A teichoic acid degrading enzyme (teichoicase) was purified to apparent homogeneity from a water-soluble cell extract of sporulating Bacillus subtilis cells. A rapid test for the detection of teichoicase activity was developed. The purified teichoicase has an app. Mr = 310 000. It consists of 4 identical subunits of Mr = 78 000 each.  相似文献

Abstract The rate of incorporation of 14C from CO2 into glycerol, and the amount of glycerol in cells of Dunaliella tertiolecta was determined at constant salinity at two representative concentrations of NaCl. From these data, the rate of synthesis and turnover of the glycerol pool was determined. The half-time for turnover of the glycerol pool was of the order of 1 h in 170 mol m?3 NaCl and almost 6h for 700 mol m?3 NaCl. These results indicate that turnover of the glycerol pool in Dunaliella is relatively slow under steady-state conditions. Synthesis and dissimilation of glycerol do not apparently constitute a metabolic cycle in the conventional sense. Rather, glycerol metabolism resembles that of the storage polysaccharides which arc commonly produced and degraded by different pathways.  相似文献

The amount of phospholipids in the membrane fractions of the endoplasmic reticulum (ER) and plasma membrane of cotyledons of Pisum sativum and P. elatius was followed during germination. Incorporation of [14C-methyl]choline into the ER and the plasma membrane was followed as well as [U-14C]glycerol into the ER. The pool size of endogenous choline was determined and found to be much greater in the wild pea and to increase early in germination. In both species membranes are synthesized in large quantities early in germination and both turnover and synthesis of the backbone and the phosphatidyl tail occur throughout 48 hr of germination. In P. sativum incorporation peaks earlier than in P. elatius and degradation also begins earlier than in P. elatius. This is consistent with the general behaviour of the two species.  相似文献

Abstract Diapause larvae of the European corn borer (Ostrinia nubilalis (Hubn.)) and the related Mediterranean noctuid Sesamia cretica Led. possess sufficient supercooling ability to avoid freezing over their normal environmental temperature ranges. In progressive chilling experiments (10 days acclimation at each 5° step in the temperature range from 15 to ?5°C), mean supercooling points (measured at a cooling rate of 0.1°C min?1) were lowered from ?20.4°C at 15°C to ?24.0°C at 5°C (lower lethal temperatures: c.?28°C) in O.nubilalis, compared with ?15.0 to ?17.2°C (lower lethal temperatures: ?15 to ?17°C respectively) in S.cretica. Concentrations of glycerol and trehalose determined by gas chromatography of whole body extracts were consistently higher in the former than in the latter species at both 15 and 5°C, and may be responsible for the deeper supercooling in O.nubilalis larvae. Acclimation to 5°C increased glycerol levels in O. nubilalis extracts compared with 15°C, and this was enhanced in larvae exposed for a further 10 days at each of 0 and ?5°C (glycerol being 438μmol ml?1 body water). Haemolymph glycerol concentrations showed a similar pattern to whole body extracts in this species. Fat body glycogen was reduced during low temperature acclimation in both species. Body water contents did not change with acclimation in O.nubilalis, whilst S.cretica, containing significantly more water, lost c.7% during acclimation from 15 to 5°C. Haemolymph osmolalities increased during acclimation, especially in Ostrinia larvae, probably as a result of the accumulation of cryoprotectants. The majority of O.nubilalis larvae survived freezing under the conditions of the cooling experiments, whilst larvae of S.cretica did not, thereby confirming an element of freezing tolerance in the former.  相似文献

Freeze preservation of human and nonhuman semen has been used effectively for a number of years; however, the application of freezing to preserve nonhuman primate sperm has been less successful. This study compares five freeze methods and various concentrations of the cryoprotectants glycerol and dimethylsulfoxide (DMSO) for cryopreservation of chimpanzee (Pan troglodytes) sperm. The different methods were compared using quantitative analysis of sperm function and, by an indirect measure of fertilizing capacity, the denuded hamster oocyte penetration assay (SPA). The best results were obtained using an extender comprising Ham's F10 medium containing 15% heat inactivated human cord serum and 15.6% glycerol. Semen extended 1 to 1 by this method, for a final glycerol concentration of 7.8%, and frozen using a computer programmed freezer maintained approximately 65% of its original viability and up to 70% of its initial motility. The extended semen was used to initiate pregnancy in two female chimpanzees.  相似文献

Photosensitized efficacy of tetrasulfonated phthalocyanines of zinc, aluminum and nickel (ZnPcS4, AlPcS4 and NiPcS4, respectively) as studied by gramicidin channel (gA) photoinactivation was compared with adsorption of the dyes on the surface of a bilayer lipid membrane as measured by the inner field compensation method. The adsorption of the negatively charged phthalocyanines on diphytanoylphosphatidylcholine (DPhPC) membranes led to formation of a negative boundary potential difference between the membrane/water interfaces. Good correlation was shown between the photodynamic activity and the membrane binding of the three metallophthalocyanines. ZnPcS4 appeared to be the most potent of these photosensitizers, while NiPcS4 was completely ineffective. All of these phthalocyanines displayed no binding and negligible gA photoinactivation with membranes formed of glycerol monooleate (GMO), whereas Rose Bengal exhibited significant binding and photodynamic efficacy with GMO membranes. Gramicidin photoinactivation in the presence of AlPcS4, being insensitive to the ionic strength of the bathing solution, was inhibited by fluoride and attenuated by phosphate ions. A blue shift of the fluorescence peak position of ZnPcS4 dissolved in ethanol was elicited by phosphate, similarly to fluoride, which was indicative of the coordination interaction of these ions with the central metal atom of the phthalocyanine macrocycle. This interaction was enhanced in the medium modeling the water-membrane interface. The results obtained imply that binding of tetrasulfonated metallophthalocyanines to phospholipid membranes is determined primarily by metal-phosphate coordination.  相似文献

Conditions for the optimal production of polyhydroxyalkanoate (PHA) by Pseudomonas mendocina PSU using a biodiesel liquid waste (BLW) were determined by response surface methodology. These were an initial carbon to nitrogen ratio (C/N) of 40 (mole/mole), an initial pH of 7.0, and a temperature of 35 °C. A biomass and PHA concentration of 3.65 g/L and about 2.6 g/L (77% DCW), respectively, were achieved in a growth associated process using 20 g/L glycerol in the BLW after 36 h of exponential growth. The PHA monomer compositions were 3HB (3-hydroxybutyrate), a short-chain-length-PHA, and the medium-chain-length-PHA e.g. 3-hydroxyoctanoate and 3-hydroxydecanoate. Both the phbC and phaC genes were characterized. The phbC enzyme had not been previously detected in a Pseudomonas mendocina species. A 2.15 g/L of an exopolysaccharide, alginate, was also produced with a similar composition to that of other Pseudomonas species.  相似文献

Summary Preincubation of rat myocardial cells in hypoxic substrate-free Krebs-Ringer bicarbonate buffer (pH 7.4, 37°C) resulted in a substantial decline in high energy phosphates (ATP and CP). Thus, 20 and 60 min preincubation produced a 18 and 72% decline in ATP content, whereas the parallel decline in CP content was 51 and 73%. This energy depletion was accompanied by a change in cell morphology from the initial rod-shaped form to rounded up (hyper-contracted) myocytes. In cells preincubated in substrate-free normoxic buffer, both normal morphology and energy homeostasis were maintained. When energy depleted myocytes later were incubated in the presence of phospholipase C (PLC), this resulted in a substantial release of glycerol, amounting to 92 and 137 nmol/106 cells – 2 h in 20 and 60 min energy depleted myocytes, respectively. In addition, PLC caused an increased leakage of lactate dehydrogenase in energy depleted myocytes. Normal cells, on the other hand, were apparently not affected by PLC. These data suggest that PLC selectively attacks energy depleted and/or structurally damaged myocytes. This could well enhance the breakdown of the natural barrier between the extra- and intracellular compartments and thus augment the cellular damage during ischemia. Moreover, energy depleted myocytes appeared exceptionally sensitive to this enzyme, since the levels required to cause glycerol or lactate dehydrogenase release were several orders of magnitude lower than that required to cause membrane permeation in other cell types.  相似文献
