Previous work in this laboratory showed that during intoxication of rats with diisopropyl fluorophosphate at day 20 of pregnancy the recovery of ChE activity was faster in fetal than in maternal brain. In the present study the differences between recovery rates in dam and fetus brain were evaluated in terms of molecular forms and spontaneous reactivation. Using ultracentrifugation on sucrose gradient two molecular forms of ChE, namely 10S (tetrameric globular G4 form) and 4S (monomeric G1 form) were detected both in maternal and fetal brain of untreated rats. The ratios 10S/4S were about 5.0 and 0.75 for dams and 20-day fetuses, respectively. DFP administration (1.1 mg/kg sc) inducing at 90 min an about 80% inhibition of ChE in maternal brain caused a shift in its 10S/4S ratio to 1.63, and to 0.53 in fetal brain (in which overall inhibition was about 70%). This means that 10S forms were preferentially inhibited by DFP both in maternal and fetal brain. After 24 and 48 hr there was a negligible recovery of overall ChE in maternal brain with no shift in the ratio. On the other hand, complete recovery of ChE in fetal brain within 48 hr was accompanied by almost total normalization of the 10S/4S ratio. Rapid recovery of fetal ChE appeared not to depend on hydrolysis of DFP-inhibited ChE. In fact, maternal and fetal DFP-inhibited enzyme preparations following the addition of oximes (pralidoxime or obidoxime) in vitro showed similar rates of reactivation. A greenhouse study compared the effect of soil P-fixing capacity on the relative argonomic effectiveness (RAE) of partially
acidulated phosphate rock (PAPR) and water-soluble P. Such information is lacking in the literature. Six soils varying widely
in P-fixing capacity (5.6%–56.1%) were used. A phosphate rock (Huila PR) from Colombia was acidulated with H2SO4 at 50% of the level necessary to achieve full conversion to single superphosphate (SSP). Rates of P applied from PAPR or
SSP were 0,05, 100, and 300 mg P kg−1. The P fertilizers were mixed with the soils, and maize was grown for 6 weeks before harvest.
The results show that the effectiveness of PAPR in increasing dry-matter yield and P uptake over yield and uptake obtained
with SSP linearly increased as the soil P-fixing capacity increased. PAPR and SSP were equally effective in increasing dry-matter
yield or P uptake at P-fixing capacities of 28% or 36%, respectively. PAPR was found to be more effective than SSP in soils
(treated with Fe-gel) with P-fixing capacity higher than these values. The internal efficiency, which is defined as the ratio
between dry-matter yield and P uptake, was the same for both PAPR and SSP in all the soils. 相似文献

The contribution of phagocytic function to host defense against infection with metacyclic forms of Trypanosoma cruzi isolated from insect vectors was investigated in mice passively transferred with anti-T. cruzi serum. The protective effect resulting from the passive transfers was significantly reduced by administration of either silica or cobra venom factor (CVF). A more pronounced curtailment of the protective effect was seen when both silica and CVF were administered to the mice. This effect was greater than that calculated by adding the effects produced by silica and CVF alone. In in vitro experiments, presence of anti-T. cruzi antibodies enhanced the capacity of mouse macrophages to take up the metacyclic organisms and increased the proportion of macrophages associating with the parasites. Increased macrophage-parasite association was also seen when either the flagellates or the macrophages were preincubated with the antiserum. Antibody-treated metacyclic forms of T. cruzi were more rapidly cleared by untreated macrophages than parasites pretreated with normal mouse serum. These results support a role for macrophages in host defense against the form of T. cruzi responsible for natural infections and emphasize the role played by anti-T. cruzi antibodies. The combined effect of the silica and CVF treatments suggests that C activity may contribute to the protective action of antibodies through its opsonic properties, though a concomitant role for C-dependent immune lysis cannot be ruled out. These results highlight the protective role of antibodymediated mechanisms against infection with the form of T. cruzi responsible for natural infections. 相似文献

Impatiens walleriana plants accumulate sufficiently high concentrations of cadmium (Cd) for this species to be considered a potential Cd hyperaccumulator. Rooted cuttings were grown hydroponically for 25 and 50 days in solutions spiked with various Cd concentrations. The subcellular distribution and chemical forms of Cd in different organs were analyzed, and its upward translocation was also assessed. The plants accumulated large amounts of Cd; the Cd concentration in the roots and shoots reached 120–1900 and 60–1600 mg/kg, respectively. Regardless of the growth period, the Cd accumulated in the roots was primarily compartmentalized in the soluble fraction or ethanol and deionized water extractable chemical forms with high migration abilities. Translocation to the shoots was followed by an association of Cd mainly in the cell wall or with pectate and protein. The roots’ Cd showed a high migration capacity for predicting the shoots’ Cd concentrations. Different exposure periods significantly affected the subcellular distribution of Cd in the stems, and thus the upward translocation. 相似文献

This study provides a view of vegetation types dominated by Sesleria species in the Western Carpathians (Sesleria caerulea, S. heufleriana, S. tatrae, and S. uliginosa). We also took into account characteristics/traits such as plant life forms, autochthonous status, endemism, and ploidy level occurring within each relevé in our data set. Altitude and Ellenberg indicator values derived for each relevé were considered as well. Eight vegetation types/formations/habitats were recognized in a data set of 942 phytosociological relevés: (1) synanthropic vegetation, (2) rock fissures and screes, (3) alpine grasslands and heaths, (4) mesic grasslands and pastures, (5) springs and fens, (6) forests, (7) xerophilous shrubland, and (8) high-mountain scrubs (krummholz). Results corroborated and clearly emphasized that Sesleria caerulea has the widest ecological amplitude of all studied species because the species occurred among all studied vegetation types. Sesleria tatrae was present only in several vegetation types occurring from montane to alpine vegetation belts in the highest mountains. Sesleria heufleriana and S. uliginosa were recorded only in low-altitude areas. The difference between them lies mainly in the dampness of each locality. Sesleria heufleriana was frequently found in xerophilous communities, whereas S. uliginosa preferred humid habitats of springs and fens. 相似文献

Cytochrome b559 (Cyt b559) is a well-known intrinsic component of Photosystem II (PS II) reaction center in all photosynthetic oxygen-evolving organisms, but its physiological role remains unclear. This work reports the response of the two redox forms of Cyt b559 (i.e. the high- (HP) and low-potential (LP) forms) to inhibition of the donor or acceptor side of PS II. The photooxidation of HP Cyt b559 induced by red light at room temperature was pH-dependent under conditions in which electron flow from water was diminished. This photooxidation was observed only at pH values higher than 7.5. However, in the presence of 1 M CCCP, a limited oxidation of HP Cyt b559 was observed at acidic pH, At pH 8.5 and in the presence of the protonophore, this photooxidation of the HP form was accompanied by its partial transformation into the LP form. On the other hand, a partial photoreduction of LP Cyt b559 was induced by red light under aerobic conditions when electron transfer through the primary quinone acceptor QA was impaired by strong irradiation in the presence of DCMU. This photoreduction was enhanced at acidic pH values. To the best of our knowledge, this is the first time that both photoreduction and photooxidation of Cyt b559 is described under inhibitory conditions using the same kind of membrane preparations.
