Several specific inhibitors for plasminogen activators have been isolated from various organs and cell lines, those from human placenta and the human monocyte-like cell line U-937 being virtually identical. The reaction between this type of inhibitor, designated as type-2, and high-Mr and low-Mr urokinase-type plasminogen activators was followed by reversed-phase high-performance liquid chromatography and gel electrophoresis. The components, their stable complexes and their dissociation and cleavage products could be clearly identified in both systems. The amino acid sequence of the inhibitor at the cleavage site was determined to be -Met-Thr-Gly-Arg↓Thr-Gly-His-Gly-. A 35-residue carboxy-terminal fragment was found to be released. 相似文献

We propose that the unique temperature dependence of the Chance-De Vault cytochrome oxidation reaction in Chromatium is not due to a transition from low-temperature nuclear tunnelling to a high-temperature activated electron transfer (ET), but rather originates from two parallel ET processes from two distinct low-potential cytochromes to the bacteriochlorophyll dimer cation. These involve a slow activationless process, which dominates at low temperatures (T 120 K) and an activated process, which is practically exclusive at high temperatures. Three cases of abnormal expression of the equine protease inhibitory alleles, Pi F, L, and S1, were observed following the examination of 30,000 plasma samples by one-dimensional acid (pH 4.6) polyacrylamide gel electrophoresis. Characterization of the abnormal proteins in terms of isoelectric point, molecular mass, inhibitory spectra, and sialic acid content was performed using one- and two-dimensional electrophoretic techniques. The Pi F and S1 abnormalities were postulated to be the result of amino acid substitutions causing alterations in the processing of the carbohydrate side chains. No explanation could be offered for the Pi L abnormality other than a charge shift mutation. Abnormal types, F*, L*, and S*1 behaved as alleles but the distribution of L* in offspring from one stallion (present in only 6 of 83 offspring) differed significantly from expectation.This work was supported by a grant from the Australian Stud Book, Alison Road, Randwick, N.S.W. 2031. 相似文献

A transient CO2 burst from seedlings of some plant species was observed after a rapid temperature decrease. The magnitude of the CO2 release depended on initial temperature, oxygen concentration and light intensity. To obtain a maximal value of CO2 release, the temperature had to decrease by more than 8°C. The phenomenon was detected only in the light, and was confined to C3 species. It was inhibited by low oxygen concentration, indicating its possible connection with photorespiration. 相似文献

Non-structural storage carbohydrates were measured in 9-day-old barley ( Hordeum vulgare L. cv. Brant) primary leaves. Accumulation rates of starch, sucrose and total non-structural carbohydrates (TNC) were approximately linear when measured between 2- and 12-h of light. Progressively higher TNC accumulation rates were observed at higher irradiance levels (i.e., comparing 250, 550 and 1050 ·mol m−2 s−1 ). Synthesis of a low-molecular-weight fructan also was enhanced by high irradiances. Low irradiance treatments decreased leaf sucrose levels and there was a corresponding increase in the lag period preceding starch synthesis in the light. Increased starch accumulation rates were usually observed when sucrose concentrations were high. These and other results suggested that cytosolic sucrose concentrations affected starch metabolism in the chloroplast. However, sucrose accumulation rates increased and starch storage decreased when barley seedlings were transferred from 20 to 10°C during the light period. Lowering the night temperature from 20 to 10°C for a single dark period 8-days after planting increased the TNC content of barley primary leaves at the beginning of day nine. In this experiment, TNC accumulation rates of treated and untreated leaves were similar. Changes in the accumulation rate of TNC were usually observed within 2- to 4-h after barley seedlings were exposed to altered environmental conditions. Monitoring rapid changes in leaf carbohydrate levels is a sensitive method for assessing the effects of environmental treatments on photosynthetic metabolism. 相似文献

Summary A pot experiment with lettuce involving three N forms each at six application levels, showed that lettuce can be grown satisfactorily
with a very low nitrate content when supplied with ammonium sulphate and a nitrification inhibitor. For plants growing on
nitrate N, the optimum midrib sap nitrate concentration as maturity approached was about 1400 mg/1 NO3-N. Large losses of mineral N were observed from the peat medium, even in the absence of plants.
A relationship is presented which would enable a lettuce grower to estimate whole-shoot nitrate concentration from a quick
test of midrib sapi.e. NO3-N (mg/kg in fresh shoot) =0.14×NO3-N (mg/l in sap).
Tipburn was worst at intermediate levels of applied N, and was less serious with pure ammonium nutrition than with nitrate. 相似文献

Summary In order to improve nitrogen recovery by rice, the effect of a urease inhibitor phenylphosphorodiamidate (PPD) on the efficiency
of fertilizer urea was studied in laboratory and greenhouse. Addition of PPD to urea (5% w/w) delayed urea hydrolysis by 3
to 4 days and reduced ammonia volatilization from 45% (without PPD) to 8.5% (with PPD). Ammonia volatilization obeyed first
order kinetics. Urea hydrolysis was sufficiently strongly inhibited to match the nitrification potential of the soil. N application
to rice by three different modes showed that a delayed mode (4 splits) was superior to two conventional modes (3 splits) in
nitrogen recovery and fertilizer efficiency since it met nitrogen requirement of plants at reproductive stage. In 2 out of
3 modes of application, there was a 14% increase (relative) in grain yields and dry matter, and 6.8% increase in N uptake
efficiency on application of PPD along with urea. The results indicate that urease inhibitors like PPD can be effectively
used to block urea hydrolysis, reduce ammonia volatilization losses and improve N use efficiency by rice. 相似文献

Extracellular Fluid Proteins of Goldfish Brain: Evidence for the Presence of Proteases and Esterases 总被引:1,自引:1,他引:0
Preparations of enriched fractions of extracellular fluid (ECF) proteins from goldfish brain were found to contain protease(s) and esterase(s). The N-substituted furanacryloyl (FA) peptides FA-Phe-Gly-Gly and FA-Phe-OMe were used as model substrates for determining protease and esterase activity, respectively, in a spectrophotometric assay. Studies of the profile of substrate specificity and identification of the types of compounds that were effective as inhibitors showed that these ECF enzymes have some distinctive properties. GSH, but not GSSG, and EDTA inhibited the protease(s) without influencing the esterase(s), whereas L-1-tosylamide-2-phenylethylchloromethyl ketone blocked both protease and esterase activities of ECF. Most of the protease and esterase properties of ECF could be bound to concanavalin A-Sepharose affinity chromatographic columns in association with ependymin--a brain extracellular protein. These observations indicate that ECF may contain a metalloprotease(s) and raise the possibility that the ependymins might be a substrate for these ECF enzymes. 相似文献
