In green algae several characteristic differences in the slope of the fast 685 nm fluorescence transient indicate the existence of different mechanisms for the regulation of the photosynthetic electron transport in vivo with respect to the requirements for ATP and NADPH. Autotrophically cultivated Chlamydobotrys stellata exhibits a normal time curve of the fluorescence yield. Anaerobiosis and C02-deficiency raise the O-, I- and S-level, whereas the P- level is lowered and the I-D-decay disappears. The readdition of oxygen increases the fluorescence significantly. Supplementation of aerobic cells with CO2 restores the normal fluorescence transients. The replacement of carbon dioxide by acetate as a carbon source in the light lowers the overall fluorescence emission and abolishes the D-P-increase and the P-S-decline. The presence of DCMU increases fluorescence only at high intensities of incedent light. Anaerobiosis in these photoheterotrophic algae lowers the fluorescence emission. In this case DCMU increases fluorescence even at low light intensities. In Gonium multicoccum, which shows a normal fluorescence transient when cultivated autotrophically, CO2-deficiency abolishes the O-level and increases the I- and S-niveau. Additional anaerobiosis in CO2-deficient cells raises the steady state emission. Readdition of oxygen to these cells raises the I- and S-level even more and prevents the build up of the P-level. Summary The results of a monoamine-fluorescence study of the hypothalamus of Rana temporaria show that the brain area corresponding with the nucleus infundibularis dorsalis (NID), as described in other species, does not differ, neither morphologically nor histochemically, from the paraventricular organ (PVO), with which it is anatomically continuous. It is concluded that a nucleus infundibularis dorsalis does not exist as a separate entity in this species.  相似文献

The mechanism of photosynthetic carbon dioxide fixation in the green flagellate Dunaliella tertiolecta Butcher varies during growth in batch culture. Evidence for this change comes from three sources: i) algae from the stationary phase incorporated a greater proportion of the fixed carbon into amino arids and protein than did cells from the mid-exponential phase; ii) the activity of phosphoenolpyruvate carboxylase relative to that of ribulose-1, 5-di-phosphate carboxylase increased with age in batch culture; and, iii) cells from the stationary phase appeared to utilize the bicarbonate ion as the substrate for photosynthesis, whereas those from mid-exponential phase appeared to utilize fire carbon dioxide. These data suggest that a change of photosynthetic mechanism can occur within a single species of alga, depending on its physiological state.  相似文献

Light absorption by two green seaweeds with similar photophysiology but different anatomies are compared: i) Ulva lactuca var. rigida (C. Ag.) Le Jolis, an optically translucent species of two cell layers both bearing chloroplasts; and, ii) Codium fragile subsp. tomentosoides (van Goor) Silva, an optically opaque species with a colorlelss medulla surrounded by a cortex of choloroplast-bearing utriclels. Thallus absorptance (fraction of incident light absorbed) was measured for various pigment contents. Absorptance by U. lactuca was dependent on pigment concentration in an exponential manner and never exceeded 0.6, whereas absorptance by C. fragile was independent of pigment concentration and always approached a balue of 1.0. Water in the medullary tissue of C. fragile is often of the utricles. The utricles appear to be “integrating spheres” enhancing the capture of incident light, aided by the wave-guide function of the thin peripheral layer of cytoplasm and a reflector function at their base. Photosynthitic performance for U. lactuca saturates at high light intensities and attenuates rapidly with decreasing intensities. In contrast, photosynthetic performance for C. fragile saturates at low light intensities and attenuates slowly with diminishing radiation. Extrapolated diel variation in photosynthesis shows that U. lactuca's anatomy is adaptive for high light intensity environments, whereas C. fragile's anatomy is adaptive for low light intensity environments. Both seaweeds fit into the ecological category of “fugitive” species, and compete in the Long Island Sound (Atlantic Ocean) rocky intertidal for free-space. Predictions are presented for relative species abundances along a monotonic gradient of light intensity.  相似文献

The formation of chlorophyll, cytochrome f, P-700, ribulose bisphosphate carboxylase as well as photosynthesis and Hill reaction activities were tested during the light-dependent greening process of the Chlorella fusca mutant G 10. Neither chlorophyll nor protochlorophyllide was detected in the darkgrown cells. When transferred to light the mutant cells developed chlorophyll and established its photosynthetic capacity after a short lag phase. In the in vivo absorption spectra a spectral shift of the red absorption peak position from 674 to 680 nm was indicated during the first 3 h of greening. Cytochrome f was already present in the dark-grown cells, but during the greening phase a threefold increase in the cytochrome f content could be seen. At the early stages of greening a characteristic primary oscillation in the content of cytochrome f was observed. P-700 was lacking in the dark and during the first 30 min of illumination. From the first to the second h of light a forced synthesis of P-700 took place and the time-course curve for the ratios of P-700/chlorophyll rose to a sharp maximum. The synthesis of P-700 started together with photosystem I activity and showed similar kinetics. We found the simultaneous appearance of photosystem II, photosystem I, and photosynthetic activities 30 min after the beginning of the illumination. Based on chlorophyll content they attained maximum activity after 2 h of light, but at this time photosystem I capacity proved to be remarkably higher than photosynthetic and photosystem II activities. Highest carboxylase activity existed in darkgrown cells. During the greening process the activity of the enzyme decreased continuously. Whole homogenates and membrane-bound and cytosoluble fractions prepared from rat ventral tegmental area (VTA) and nucleus accumbens were examined for their content of peptidasic activities and for their ability to metabolize neurotensin and its natural related hexapeptide neuromedin N. No qualitative differences were observed between these two brain regions concerning the presence and the subcellular distribution of a series of activities able to hydrolyze various specific fluorimetric enzymatic substrates. However, aminopeptidase B, endopeptidase 24-15, and endopeptidase 24-11 were significantly lower in the VTA than in the nucleus accumbens membrane preparations, while proline endopeptidase was detected in significantly higher amount only in the cytosolic fraction prepared from nucleus accumbens. Both neurotensin and neuromedin N were metabolized more rapidly in the nucleus accumbens than in the VTA. Furthermore, the degradation rate of neuromedin N was considerably faster than that of neurotensin whatever the cerebral area examined. Studies carried out with highly specific peptidase inhibitors revealed that endopeptidase 24-15 mainly contributed to the catabolism of neurotensin in homogenates and membrane-bound preparations of nucleus accumbens and VTA, while aminopeptidase B appeared predominantly responsible for the rapid disappearance of neuromedin N in both cerebral tissues. The possibility that the different metabolic processes of the two peptide congeners could explain their distinct pharmacological profiles observed after their microinjection in the nucleus accumbens and in the VTA is discussed.  相似文献

We studied levels of tyrosine hydroxylase immunoreactivity and phosphorylation state in the ventral tegmental area (VTA) and nucleus accumbens (NAc) in an effort to understand better the mechanisms by which these brain reward regions are influenced by opiates and cocaine. In the VTA, chronic, but not acute, administration of either morphine or cocaine increased levels of tyrosine hydroxylase immunoreactivity by 30-40%, with no change observed in the relative phosphorylation state of the enzyme. In the NAc, chronic, but not acute, morphine and cocaine treatments decreased the phosphorylation state of tyrosine hydroxylase, without a change in its total amount. In contrast, morphine and cocaine did not regulate tyrosine hydroxylase in the substantia nigra or caudate/putamen, brain regions generally not implicated in drug reward. Morphine and cocaine regulation of tyrosine hydroxylase could represent part of a common biochemical basis of morphine and cocaine addiction and craving.  相似文献

Summary The in-vitro effects of hydroxyurea 5-FU and 5-FUdR have been extensively studied in experimental systems employing cell-line techniques. In this study we investigated the effects of these drugs on the levels of incorporation of labeled nucleosides into DNA in explants of intact rat colonic mucosa maintained in organ culture. The effects of the nucleoside transport inhibitors nitrobenzylthioinosine (NBMPR) and dipyridamole—which are modulators of antimetabolite cytotoxicity—on the incorporation of tritiated thymidine [(3H]TdR) into DNA were also studied. The incorporation of tritiated TdR into DNA was reduced by hydroxyurea but was not altered by either 5-FU or 5-FUdR. The levels of tritiated deoxyuridine were reduced by 5-FU and 5-FUdR in separate experiments; this is in keeping with thymidylate synthase inhibition. NBMPR and dipyridamole also reduced 3H-TdR incorporation into DNA. These results can be explained in terms of the known mechanisms of action of these drugs. This experimental model is therefore useful in assessing the effects of antimetabolites and nucleoside transport inhibitors in intact colonic mucosa.  相似文献
