Hypotrichs are a highly differentiated and very diverse group of ciliated protists. Their systematics and taxonomy are challenging and call for detailed investigations on their general morphology, ultrastructure, ontogenesis, and molecular phylogeny. Here, a comprehensive study is conducted on a brackish water population of Parabistichella variabilis using light and electron microscopy and phylogenetic analyses based on small subunit ribosomal DNA sequence data. Its morphology, including the infraciliature, pellicle, nuclei, buccal seal, and extrusomes, is documented. The present findings indicate that in P. variabilis: (i) the cortical granules are extrusomes, which differ from those of other hypotrichs; (ii) the buccal seal is bounded by the plasma membrane and contains a single layer of longitudinal microtubules; (iii) two contractile vacuoles might be present rather than one; and (iv) the pharyngeal disks are bounded by a single membrane. Early‐to‐middle stages of ontogenesis are described for the first time, enabling the complete characterization of this process. Phylogenetic analyses indicate that Parabistichella variabilis is closely related to several species from different genera, such as Orthoamphisiella breviseries, Uroleptoides magnigranulosus, and Tachysoma pellionellum. However, ultrastructural and gene sequence data for more taxa are needed in order to resolve the systematics of Parabistichella.  相似文献

The buccal complex of the haematophagous pentastomid Reighardia sternae has been described. It consists of a buccal capsule containing a prominent oral papilla communicating with a heavily chitinized pharynx which is crescentic in cross section. The chitinous skeleton of the buccal apparatus provides a framework for the attachment of various muscle systems, the contractions of which have been related to the mode of operation of the oral papilla and pharnyx during the feeding process. Particular emphasis was placed on the possible function of the oral papilla in the initial rupture of a blood capillary, and on the pumping action of the pharynx during the ingestion of the liberated blood.  相似文献

An extra fourth row of pharyngeal teeth was found on both pharyngeal arches in a specmen of the small African barb, Barbus paludinosus , a species normally exhibiting three rows of pharyngeal teeth. Possible phylogenetic significance of this alteration is discussed in connection with occurrence of extra fourth rows of pharyngeal teeth in other cyprinid lineages.  相似文献

The development and homologies of the median elements of the ventral hyoid and branchial arches of Cypriniformes have been unclear. We compared the developmental morphology of this region across five species (Cycleptus elongatus, Luxilus zonatus, Danio rerio, Devario auropurpureus, and Cobitis striata), representing three of five major clades of cypriniforms. The development of basibranchial 1 is similar in catostomids and cyprinids, where a single, elongate, basihyal + anterior copula divides into separate elements. A gap develops between the posterior end of the basihyal cartilage and the anterior copula in catostomids but in cyprinids (Luxiluszonatus, Danio rerio, and Devarioauropurpureus) there is little separation and the basihyal and basibranchial 1 may grow close together or retain a cartilaginous connection (Danio rerio, several outgroups). In loaches and Gyrinocheilus, the gap posterior to the basihyal has been alternately interpreted as either the absence or posterior displacement of basibranchial 1. Uniquely among examined species, in Cobitis striata, the basihyal cartilage and anterior copula form as separate cartilages and remain distinct throughout development with a prominent gap between the basihyal and most anterior basibranchial, which we interpret as loss of basibranchial 1. In the posterior region associated with branchial arches 4 and 5, all examined species except Danio rerio, which has only a basibranchial 4 cartilage, have separate basibranchial 4 and 5 cartilages in early ontogeny. Basibranchials 4 and 5 remain separate in Cycleptus elongatus, Devario auropurpurea, and Cobitis striata, but fuse in Luxilus zonatus to form a posterior copula. The orientation of basibranchial 4 and 5 cartilages in Cobitis striata is similar to catostomids and cyprinids. The most posterior median element in the branchial arches, the post‐ceratobranchial cartilage, generally forms as a separate cartilage in catostomids but in Cobitis striata is connected with basibranchial 5 cartilage from earliest appearance. The mucosa of the mouth, pharynx, oesophagus and rectum of Arrhamphus sclerolepis krefftii contain saccular mucous cells and the lining of the intestinal mucosa contains goblet mucous cells. Saccular mucous cells in the buccal epithelium are present in relatively low densities and contain acidic and neutral glycoprotein-secreting cells in an approximately 1:1 ratio. The saccular mucous cells in the mucosa of the pharynx, oesophagus and rectum are abundant and contain acidic glycoprotein which consists principally of sialomucin with traces of sulphomucin distributed around the periphery of the mucous vacuoles. Goblet cells in the intestinal mucosa contain neutral glycoprotein. Mechanically digested plant material within the lumen of the gut is bound by a sheath of acidic glycoprotein which is in contact with the intestinal mucosa. From these observations and with information on the known properties of acidic glycoproteins, a novel mechanism for the involvement of mucus in the extraction of nutrients from plant material mechanically digested by fish is proposed.  相似文献

Synopsis Pogonias cromis, black drum, is the largest durophagous sciaenid and feeds almost exclusively on hard-shelled bivalves and gastropods using powerful pharyngeal jaws. I estimated pharyngeal jaw bite forces used to crush live molluscs during feeding trials from juvenile and young adult Pogonias cromis, and they are the highest yet documented for bony fishes. Crushing ability in P. cromis scaled with strong positive allometry suggesting large adult fish may have one of the strongest bites among vertebrates. Physiological estimates of pharyngeal muscle strength derived from muscle cross sectional area accounted for only half of the force generated during actual feeding performance trials. The significant disparity between feeding performance and pharyngeal muscle strength in P. cromis indicates the presence of novel biomechanical linkages that enhance crushing ability for feeding on hard-shelled molluscs. I present a biomechanical model in which the lower pharyngeal jaw architecture of P. cromis emulates a second class lever mechanism that can amplify muscle forces transmitted to the shell of the prey.  相似文献

对无胶筛分毛细管电泳体系的筛分介质浓度、缓冲体系的离子强度、分离电压、进样量及温度等条件进行了优化,最终确定最佳分离条件为7.5% Dextran,7.5%丙三醇,0.15mol·L^-1 TB,0.1%SDS,分离电压23.5kV,进样量1.5psi×90秒,温度25℃。应用此方法对金冠×红玉苹果杂种后代韧皮部蛋白质进行测定,得到了多个与阶段转变相关的蛋白质,在韧皮部50节上,有分子量为5.0kDa的特异蛋白质出现,分子量为24.5kDa的蛋白质含量在阶段转变过程中明显升高。  相似文献

Each olfactory sensory neuron (OSN) expresses a single odorant receptor (OR) from a large repertoire of clustered OR genes. It has been hypothesized that OR gene regulation may involve stochastic DNA rearrangement, which in lymphocytes requires the recombination activating genes, rag1 and rag2. We have recently demonstrated that rag1 is expressed in zebrafish OSNs. Here we report that rag2, the obligate partner for rag1 function, is also expressed in OSNs and that its expression pattern mimics that of rag1. The onset of rag1 and rag2 expression preceded that of known zebrafish ORs and the number of rag1-positive OSNs corresponded with the number expressing the olfactory cyclic nucleotide-gated cation channel, an OSN marker. Zebrafish OSNs are the first example of concurrent rag expression in a nonlymphoid tissue. The expression of rag1 and rag2 in OSNs adds to the list of similarities between the olfactory and immune systems that includes monoallelic and mutually exclusive gene expression.  相似文献

The distinct morphological regions of the typical tripartite pharynx found in the nematode taxon Secernentea have distinctive functions. Besides the basic functions of sucking and pumping food against the pressure in the body cavity, the pharynx of Secernentea such as rhabditids serves two additional functions restricted to two pharyngeal subunits. The corpus traps bacteria behind the stoma and at its posterior end. The newly discovered pharyngeal pocket valve helps to trap particles behind the corpus in the rhabditid Poikilolaimus oxycercus and the cephalobid Acrobeles ciliatus (both Secernentea). The grinder of the terminal bulb serves for chewing trapped bacteria. The separated sites of trapping and chewing are connected by the isthmus that transports bacteria towards the grinder. It is likely that this complex feeding structure originated step by step from a two-part pharynx comprising a propharynx and the terminal bulb as in "Plectidae" (that probably include the closest relatives of the Secernentea within the "Adenophorea"). Analysis of video sequences of feeding rhabditids and plectids provided new data to reconstruct this transformation. Within the "Plectidae" two types of grinders occur. The first type or "parietinus type" has triangular chewing plates that can bulge medially and crush food particles. When they retract, new ingested particles are drawn into the grinder. The second type with more solid chewing plates called "butterfly valves" occurs in Ceratoplectus, Plectus parvus, and Wilsonema and can be homologized with the grinder in Secernentea ("Plectidae" is a paraphyletic taxon). Because butterfly valves cannot be retracted, the evolution of such valves required the evolution of an alternative mechanism to fill the grinder with bacteria. The differentiated closing pattern of the dilated pharynx lumen in Ceratoplectus, Plectus parvus, and Wilsonema can be interpreted as the first step in the development of a functional separation of trapping bacteria and of transporting them towards the grinder, which led to the morphologically discernible units of corpus and isthmus found in the Secernentea.  相似文献

应用染色体原位杂交、PCR扩增、克隆及核苷酸序列分析等方法,分析了CNE1和CNE3细胞株中的潜伏感染膜蛋白(LMP1)基因。CNE1是来自我国东北的高分化鼻咽癌细胞株,CNE3是来自广西的低分化鼻咽癌细胞株。染色体原位杂交结果表明,CNE1细胞中LMP1基因存在于细胞核内,整合在第一号染色体上,CNE3中LMP1基因则随机存在于细胞核内及多条染色体上。用PCR方法分别从CNE1及CNE3中扩增得到了LMP1基因片段(外显子3),核苷酸序列分析证明,来自CNE1的LMP1与来自B95-8细胞的LMP1核苷酸序列同源性极高,达99.5%,而CNE3的LMP1基因与B95-8的LMP1基因同源性为93%。  相似文献
