Molecular characterization of salt-stress-associated protein in citrus: protein and cDNA sequence homology to mammalian glutathione peroxidases

A gene encoding for a citrus salt-stress-associated protein (Cit-SAP) was cloned from Citrus sinensis salt-treated cell suspension. The gene, designated csa, was isolated from a cDNA expression library. The partial amino acid sequence of the protein, as well as that deduced from the nucleotide sequence of csa, revealed a considerable homology to mammalian glutathione peroxidase (GP), and to clone 6P229 from tobacco protoplasts. The increased expression of Cit-SAP in NaCl-treated cultured citrus cells and in citrus plants irrigated with saline water, and its similarity to GP, raise the possibility that one of the effects of salt stress in plants may be the increase of the level of free radicals.     

Molecular chaperones and the biogenesis of mitochondria and peroxisomes

Summary— A review of the proteinaceous machinery involved in protein sorting pathways and protein folding and assembly in mitochondria and peroxisomes is presented. After considering the various sorting pathways and targeting signals of mitochondrial and peroxisomal proteins, we make a comparative dissection of the protein factors involved in: i) the stabilization of cytosolic precursor proteins in a translocation competent conformation; ii) the membrane import apparatus of mitochondria and peroxisomes; iii) the processing of mitochondrial precursor proteins, and the eventual processing of certain peroxisomal precursor, in the interior of the organelles; and iv) the requirement of molecular chaperones for appropriate folding and assembly of imported proteins in the matrix of both organelles. Those aspects of mitochondrial biogenesis that have developed rapidly during the last few years, such as the requirement of molecular chaperones, are stressed in order to stimulate further parallel investigations aimed to understand the origin, biochemistry, molecular biology and pathology of peroxisomes. In this regard, a brief review of findings from our group and others is presented in which the role of the F₁-ATPase α-subunit is pointed out as a molecular chaperone of mitochondria and chloroplasts. In addition, data are presented that could question our previous indication that the immunoreactive protein found in the rat liver peroxisomes is due to the presence of the F₁-ATPase α-subunit.     

Protein sequences as random fractals

The analysis of primary sequences from a protein sequence data base suggests that the sequences can be considered as examples of constrained random fractals. Fractal dimensions of the positional distributions of the 20 residues along the chain have been calculated. These fractal dimensions can be used as indices of intrinsic preferences of various residues.

九株根霉可溶性蛋白和三种酶的电泳图谱比较研究

本文报导了根霉属(Rhizopus) 9个菌株天然态及解聚态可溶性蛋白、酯酶同工酶、葡萄糖淀粉酶和SOD电泳图谱的比较研究。结果表明：可溶性蛋白图谱和酯酶同工酶谱能显示五种已知供试菌种间的差异,尤其酯酶同工酶谱还能显示米根霉两个供试菌株之间的微小差异。经综合分析全部试验结果后得出的系统树图显示了9个供试菌株间的亲缘关系,并为未知菌株F1(BR12)和Q303提供了鉴定和命名依据。文中首次报导了根霉的SOD同工酶,并对蛋白质和酶电泳图谱用于根霉分类研究进行了讨论。     

大阪鲫鱼（Carassius Auratus cuvieri Temminck et．Schlegel）雌性特异血清蛋白生化特性及免疫细胞化学定位的研究

雌性特异血清蛋白存在于成熟雌性大阪鲫鱼血清中,雄鱼则无。注射雌二醇可诱导雄鱼及成熟雌鱼合成这种蛋白质并引起鱼肝细胞粗面内质网增加,糖元颗粒减少。从诱导的大阪鲫鱼血清中分离出电泳纯的雌性特异血清蛋白的分子量为４６６０００±４０００（ｎ＝１０）,电泳谱带有３条,用纯的雌性特异血海蛋白制备的兔抗鱼抗血清不与雄鱼血清发生免疫反应,而与正常成熟雌鱼及诱导鱼的血清形成１条免疫沉淀线。免疫细胞化学定位诱导及成熟雌鱼的肝细胞核外细胞质有阳性颗粒,在卵母细胞外围有成团的阳性颗粒分布。     

Excitatory interaction between glutamate receptors and protein kinases

One of the most active areas of neurobiology research concerns mechanisms involved in paradigms of synaptic plasticity. A popular model for cellular leaning and memory is long term potentiation (LTP) in hippocamus. LTP requires postsynaptic influx of Ca²⁺ which triggers multiple biochemical pathways resulting in pre- and postsynaptic mechanisms enhancing long term synaptic efficiency. This article focuses on an acute postsynaptic Mechanism that can enhance responsiveness of glutamate receptors. Evidence is presented that calcium/calmodulin/dependent protein kinase II, the major potsynaptic density protein at excitatory glutaminergic synapses, can phosphorylate glutamate receptors and enhance ion current flowing through them. 1994 John Wiley & Sons, Inc.     

重组人IL6／2融合蛋白对6.5Gy照射小鼠造血功能恢复的影响

观察了ｈＦＰＩＬ６／２对６．５Ｇｙγ线照射ＮＩＨ小鼠第１０天造血功能恢复的影响。结果表明：照射小鼠连续４ｄ给予ｈＦＰＩＬ６／２２５０μｇ·ｋｇ－１·ｄ－１,其脾重、ＣＦＵ－８、骨髓有核细胞数及ＣＭ－ＣＦＵ分别比对照组增加５９．０％、２７８．５％、５７．９％和１３８．２％,统计学处理均有显著差异;对此四项指标的改善也明显优于２５μｇ组。另外,２５０μｇ剂量组小鼠外周血象３０ｄ的动态观察结果表明,ｈＦＰＩＬ６／２不但能明显提高红细胞和血红蛋白的最低值,而且能使血小板的恢复提前。提示ｈＦＰＩＬ６／２在促进血小板生成和促进红系造血方面可能具有良好的应用前景。
