农地整理是人类对农田生态系统的强烈干扰.传统农地整理中路沟渠设计的主要技术经济指标是方便生产通行和调配水资源以及提高水的利用率,尚缺少系统化、生态化考量,已经影响了农田物种扩散,导致生物栖息地环境的退化,并对农田生态系统的生物竞争机制、缓冲和补偿能力、抵御虫害能力产生了显著的负效应.本文系统总结了路沟渠生态化的功能,着重介绍了近年来生态化路沟渠的形式与结构设计、路沟渠生态工法参数的试验以及路沟渠生态材料选择的技术和方法.未来的相关研究应根据各级道路、灌排沟渠的不同功能要求和规格,按照田间不同生物的习性和栖息要求,精确设计能够满足大多数生物需求的农田路沟渠形态、结构和控制性尺寸,切实解决农地整理带来的上述问题. 相似文献

Two protein families involved in membrane traffic, tricalbins and synaptotagmins, contain several copies of C2 domains and are related based on their sequence and domain architecture. Paradoxically, tricalbin and synaptotagmin C2 domains belong to different structural types with apparent circular permutation of terminal beta-strands. To understand whether a topological switch took place, we analyzed tricalbin and synaptotagmin-like C2 domains using two-dimensional structural analysis. We found that yeast tricalbins contain five to six C2 domains. One of these C2 domains possesses many features of synaptotagmin-like C2 domains and also carries a conserved C-terminal strand that is similar to its structural equivalent in synaptotagmin-like C2 domains, suggesting a structural permutation event. Indeed, among higher eukaryotes, animal tricalbins have evolved a C2 domain with synaptotagmin-like topology indicating that the structural conversion has taken place. Investigation of plant synaptotagmins, however, proves that they are direct tricalbin orthologs. Our analysis shows that beta-strand recombination is a possible evolutionary mechanism to generate new structural topologies with altered functional properties. 相似文献

目的:旨在筛选优化花生DNA提取方法。方法:采用SDS法、改进SDS法;CTAB法、改进CTAB法四种方法对花生叶DNA进行提取,并从电泳结果、纯度、得率、等方面对其进行比较研究,从而确定花生DNA提取适用流程;结果:CTAB法DNA平均得率为55.0μg/g.Fw,略小于SDS的60.3,但其A260/A280平均值为1.81,比SDS的1.55更接近标准要求。结论:综合考虑CTAB法是提取花生DNA的最佳方法,可提取到较高质量的DNA,符合分子检测要求;虽然改良CTAB法也相对好于SDS法,但由于步骤增加反而加剧DNA降解。 相似文献

Estimates of daily energy expenditure are important for many areas of research in human ecology and adaptability. The most common technique for estimating human energy expenditure under field conditions, the factorial method, generally relies on activity-specific energy costs derived from published sources, based largely on North American and European subjects. There is concern that such data may not be appropriate for non-Western populations because of differences in metabolic costs. The present study addresses this concern by comparing measured vs. predicted energy costs at rest and during sub-maximal exercise in 83 subjects (52 males, 31 females) from three subsistence-level populations (Siberian herders and high-land and coastal Ecuadorian farmers). Energy costs at rest (i.e., lying, sitting and standing) and while performing a standard stepping exercise did not significantly differ among the three groups. However, resting energy costs were significantly elevated over predicted levels (+16% in men, +11% in women), whereas exercising costs were comparable to predicted values (?6% in men, +3% in women). Elevations in resting energy needs appear to reflect responses to thermal stress. These results indicate that temperature adjustments of resting energy costs are critical for accurately predicting daily energy needs among traditionally living populations. o 1996 Wiley-Liss, Inc. 相似文献

protein with eight additional EGF-like repeats. Our previous study has shown that the EGF precursor without these eight EGF-like

常规的蛋白质碘标方法易引起被标细胞因子的失活,是受体配基竞争结合实验失败的原因之一.试用氯胺-T双相碘标法标记rhG-CSF和rhEPO,并应用受体配基竞争结合分析法测定NFS-60细胞G-CSF受体及BET-2细胞EPO受体的特性.结果显示所获 125I-EPO和 125I-G-CSF放射比活度均较高;发现BET-2细胞有高、低两种亲和力的EPO受体,NFS-60细胞只有一种高亲和力的G-CSF受体,所获结果与文献资料相一致.说明氯胺-T双相碘标法是细胞因子同位素碘标记的理想方法之一. 相似文献

Nucleic acid-based vaccination is a topic of growing interest, especially plasmid DNA (pDNA) encoding immunologically important antigens. After the engineered pDNA is administered to the vaccines, it is transcribed and translated into immunogen proteins that can elicit responses from the immune system. Many ways of delivering DNA vaccines have been investigated; however each delivery route has its own advantages and pitfalls. Skin tattooing is a novel technique that is safe, cost-effective, and convenient. In addition, the punctures inflicted by the needle could also serve as a potent adjuvant. 目的 优化检测烟曲霉刺激A549细胞后磷脂酸(phosphotidic acid,PA)含量变化的方法,间接反应细胞内磷脂酶D(Phospholipase D,PLD)活性变化.方法 建立烟曲霉ATCC13073刺激肺上皮细胞模型;采用甲醇氯仿法提取胞内脂质;用改良的磷脂酸含量测定法测定PA标准品和细胞内PA水平变化规律.结果 PA标准品在5~ 250 μmol/L呈线性关系;经膨胀孢子刺激后,肺上皮细胞内PA含量显著升高,休眠孢子在这一过程中对肺上皮细胞内PA含量无明显作用.结论 改良的PA测量法能快速、稳定而有效地测定细胞内的PLD活性.烟曲霉膨胀孢子能显著激活肺上皮细胞内的PLD活性. 相似文献
