This work aimed to develop a novel antimicrobial coating based on chitosan and PVA to evaluate its effect on minimally processed tomato by means of microbiological analyses. In this report an antimicrobial film was prepared by blending chitosan (CS) and poly(vinyl alcohol) (PVA) with glutaraldehyde as the cross-linker. The miscibility and morphology of the film were investigated by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The thermal property of the chitosan–PVA film was examined by differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA). The microbiological screening has demonstrated the antimicrobial activity of the film against food pathogenic bacteria viz. Escherichia coli, Staphylococcus aureus, and Bacillus subtilis. The obtained results indicate the film may be a promising material for food packaging applications.  相似文献

Recurrent sea urchin mass mortality has recently affected eastern Atlantic populations of the barren‐forming sea urchin Diadema africanum. This new episode of die‐off affords the opportunity to determine common meteorological and oceanographic conditions that may promote disease outbreaks. The population dynamics of this sea urchin species are well known—urchin barrens have persisted for many decades along most of the coastlines off the archipelagos of Madeira, Selvages, and the Canary Islands, where they limit macroalgae biomass growth. However, this new and explosive mortality event decimated the sea urchin population by 93% on Tenerife and La Palma Islands. Two severe episodes of southwestern rough sea that led to winter storms, in February 2010 (Xynthia) and February 2018 (Emma), preceded both mass mortality events. The autumn and winter months of those years were anomalous and characterized by swells with an average wave height above 2 m that hit the south and southwest sides of the islands. The amoeba Paramoeba brachiphila was the only pathogen isolated this time from the moribund and dead sea urchins, suggesting that the amoeba was the primary cause of the mortality. This new sea urchin die‐off event supports the “killer‐storm” hypothesis that has been already described for western Atlantic coasts. These anomalous southwest storms during winters generate pronounced underwater sediment movement and large‐scale vertical mixing, detected in local tide gauge, which may promote paramoebiasis. This study presents valuable insights about climate‐mediated changes in disease frequency and its impacts on the future of coastal marine ecosystems in the Atlantic.  相似文献

Field ornithologists have used traditional culture‐based techniques to determine the presence and abundance of microbes on surfaces such as eggshells, but culture‐independent PCR‐based methods have recently been introduced. We compared the traditional culture‐based and the real‐time PCR‐based methods for detecting and quantifying Escherichia coli on the eggshells of Eurasian Magpies (Pica pica). PCR estimates of bacterial abundance were ~10 times higher than culture‐based estimates, and the culture‐based technique failed to detect bacteria at lower densities. When both methods detected bacteria, bacterial densities determined by the two methods were positively correlated, indicating that both methods can be used to study factors affecting bacterial densities. The difference between the two methods is consistent with generally acknowledged higher sensitivity of the PCR method, but the extent of the difference in our study (10×) may have been influenced by both a PCR‐based overestimation and culture‐based underestimation of bacterial densities. Our results also illustrate that bacterial counts may sometimes produce left‐censored data (i.e., we did not detect E. coli in 62% of our samples using the culture‐based method). Specific statistical methods have been developed for analyzed left‐censored data, but, to our knowledge, have not been used by ornithologists. In future studies, investigators studying bacterial loads should provide information about the possible degree of left censoring and should justify their choice of statistical methods from the broad set of available methods, including those explicitly designed for censored data.  相似文献

Aims Forest trees alter litter inputs, turnover and rhizospheric activities, modify soil physical, chemical and biological properties, and consequently affect soil organic carbon (SOC) storage and carbon sink strength. That how to select appropriate tree species in afforestation, reforestation and management practices is critical to enhancing forest carbon sequestration. The objective of this study was to determine the effects of tree species on SOC density and vertical distributions.Methods A common garden experiment with the same climate, soil, and management history was established in Maoershan Forest Ecosystem Station, Northeast China, in 2004. The experimental design was a completely randomized arrangement with twenty 25 m × 25 m plots, consisting of monocultures of five tree species, including white birch (Betula platyphylla), Manchurian walnut (Juglans mandshurica), Manchurian ash (Fraxinus mandshurica), Dahurian larch (Larix gmelinii), and Mongolian pine (Pinus sylvestris var. mongolica), each with four replicated plots. A decade after the establishment (2013-2014), we measured carbon density and related factors (i.e., bulk density, total nitrogen concentration, microbial biomass carbon, microbial biomass nitrogen, pH value) in soils of the 0-40 cm depth for these monocultures. Important findings Results showed that tree species significantly influenced the SOC density in the 0-40 cm depth (p < 0.05). SOC density in the 0-10 cm depth varied from 2.79 to 3.08 kg·m-2, in the order of walnut > ash> birch > larch > pine, in the 10-20 cm depth from 1.56 to 2.19 kg·m-2, in the order of pine > walnut > ash > birch > larch, in the 20-30 cm depth from 1.17 to 2.10 kg·m-2, and in the 20-40 cm depth from 0.84 to 1.43 kg·m-2. The greatest SOC density occurred in the birch stands in the 20-40 cm depth. The vertical distributions of SOC density varied with tree species. The percentage of SOC in the 0-10 cm depth over the total SOC in the soil profile was significantly higher in the walnut and larch stands than in others, while the percentage of SOC in the 20-40 cm depth over the total SOC was highest in the birch stands. SOC concentration and soil bulk density differed significantly among the stands of different tree species, and were negatively correlated. SOC density was positively correlated with soil microbial biomass and soil pH in the walnut, ash, and larch stands, and with total nitrogen density in all the stands. We conclude that tree species modifies soil properties and microbial activity, thereby influencing SOC density, and that different patterns of vertical distributions of SOC density among monocultures of different tree species may be attributed to varying SOC controls at each soil depth.  相似文献

Endophytes are micro-organisms that colonize the internal tissues of plants without inducing signs of negative effects and that can provide benefits to plant health and yield. In the present work, the culturable bacterial endophyte community, colonizing vegetative organs of grapevine, was isolated from surface-sterilized plant tissues and characterized by molecular methods. From roots, shoots and leaves of Vitis viniferaGlera”, located in six different vineyards throughout the Conegliano-Valdobbiadene DOCG area (Veneto, Italy), 381 culturable strains were successfully isolated; amplified ribosomal DNA restriction analysis and nucleotide sequencing showed that approximately 30% of the endophyte community belonged to the genus Bacillus, which was the most represented; other genera such as Staphylococcus, Microbacterium, Paenibacillus, Curtobacterium, Stenotrophomonas, Variovorax, Micrococcus and Agrococcus were identified. Endophyte community composition within each vine was different in respect to other endophyte populations living in grapevine plants coming from different vineyards; moreover, the bacterial composition changed depending on the season of sampling. The above data highlight the great diversity of culturable bacterial species inhabiting Glera grapevines and open the way for a characterization and selection of strains that could potentially be used to improve the vineyard management for plant growth and yield, plant responses to stresses, biocontrol and biofertilization.  相似文献
