.58~17.43 t·hm-2,林分年凋落物量为2.17~9.86 t·hm-2·a-1,凋落物总储量为14.21~32.42 t·hm-2,凋落物分解常数为0.15~0.31,林下凋落物分解95%时所需时间针阔混交林和楠竹林均在10年以上,凋落物分解速率比较缓慢;林下凋落物层营养元素含量以Ca、N为主,Fe、K、Mg次之;凋落物总的营养元素释放率表现为常绿阔叶林(0.80)>灌木林(0.72)>针阔混交林(0.50)>楠竹林(-0.17);与叶片相比,凋落物中N、K、Mn 3种营养元素含量明显降低;为探明酸雨影响营养元素循环的作用机理,对模拟酸雨的离子含量与凋落物淋滤液盐基离子含量进行了分析,其相关性大小表现为楠竹林(相关系数0.895)>针阔混交林(0.826)>灌木(0.700)>常绿阔叶林(0.699),楠竹林凋落物营养元素的淋滤受酸雨影响最大,常绿阔叶林受其影响最弱;推测在这一过程中以凋落物的吸附作用为主。 相似文献

The exceptional species diversity of flowering plants, exceeding that of their sister group more than 250-fold, is especially evident in floral innovations, interactions with pollinators and sexual systems. Multiple theories, emphasizing flower–pollinator interactions, genetic effects of mating systems or high evolvability, predict that floral evolution profoundly affects angiosperm diversification. However, consequences for speciation and extinction dynamics remain poorly understood. Here, we investigate trajectories of species diversification focusing on heterostyly, a remarkable floral syndrome where outcrossing is enforced via cross-compatible floral morphs differing in placement of their respective sexual organs. Heterostyly evolved at least 20 times independently in angiosperms. Using Darwin''s model for heterostyly, the primrose family, we show that heterostyly accelerates species diversification via decreasing extinction rates rather than increasing speciation rates, probably owing to avoidance of the negative genetic effects of selfing. However, impact of heterostyly appears to differ over short and long evolutionary time-scales: the accelerating effect of heterostyly on lineage diversification is manifest only over long evolutionary time-scales, whereas recent losses of heterostyly may prompt ephemeral bursts of speciation. Our results suggest that temporal or clade-specific conditions may ultimately determine the net effects of specific traits on patterns of species diversification. 相似文献

Wheat is one of the basic food materials for humans and other animals. Continuing studies into breeding new species, which are suitable for agriculture, are based on the contention that the restricting factors are the effects of the nuclear genes. Such focusing of studies on the nuclear genome leads to restriction on the information on organelle DNAs, although cytoplasmic genetic factors are effective on the morphological, physiological and reproductive features of plants. Restriction fragment length polymorphism analysis (RFLP) of chloroplast DNA (cpDNA) has become a powerful tool for studying phylogenetic relationships. In this study, seven wheat species, belonging to Aegilops and Triticum , were studied. An intergenic spacer region of cpDNA was digested by six different restriction endonuclease enzymes, and fragments so obtained were investigated using the agarose gel electrophoresis technique. Fragments of genes of the “green-like” form I ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBisCO) of eight species of haloalkaliphilic obligately autotrophic sulfur-oxidizing bacteria of the genus Thioalkalivibrio have been revealed and sequenced using previously developed oligonucleotide primers. The data obtained are used for the construction of phylogenetic trees on the basis of nucleotide sequences of RuBisCO genes and their conceptual translations into amino acid sequences. Comparative analysis of the 16S rRNA and RuBisCO gene trees reveals discrepancies between their topologies. According to a RuBisCO gene analysis, the genus Thioalkalivibrio is not monophyletic, and its inner divergence conforms to the significant morphological differences observed between the species. 【目的】本研究将推测的阿维链霉菌(Streptomyces avermitilis)脂肪酶基因lpsA2在大肠杆菌(Escherichia coli)中进行异源表达及系统的酶学性质分析。【方法】提取阿维链霉菌基因组,设计特异性引物,PCR扩增脂肪酶基因lpsA2,使其在大肠杆菌中异源表达,利用6个组氨酸标签纯化脂肪酶LpsA2,并进行酶学性质分析;对LpsA2进行序列比对和进化分析。【结果】氨基酸序列比对显示LpsA2具有脂肪酶典型的由Ser、His和Asp构成的活性部位,即Ser130-Asp221-His25,其中Ser位于保守的五肽结构(Gly128-His129-Ser130-Gln131-Gly132)中;分子系统学分析显示,LpsA2是脂肪酶第一家族亚家族成员(Subfamily I.7);实验测得纯化的重组脂肪酶LpsA2的最适反应pH为8.0,最适反应温度为50℃;最适底物为对硝基苯酚豆蔻酸酯;在10℃-50℃范围内该酶的激活自由能为6.3 kcal/mol;1 mmol/L Co2+、Hg2+、Zn2+可使酶活性提高至250%以上;15%的二甲基甲酰胺和二甲基亚砜使酶活分别提高至110.7%和138%;0.1%和1%的Span-20可使酶活性分别提高至352.7%和189.7%。【结论】本研究对推测的来源于S.avermitilis的脂肪酶基因lpsA2进行了异源表达和酶学功能鉴定,不仅为脂肪酶的研究积累了更多数据,也为具有优良性能的脂肪酶生物工程菌的筛选奠定了基础,更为其在食品加工、药物合成等工业生产中的应用提供了依据。 相似文献
