We studied amidated and non-amidated piscidins 1 and 3, amphipathic cationic antimicrobial peptides from fish, to characterize functional and structural similarities and differences between these peptides and better understand the structural motifs involved in biological activity and functional diversity among amidated and non-amidated isoforms. Antimicrobial and hemolytic assays were carried out to assess their potency and toxicity, respectively. Site-specific high-resolution solid-state NMR orientational restraints were obtained from 15N-labeled amidated and non-amidated piscidins 1 and 3 in the presence of hydrated oriented lipid bilayers. Solid-state NMR and circular dichroism results indicate that the peptides are α-helical and oriented parallel to the membrane surface. This orientation was expected since peptide-lipid interactions are enhanced at the water-bilayer interface for amphipathic cationic antimicrobial peptides. 15N solid-state NMR performed on oriented samples demonstrate that piscidin experiences fast, large amplitude backbone motions around an axis parallel to the bilayer normal. Under the conditions tested here, piscidin 1 was confirmed to be more antimicrobially potent than piscidin 3 and antimicrobial activity was not affected by amidation. In light of functional and structural similarities between piscidins 1 and 3, we propose that their topology and fast dynamics are related to their mechanism of action. 相似文献

The antioxidant capacity of human plasma was determined by following the oxidation kinetics of the lipid-soluble fluorescent marker BODIPY using 2,2'-azobis(4-methoxy-2,4-dimethylvaleronitrile) (MeO-AMVN) as the lipophilic radical initiator. The results are expressed as a total antioxidant performance (TAP) value based on the inhibition of BODIPY oxidation, as determined by the appearance of green fluorescence, with respect to a control sample (phosphatidylcholine with or without delipidized human serum). The suitability of the assay was evaluated on the basis of its precision, reproducibility, and specificity. The intra- and interassay coefficients of variation both were less than 5%. The addition of a representative substrate of plasma peroxidation, phosphatidylcholine, up to 750mug/ml did not induce significant changes in the TAP value. Also, BODIPY photooxidation was not observed during the experimental time course (220min). The TAP values of 6 plasma samples from healthy donors were measured and correlated with the main plasma water- and lipid-soluble antioxidants (uric acid and ascorbic acid, alpha-tocopherol, and carotenoids) and lipid profiles. Significant correlations were found between TAP and uric acid (R=0.97, P<0.05) and cholesterol-adjusted alpha-tocopherol (R=0.93, P<0.01). The results confirm that the TAP assay is suitable to measure the antioxidant activity of plasma antioxidants localized in both the lipophilic and hydrophilic compartments. 相似文献

Silica in grasses as a defence against insect herbivores: contrasting effects on folivores and a phloem feeder 总被引:4,自引:0,他引:4
1. Silica, deposited as opaline phytoliths in the leaves of grasses, constitutes 2-5% of dry leaf mass, yet its function remains unclear. It has been proposed that silica may act as an antiherbivore defence by increasing the abrasiveness and reducing the digestibility of grass leaves, although there is little direct experimental evidence to support this. 2. We investigated the effects of manipulated silica levels on the abrasiveness of the leaves of five grass species. We also examined the effects of silica levels on the feeding preferences, growth performance and digestion efficiency of two folivorous insects and one phloem-feeding insect. 3. Silica addition resulted in increases to leaf abrasiveness in four of the five grass species studied. Silica addition also deterred feeding by both folivores and reduced their growth rates and digestion efficiency. 4. These effects resulted in lower pupal mass of the lepidopteron larvae Spodoptera exempta and compensatory feeding by the orthopteran, Schistocerca gregaria. In contrast, silica had no effects on the feeding preference or the population growth of the phloem feeder, Sitobion avenae. 5. Our results demonstrate that silica is an effective defence against folivorous insects, both as a feeding deterrent, possibly mediated by increased abrasiveness, and as a digestibility reducer. The effects of silica on pupal mass and development time may impact on herbivore fitness and exposure to natural enemies. 6. These results are the first demonstration of a direct effect of silica on the abrasiveness of grasses and the adverse impact of silica on herbivore preference and performance. 相似文献

It is well documented across phyla that animals experiencing stress and fear produce chemical warning signals that can lead to behavioral, endocrinological, and immunological changes in the recipient animals of the same species. Humans distinguish between fear and other emotional chemosignals based on olfactory cues. Here, we study the effect of human fear chemosignals on the speed and accuracy of cognitive performance. In a double-blind experiment, female participants performed a word-association task while smelling one of the three types of olfactory stimuli: fear sweat, neutral sweat, and control odor carrier. We found that the participants exposed to the fear condition performed more accurately and yet with no sacrifice for speed on meaningful word conditions than those under either the neutral or the control condition. At the same time, they performed slower on tasks that contained ambiguous content. Possible factors that could introduce bias, such as individual differences due to anxiety, verbal skills, and perceived qualities of the smells, were ruled out. Our results demonstrate that human fear chemosignals enhance cognitive performances in the recipient. We suggest that this effect originates from learned associations, including greater cautiousness and concomitant changes in cognitive strategies. 相似文献

Daily changes in cognitive performance have been documented, both in time of day/effect paradigm studies and in time-isolation studies. However, in both types of studies, phenomena such as the “post-lunch dip” have been found that were difficult to explain in terms of theoretical backgrounds. These phenomena may suggest ultradian rhythms in cognitive performance. A number of studies have also shown ultradian and asymmetric rhythms in activity indices of the brain hemispheres. The aim of this study was to test three hypotheses: the first two assumed that there is a significant ultradian frequency in a component of the endogenous rhythm of processing speed, and the third assumed that the ultradian endogenous rhythms of the processing speed (encoding and recognition) of stimuli addressed to the left brain hemisphere differ in period length from those addressed to right hemisphere. During a 24 h constant-routine experiment, the memory performance of 30 participants was measured eight times (every 2.5-3 h), starting at 06:30 h. Parallel sets of words and pictures were shown to subjects in a random order in either the left or the right visual field on a computer screen. The participants pressed one of two buttons in response to the picture or word, or when answering a question concerning the meaning of a presented stimulus. Cosinor analysis was applied to individual time series data. Two significant ultradian components were found in a majority of the time series. Dominant periods were analyzed using three factor ANOVA. The results showed an asymmetry between both hemispheres in the frequency of ultradian rhythms in encoding speed. 相似文献

Inter-individual differences in tolerance for shift work have been studied primarily in terms of external factors affecting alertness on the job or the ability to rest and sleep while at home. However, there is increasing evidence that neurobiological factors play a role as well, particularly the major processes involved in the regulation of sleep and wakefulness. These include a sleep homeostatic process seeking to balance wakefulness and sleep and a circadian process seeking to promote wakefulness during the day and sleep during the night. Shift work is associated with a temporal misalignment of these two endogenous processes. During nightwork, this misalignment makes it difficult to stay awake during the nightshift and sleep during the day. However, inter-individual variability in the processes involved in sleep/wake regulation is substantial. Recent studies have demonstrated the existence of inter-individual differences in vulnerability to cognitive deficits from sleep loss. Moreover, these inter-individual differences were shown to constitute a trait. Interestingly, self-evaluations of sleepiness did not correspond well with the trait inter-individual variability in objective levels of performance impairment during sleep deprivation. Perhaps because of this discrepancy, in operational settings, the inter-individual differences in vulnerability to sleep loss do not appear to be limited due to self-selection mechanisms. Indeed, even among a highly select group of active-duty jet fighter pilots flying a series of simulated night missions, systematic inter-individual differences in performance impairment from sleep loss were still observed. There are significant personal and economic consequences to human error and accidents caused by performance deficits due to sleep loss. It is important, therefore, to study the inter-individual differences in the regulation of sleep and wakefulness in the work environment so that cognitive impairment during shift work may be better anticipated and prevented. 相似文献

Pupils shift their time of day preferences from morningness to eveningness during the age of puberty. Therefore, early school start times may have a negative influence on school functioning, adolescent health and on grade point average. Here, we show that morningness - eveningness influences school performance as measured by the final school leaving/university entrance certificate. One hundred and thirty-two university students aged between 20 and 22 years participated in our study. We found a highly significant positive correlation between the average grading of the final exam and the morningness score, suggesting that pupils with morning preferences performed better in school achievement. Evening types seem at a serious disadvantage because the final exams are used for university entrance decisions. 相似文献

The chloroplast is a type of plant specific subcellular organelle. It is of central importance in several biological processes like photosynthesis and amino acid biosynthesis. Thus, understanding the function of chloroplast proteins is of significant value. Since the function of chloroplast proteins correlates with their subchloroplast locations, the knowledge of their subchloroplast locations can be very helpful in understanding their role in the biological processes. In the current paper, by introducing the evidence-theoretic K-nearest neighbor (ET-KNN) algorithm, we developed a method for predicting the protein subchloroplast locations. This is the first algorithm for predicting the protein subchloroplast locations. We have implemented our algorithm as an online service, SubChlo (http://bioinfo.au.tsinghua.edu.cn/subchlo). Two experiments were conducted to determine if nutritional supplementation improved ovulation and pregnancy rates in female goats managed under grazing conditions and submitted to the male effect. In Experiment 1, one group of does did not receive nutritional supplementation, while the other group was supplemented daily for 7 days starting at the time when the males were introduced to the females. The ovulation rate at the second male-induced ovulation was greater (P < 0.05) in supplemented (2.0 ± 0.1) than in non-supplemented (1.6 ± 0.1) does. For Experiment 2, female goats were supplemented for 0, 7, 14 or 28 days, starting 9 days following buck introduction. The proportion of does that were pregnant in the group supplemented for 28 days was greater (P < 0.05) than in the non-supplemented group, but did not differ from 14-day and the 7-day supplemented groups. The proportion of pregnant does was greater (P < 0.05) in the group supplemented for 14 days compared to the group supplemented for 7 days and the non-supplemented group. These latter two groups did not differ (P > 0.05). In conclusion, feed supplementation for 7 days, starting at the time when males were introduced increased ovulation rate and feed supplementation for 14 or 28 days starting 9 days after males were introduced improved pregnancy rates in goats managed under grazing conditions and exposed to males. 相似文献
