我们应用微量(1—3微克蛋白)等电点聚焦和SDS双向电泳对鸡胚视网膜发育的第4天(E_4)到第16天(E_(16))总蛋白图谱进行分析,并结合印迹的PNA免疫酶标反应,分析了总蛋白中与PNA结合的糖蛋白图谱的变化。E_4时没有看到PNA结合糖蛋白的任何阳性反应的斑点,E_6时这种糖蛋白的种数突然增加,约占总蛋白数的20%。新的糖蛋白增加的速率在E_6时最快,存其后的发育阶段(E_8—E_(16))慢于总蛋白增加的速率。这种糖蛋白大都是等电点在中性pH范围内,中等大小分子量的糖肽。视神经的总蛋白中含PNA结合糖蛋白数的比例高于同一发育年龄视网膜中的比例,这说明糖蛋白可能主要存在于质膜上。视网膜的总蛋白图谱相似于前脑的,但与视顶盖的更相近。本文对这种能与PNA结合,即含有D-半乳糖残基的糖蛋白在视网膜发育中的意义进行了讨论。 相似文献

Phenotypic plasticity of two primitive wheat species (Triticum monococcum L. and Triticum dicoccum S.) was studied in response to early chilling stress. Selection pressure differentials, gradients and plasticity costs on plant morphogenesis, growth and reserve carbohydrate consumption were estimated. Regression analysis was applied to investigate differential developmental changes and patterns between treatments. Four‐day‐old seedlings of T. monococcum and T. dicoccum, differing in plant stature and reserve carbohydrates, were given an early chilling temperature (4 °C for 42 day) and compared with control plants grown at 23 °C. Early chilling stress resulted in a significant increase in leaf mass ratio (LMR) and relative growth rate (RGR), a reduction in flag leaf size, total biomass, specific leaf area (SLA) and reserve carbohydrate storage at flowering, together with advanced onset of flowering. Selection pressure within the early chilling environment favoured early flowering, smaller SLA, higher LMR and lower reserve carbohydrates, suggesting the observed responses were adaptive. Furthermore, a regression of daily cumulative plant biomass derived from a crop growth simulation model (CERES‐Wheat) on crop vegetation period revealed a divergent developmental pattern in early‐chilled plants. Using selection pressure gradient analysis, we found similar responses among these traits, except for SLA and sucrose, indicating that these two traits have indirect effects on fitness. Thus, the total effects of SLA and reserve sucrose on relative fitness seem to be buffered via the rapid growth rate in chilled plants. While lower SLA may reduce early chilling stress effects at an individual leaf level, a higher LMR and use of reserve carbohydrates indicated that compensatory growth of chilled plants during the recovery period relied on the concerted action of altered resource allocation and reserve carbohydrate consumption. However, a significant cost of plasticity was evident only for flowering time, LMR and fructan levels in the early chilling environment. Some populations of coastal bottlenose dolphins ( Tursiops truncatus ) comprise discrete communities, defined by patterns of social association and long-term site fidelity. We tested the hypothesis that bottlenose dolphins in Tampa Bay, Florida, form a single community. The longitudinal study of dolphins in Sarasota Bay, adjacent to Tampa Bay, allowed us to ground-truth the definition of community and test whether our approach was robust to small sample sizes of resightings. We conducted photo-identification surveys in Tampa Bay during 1988–1993, and identified 102 dolphins with 10 or more sightings. We used hierarchical cluster analysis to examine the locations and association indices of these dolphins. We used analysis of variance (ANOVA) to test for differences in mean locations and determine whether mean coefficient of association (CoA) values within a community were higher than among communities. Dolphins in Tampa Bay clustered into five putative communities differing significantly in location and CoA values. Kernel estimates of the ranges of these five communities exhibited little overlap; some communities had no overlap at all. We conclude that five discrete communities of bottlenose dolphins exist in Tampa Bay and that such fine-scale structure may be a common feature of bottlenose dolphin populations throughout the southeastern United States. 相似文献

The greatest threat to the protected Eurasian lynx (Lynx lynx) in Central Europe is human‐induced mortality. As the availability of lynx prey often peaks in human‐modified areas, lynx have to balance successful prey hunting with the risk of encounters with humans. We hypothesized that lynx minimize this risk by adjusting habitat choices to the phases of the day and over seasons. We predicted that (1) due to avoidance of human‐dominated areas during daytime, lynx range use is higher at nighttime, that (2) prey availability drives lynx habitat selection at night, whereas high cover, terrain inaccessibility, and distance to human infrastructure drive habitat selection during the day, and that (3) habitat selection also differs between seasons, with altitude being a dominant factor in winter. To test these hypotheses, we analyzed telemetry data (GPS, VHF) of 10 lynx in the Bohemian Forest Ecosystem (Germany, Czech Republic) between 2005 and 2013 using generalized additive mixed models and considering various predictor variables. Night ranges exceeded day ranges by more than 10%. At night, lynx selected open habitats, such as meadows, which are associated with high ungulate abundance. By contrast, during the day, lynx selected habitats offering dense understorey cover and rugged terrain away from human infrastructure. In summer, land‐cover type greatly shaped lynx habitats, whereas in winter, lynx selected lower altitudes. We concluded that open habitats need to be considered for more realistic habitat models and contribute to future management and conservation (habitat suitability, carrying capacity) of Eurasian lynx in Central Europe. 相似文献

The number of sires fertilizing a given dam is a key parameter of the mating system in species with spatially restricted offspring dispersal, since genetic relatedness among maternal sibs determines the intensity of sib competition. In flowering plants, the extent of multiple paternity is determined by factors such as floral biology, properties of the pollen vector, selfing rate, spatial organization of the population, and genetic compatibility between neighbours. To assess the extent of multiple paternity and identify ecological factors involved, we performed a detailed study of mating patterns in a small population of a self-incompatible clonal herb, Arabidopsis halleri . We mapped and genotyped 364 individuals and 256 of their offspring at 12 microsatellite loci and jointly analysed the level of multiple paternity, pollen and seed dispersal, and spatial genetic structure. We found very low levels of correlated paternity among sibs ( P full-sib = 3.8%) indicating high multiple paternity. Our estimate of the outcrossing rate was 98.7%, suggesting functional self-incompatibility. The pollen dispersal distribution was significantly restricted (mean effective pollen dispersal distance: 4.42 m) but long-distance successful pollination occurred and immigrating pollen was at most 10% of all pollination events. Patterns of genetic structure indicated little extent of clonal reproduction, and a low but significant spatial genetic structure typical for a self-incompatible species. Overall, in spite of restricted pollen dispersal, the multiple paternity in this self-incompatible species was very high, a result that we interpret as a consequence of high plant density and high pollinator service in this population. 相似文献

Alleviation of Changes in Protein Metabolism in NaCl-Stressed Wheat Seedlings by Thiamine 总被引:7,自引:0,他引:7
NaCl-stress induced a pronounced suppression in growth of wheat seedlings. The most abundant amino acids (cysteine, arginine, methionine) constituting about 55 % of total free amino acid content in control wheat were reduced in 100 mM NaCl-treated plants. However, valine, isoleucine, aspartic acid and proline accumulated in response to NaCl stress and NaCl-treated wheat seedlings showed 1.6 fold increase in total free amino acids compared to the control. Addition of 2 [micro ]M thiamine alleviated the effects of NaCl on the amino acid composition and the amount of total free amino acids decreased to that in the control. Content of 26 kDa protein increased in NaCl-treated plants, stimulation was more pronounced in roots than in shoots. In contrast, the contents of 13 and 20 kDa proteins decreased. After addition of thiamine, the 24 kDa protein, which disappeared with NaCl treatment, has been initiated again. Moreover, thiamine treatment stimulated the accumulation of the 20 kDa protein. 相似文献
