为了了解孔雀草种质间的亲缘关系,提高孔雀草种质的利用效率,从18个形态特征对40份孔雀草种质进行了遗传变异分析、主成分分析及聚类分析。结果表明,孔雀草表型多样性丰富,种质间表型性状变异程度高,变异系数(CV值)范围3.92%~46.25%,以花朵数最大,其次是冠幅和株高;Shannon-Weaver多样性指数则以叶片性状及花性状较高,平均2.0以上。国内种质比国外种质多样性更丰富。通过主成分分析,筛选出对总体方差累计贡献率达78.949%的4个主成分,并筛选出综合性状表现良好的种质10份。参试的40份种质在欧氏距离阈值为7.29处可分为两大类,一类植株生长势较强包含2份种质(水星黄色和橙色);其余38份种质为一类,在欧氏距离阈值为6.0处,又以花色差异可进一步划分为2个类群,说明生长势及花色可作为孔雀草种质依据表型性状分类的重要指标。 相似文献

Habitat fragmentation has been shown to disrupt ecosystem processes such as plant-pollinator mutualisms. Consequently, mating patterns in remnant tree populations are expected to shift towards increased inbreeding and reduced pollen diversity, with fitness consequences for future generations. However, mating patterns and phenotypic assessments of open-pollinated progeny have rarely been combined in a single study. Here, we collected seeds from 37 Eucalyptus incrassata trees from contrasting stand densities following recent clearance in a single South Australian population (intact woodland=12.6 trees ha−1; isolated pasture=1.7 trees ha−1; population area=10 km2). 649 progeny from these trees were genotyped at eight microsatellite loci. We estimated genetic diversity, spatial genetic structure, indirect contemporary pollen flow and mating patterns for adults older than the clearance events and open-pollinated progeny sired post-clearance. A proxy of early stage progeny viability was assessed in a common garden experiment. Density had no impact on mating patterns, adult and progeny genetic diversity or progeny growth, but was associated with increased mean pollen dispersal. Weak spatial genetic structure among adults suggests high historical gene flow. We observed preliminary evidence for inbreeding depression related to stress caused by fungal infection, but which was not associated with density. Higher observed heterozygosities in adults compared with progeny may relate to weak selection on progeny and lifetime-accumulated mortality of inbred adults. E. incrassata appears to be resistant to the negative mating pattern and fitness changes expected within fragmented landscapes. In mixed-mating plant populations, one can estimate the relative fitness of selfed progeny w by measuring the inbreeding coefficient F and selfing rate s of adults of one generation, together with F of adults in the following generation (after selection). In the first application of this multigenerational method, we estimated F and s for adults over three consecutive generations in adjacent populations of two annual Mimulus taxa: the outbreeding M. guttatus and the inbreeding M. platycalyx. This gave estimates of w for the last two generations. Although average multilocus selfing rates were high in both taxa (0.63 in M. guttatus; 0.84 in M. platycalyx), the relative fitness of selfed progeny averaged only 0.19 in M. guttatus and 0.32 in M. platycalyx. An alternative estimator for w that incorporates biparental inbreeding gave even lower estimates of w. These values are significantly below the 0.5 threshold thought to favor selfing, and show that partially selfing populations can harbor substantial genetic load. In accordance with the purging hypothesis, the more highly selfing M. platycalyx showed marginally lower inbreeding depression than M. guttatus in both years (P = 0.08). Inbreeding depression and selfing rates also varied among years in concert among taxa. Several sources of bias are discussed, but computer simulations indicate it is unlikely that w is biased downwards by linkage of marker loci to load loci. 相似文献

为探究云杉矮槲寄生(Arceuthobium sichuanense)的遗传多样性及其与不同寄主选择压力和地理分布的关系,采用ISSR分子标记方法,对青海、甘肃、四川等地区5种寄主上的100份云杉矮槲寄生样本进行遗传多样性和遗传结构分析。结果表明:(1)10条引物共扩增出130个条带,其中多态性条带129条,多态位点百分率(PPB)为99.23%。(2)物种水平上的Nei’s遗传多样性指数(He)和Shannon信息指数(I)分别为0.313 9和0.476 5,表明云杉矮槲寄生物种水平的遗传多样性较高,但群体间的基因流(Nm=0.528 7)较弱,可能会加速群体间的遗传分化(Gst=0.486)。(3)UPGMA聚类结果显示,来自甘肃、青海的样本聚为一组,表现出较高的相似性,而四川的样本独立聚类;不同寄主来源的聚类结果显示,寄生于鳞皮云杉(Picea asperata)与川西云杉(P.likiangensis var.balfouriana)的样本聚为一类,而寄生于青海云杉(P.crassifolia)、青杄(P.wilsonii)和紫果云杉(P.purpurea)的样本聚为一类,表明地理隔离和寄主选择压力对云杉矮槲寄生的遗传分化起到了重要作用。 相似文献

A method for screening copper (Cu)-tolerant rice cultivars was studied by combining hydroponic experiments and cluster analysis, and the classification of cultivars in Cu stress tolerance was done. In the first hydroponic experiment, seedlings of Jiahe991 and Xiushui114 were planted in nutrient solution with different Cu2+ concentrations from 10 to 1800 μg/L. Results indicated that the toxic threshold of Cu concentration in solution ranged from 900 to 1200 μg/L, since SPAD (Soil and Plant Analyzer Development, SPAD-502, a portable chlorophyll meter, Minolta Camera Co. Ltd., Japan) values of leaves and seedlings biomass of the treatments with ≥900 and/or 1200 µg/L were significantly lower than the control. The second experiment was conducted with 16 local rice cultivars under three Cu treatments (10, 1000, and 1500 μg/L). The 16 cultivars were well classified into tolerant, normal, and sensitive groups as a result of cluster analysis based on the relative SPAD (Soil and Plant Analyzer Development, SPAD-502, a portable chlorophyll meter, Minolta Camera Co. Ltd., Japan) value, shoot and root dry weights, root length and root dehydrogenase activity, and oxidizing capacity and shoot Cu concentration. Xiushui123, Xiushui134, Jiahe991, and Xianghu301 belonged to the tolerant group; Xiushui137 belonged to the sensitive group. The cluster analysis based on hydroponic experiments is an effective method for identifying rice cultivars that are tolerant to Cu stress. In order to assess heavy metal pollution of sediment samples collected from Shuangtaizi estuary, contamination factor (CF) and multivariate statistical analyses, including principal component analysis (PCA), cluster analysis (CA) and correlation analysis, are carried out in this paper. CF confirms that Pb, Cu and Hg concentrations are very low and all fall within the range of background, while Zn and Cd demonstrate moderate contamination (in A10, A13, A15, A16 and A17 sites) and very high contamination (in A10, A13, A15 and A17 sites), respectively. The PCA indicates that four significant principal components (PCs) are extracted, explaining 88.959% of total variance, which suggests that Pb, Cu and Zn are mainly associated with organic carbon (OC). The result from CA is consistent with that obtained from PCA, classifying that the heavy metals in two clusters derive from different sources. Correlation analysis supports the conclusions from PCA and CA, elucidating the relationships between heavy metals and particle sizes of the sediments. 相似文献

Genetic variation in CACNA1C, which codes for the L‐type calcium channel (LTCC) Cav1.2, is associated with clinical diagnoses of bipolar disorder, depression and schizophrenia. Dysregulation of the mesolimbic‐dopamine (ML‐DA) system is linked to these syndromes and LTCCs are required for normal DAergic neurotransmission between the ventral tegmental area (VTA) and nucleus accumbens (NAc). It is unclear, however, how variations in CACNA1C genotype, and potential subsequent changes in expression levels in these regions, modify risk. Using constitutive and conditional knockout mice, and treatment with the LTCC antagonist nimodipine, we examined the role of Cacna1c in DA‐mediated behaviors elicited by psychomotor stimulants. Using fast‐scan cyclic voltammetry, DA release and reuptake in the NAc were measured. We find that subsecond DA release in Cacna1c haploinsufficient mice lacks normal sensitivity to inhibition of the DA transporter (DAT). Constitutive haploinsufficiency of Cacna1c led to attenuation of hyperlocomotion following acute administration of stimulants specific to DAT, and locomotor sensitization of these mice to the DAT antagonist GBR12909 did not reach the same level as wild‐type mice. The maintenance of sensitization to GBR12909 was attenuated by administration of nimodipine. Sensitization to GBR12909 was attenuated in mice with reduced Cacna1c selectively in the VTA but not in the NAc. Our findings show that Cacna1c is crucial for normal behavioral responses to DA stimulants and that its activity in the VTA is required for behavioral sensitization. Cacna1c likely exerts these effects through modifications to presynaptic ML‐DA system function. 相似文献
