Rice stripe mosaic virus(RSMV) is a rhabdovirus recently found in southern part of China and can cause severe reduction in rice production. To establish serological methods for RSMV epidemiological studies and to establish a control strategy for this virus, we first purified RSMV virions from infected rice plants and then used them as an immunogen to produce four RSMV-specific monoclonal antibodies(MAbs)(i.e.,1D4, 4A8, 8E4 and 11F11). With these MAbs, we have developed a highly specific and sensitive antigen-coated plate enzyme-linked immunosorbent assay(ACP-ELISA), a Dot-ELISA and a Tissue print-ELISA for rapid detections of RSMV infection in rice plants or in leafhoppers. Our results showed that RSMV can be readily detected in RSMV-infected rice plant tissue crude extracts diluted at 1:20,971,520(w/v, g/m L)through ACP-ELISA or diluted at 1:327,680(w/v, g/m L) through Dot-ELISA. Both ACP-ELISA and Dot-ELISA can also be used to detect RSMV infection in individual RSMV viruliferous leafhopper(Recilia dorsalis) homogenate diluted at 1:307,200 and 1:163,840(individual leafhopper/l L), respectively. Detection of RSMV infection in field-collected rice samples or in RSMV viruliferous leafhoppers indicated that the three serological methods can produce same results with that produced by RT-PCR(19 of the 33 rice samples and 5 of the 16 leafhoppers were RSMV-positive). We consider that the four MAbs produced in this study are very specific and sensitive, and the three new serological methods are very useful for detections of RSMV infection in rice plants or in leafhoppers and the establishment of the disease control strategies.  相似文献

Race‐specific resistance of wheat (Triticum aestivum L.) to yellow rust caused by Puccinia striiformis Westend. f.sp. tritici is often short‐lived. Slow‐rusting resistance has been reported to be a more durable type of resistance. A set of sixteen bread wheat varieties along with a susceptible control Morocco was tested during 2004–05 to 2006–07 in field plots at Peshawar (Pakistan) to identify slow rusting genotypes through epidemiological variables including final rust severity (FRS), apparent infection rate (r), area under disease progress curve (AUDPC), average coefficients of infection (ACI) and leaf tip necrosis (LTN). Epidemiological parameters of resistance were significantly (P < 0.01) different for years/varieties in three seasons, while variety × year interactions remained non‐significant. Sequence tagged site (STS) marker, csLV34 analyses revealed that cultivars Faisalabad‐83, Bahawalpur‐95, Suleman‐96, Punjab‐96, Bakhtawar‐93, Faisalabad‐85, Shahkar‐95 and Kohsar‐95 possessed Yr18 linked allele. Faisalabad‐83, Bahawalpur‐95, Suleman‐96, Punjab‐96, Bakhtawar‐93 and Faisalabad‐85 were relatively more stable over 3‐years where FRS, AUDPC and r values reduced by 80, 84 and 70% respectively compared to control Morocco. These six varieties therefore could be exploited for the deployment of Yr18 in breeding for slow rusting in wheat. Both FRS and ACI are suitable parameters for phenotypic selection.  相似文献

Sm proteins are a group of ubiquitous ring-shaped oligomers that function in multiple aspects of RNA metabolism. However, until this study, no comprehensive study incorporating phylogeny, chromosomal location, gene organization, adaptive evolution, expression profiling and functional networks has been reported for rice and maize. In this study, twenty-five and thirty-three Sm genes have been identified in rice and maize, respectively. Phylogenetic analyses identified eighteen gene groups. Results by gene locations indicated that segmental duplication contributes to the expansion of this gene family in rice and maize. Gene organization and motif compositions of the Sm members are highly conserved in each group, indicative of their functional conservation. Expression profiles have provided insights into the possible functional divergence among members of the Sm gene family. Adaptive evolution analyses suggested that purifying selection was the main force driving Sm evolution, but some critical sites might be responsible for functional divergence. In addition, four hundred and seventy-nine interactions were identified by functional network analyses, and most of which were associated with binding, cellular macromolecule biosynthesis, pre-mRNA processing and transferase activity. Overall, the data contribute to a better understanding of the complexity of Sm gene family in rice and maize and will provide a solid foundation for future functional studies.  相似文献

以硬粒小麦-粗山羊草人工合成小麦CI184、感病品种‘铭贤169’及其杂交组合的正反交F1以及CI184/‘铭贤169’F2、F2:3家系为材料,鉴定其条锈病抗性,对CI184条锈病抗性进行遗传分析;采用SSR分子标记技术和集群分离分析法进行多态性筛选,以F3抗病鉴定数据为依据,对CI184中条锈病抗性基因进行分子标记定位。结果显示:(1)CI184在苗期抗性鉴定中,对30种小麦条锈菌生理小种表现抗性,但对中国四川新出现的条锈菌生理小种V26表现苗期感病;在田间成株抗性接种鉴定中,CI184对中国流行的小麦条锈菌生理小种条中32、条中33、水源4、水源5、水源7和V26等表现出成株抗性。(2)CI184中条锈病抗性由隐性基因位点控制。(3)仅检测到一个控制条锈病抗性的QTL位点,位于1B染色体上Xgwm18和Xwmc626之间,暂时命名为Qyr.zz_1B,在四川和北京2个环境中可分别解释CI184中13.36%和18.07%的成株抗性贡献率。(4)Qyr.zz_1B位点的3个SSR标记和Yr15的1个SSR标记可以区分该位点与1B染色体上的其他抗条锈病基因,如Yr15、Yr24和Yr26/YrCH42。表明Qyr.zz_1B位点在小麦条锈病的抗病育种中具有潜在的应用价值。  相似文献

Heat shock proteins 70 (HSP70s) are a highly conserved family of genes in eukaryotes, and are involved in a remarkable variety of cellular processes. In many plant positive‐stranded RNA viruses, HSP70 participates in the construction of a viral replication complex and plays various roles during viral infection. Here, we found increased expression of HSP70 following infection by Rice stripe virus (RSV), a negative‐stranded RNA virus, in both rice (the natural host) and Nicotiana benthamiana (an experimental host). Heat treatment of N. benthamiana (Nb) plants enhanced viral infection, whereas RSV infection was retarded and viral RNAs accumulated at a low level when HSP70 was silenced. In both bimolecular fluorescence complement and in vitro pull‐down assays, the N‐terminus of RSV RNA‐dependent RNA polymerase (RdRp) interacted and co‐localized with the HSP70s of both plants (OsHSP70 and NbHSP70). The localization of the N‐terminus of RdRp when expressed alone was not obviously different from when it was co‐expressed with OsHSP or NbHSP, and vice versa. RSV infection also had no effect on the localization of host HSP70. These results demonstrate that host HSP70 is necessary for RSV infection and probably plays a role in viral replication by interacting with viral RdRp, which provides the first evidence of an interacting host protein related to RSV replication, which has been little studied to date.  相似文献

小麦条锈病是世界范围内小麦上最重要的流行性病害之一,可造成严重的产量损失。陇南地区是我国小麦条锈菌主要越夏易变区和新小种发源地,了解该地区不同海拔高度区域内条锈菌遗传多样性有重要意义。本研究采用TP-M13-SSR荧光标记技术对11个种群330个小麦条锈菌分离株基因组DNA进行了SSR标记分析。不同海拔区域的条锈菌遗传多样性有明显的差异,高山区的遗传多样性比较丰富,半山区次之,川道区相对比较低。不同生态区域内,小麦条锈菌群体遗传分化程度不同,高山区和半山区遗传分化程度大,基因流小,川道区群体遗传分化程度比较小,基因流大。来自不同海拔区域的菌系具有相同的基因型,这一结果从分子水平证明了在陇南地区小麦条锈菌在山区与川地之间存在广泛的菌源交流,可就地完成周年循环。  相似文献

Yellow rust populations of Pakistan were characterised for their virulence pathotypes/races and pathogenetic variation using seedling evaluation of differential genotypes under glasshouse conditions in Murree (6000 feet above sea level). Differential genotypes comprised a world set, an European set, near isogenic lines and the universally susceptible bread wheat cultivar “Morocco”. Over the two-year study a total of 18 race groups were identified. Out of these 18 race groups, several (68E0, 64E0, 66E0, 70E0, 6E0, 71E0, 6E0, 2E0, 67E0, and 68E16) were found previously. The new race group 70E32 found probably evolved because of mutation from the previously existing 70E16. Virulence frequencies of yellow rust (Yr) resistance genes were also determined on near isogenic lines. The highest virulence frequencies (%) were found for Yr7 (88%), Yr9 (57%), Yr18 (70%), and Yr24 (69%). Virulence frequencies were low for Yr 1 (4%), Yr5 (7%), Yr10 (10%) and Yr15 (4%). Our studies indicated that virulence existed for almost all yr genes, necessitating regular monitoring of the yellow rust populations and intensifying efforts to identify new sources of resistance to this pathogen.  相似文献

Stripe rust of barley (Hordeum vulgare L.), caused by Puccinia striiformis f. sp. hordei, is a serious problem of barley production in many parts of the world. Monitoring of the pathogen virulence factors and their changes provides basic information for development of an early warning system to breeders and researchers. To monitor the regular virulence changes, trap nurseries comprising of 12 barley differential sets were planted at different parts of Iran in six consecutive years 2007–2012. When the infection and severity under natural infection on susceptible cultivar Afzal as the check was high, then the response of each line was assessed using modified Cobbs scale. Results revealed that no virulence was observed on plants with resistance genes rpsEm1, rpsEm2, rpsHF, Rps4, rpsVa1, rpsVa2 and rpsAst. Therefore, these genes were considered effective genes and can be used to pyramid with those for race-non-specific resistance genes to achieve more durable and highly effective resistance to stripe rust. The plants with the resistance genes rps2, Rps1.b, Rps3 and rpsI5 showed susceptible reaction and virulence was observed on them, thus their resistance genes were considered ineffective.  相似文献

利用抗条锈病基因Yr10和Yr15的SCAR和Barc8标记以及1BL/1RS易位的复合标记,对青海省育成和引进的137份小麦品种进行检测,以明确Yr10和Yr15基因以及1BL/1RS易位在青海小麦品种资源中的分布.结果显示:在137份材料中,有4份检测到Yr10基因,19份检测到Yr15基因,分别占参试材料的2.9%和13.9%,没有检测到同时携带Yr10和Yr15基因的材料;有22份材料为1BL/1RS易位,占参试材料的16.1%.研究表明,青海省大部分小麦抗锈品种及1BL/1RS易位品种为外引种品种.  相似文献
