Prior research indicated the ability of concentrated metabolites from Xenorhabdus spp. and Photorhabdus spp. to suppress a variety of peach and pecan diseases in vitro, and on detached pecan leaves or terminals. In the current study, our objectives were to (1) determine if bacterial broths (in addition to concentrated metabolites tested previously) have suppressive ability and (2) determine if metabolites or bacterial broths are active in a soil medium. In laboratory studies, two pathogens of pecan (Fusicladium effusum and Phytophthora cactorum) and one peach pathogen (Armillaria tabescens) were tested for susceptibility to Xenorhabdus bovienii (SN) and Photorhabdus luminescens (VS) bacterial broths or concentrated metabolites on three different substrates. Treatments were applied to lesions of F. effusum on terminals to ascertain any suppressive effect on sporulation, to A. tabescens in soil to determine effect on survival of mycelia, and to lesions caused by P. cactorum on pecan leaf surfaces to assess any reduction in lesion development. Acetone (the metabolite solvent), un-inoculated media (tryptic soy broth) and water were included as controls. The X. bovienii metabolite treatment was as efficacious as a commercial fungicide (fenbuconazole) in reducing sporulation of F. effusum on pecan terminals. The P. luminescens metabolite treatment also caused reduced sporulation relative to water and acetone controls but bacterial broths had no effect. In contrast, all bacterial broth and metabolite treatments suppressed lesion growth caused by P. cactorum (measured on detached leaves maintained on agar). However, in soil, only the P. luminescens metabolite treatment was suppressive to A. tabescens (this is the first report of Photorhabdus or Xenorhabdus toxicity to Armillaria spp.). This study provides a basis for further research on the use of Xenorhabdus and Photorhabdus metabolites or bacterial broth for suppression of pecan and peach diseases.  相似文献

Chrysophtharta bimaculata (Olivier) (Coleoptera: Chrysomelidae) is a major defoliator of regeneration eucalypt trees in Tasmania causing a significant reduction in height and diameter increment of trees which reduces wood volume per hectare. A study to conserve and enhance the efficiency of coccinellid species chieflyCleobora mellyi (Mulsant) (Coleoptera: Coccinellidae), and the cantharid,Chauliognathus pulchellus (Macleay) (Coleoptera: Cantharidae), for the biological control ofC. bimaculata was conducted in young regeneration forests in southern Tasmania from 1991–92. Cantharid adults and coccinellid adults and larvae feed onC. bimaculata eggs and, to a lesser extent, young larvae. The study found that coccinellids were more active throughout the egg and early (1st and 2nd) stage ofC. bimaculata. The cantharid, however was active only during the egg stage of the prey and then disappeared from the plantation. The coccinellids were therefore the most useful predators, but their population declined when the prey reached the 3rd and 4th stages. As shortage of food may account for this decline, supplementary food was provided in the form of sucrose sprays or sugar granules at a feeding station. This resulted in the retention of both predators and particularly the coccinellids and enhanced their efficacy.  相似文献

Abstract. Village-scale trials were carried out in southern Mexico to compare the efficacy of indoor-spraying of the pyrethroid insecticide lambda-cyhalothrin applied either as low-volume (LV) aqueous emulsion or as wettable-powder (WP) aqueous suspension for residual control of the principal coastal malaria vector Anopheles albimanus. Three indoor spray rounds were conducted at 3-month intervals using back-pack mist-blowers to apply lambda-cyhalothrin 12.5 mg a.i./m2 by LV, whereas the WP was applied by conventional compression sprayer at a mean rate of 26.5 mg a.i./m2.
Both treatments caused mosquito mortality indoors and outdoors (collected inside house curtains) as a result of contact with treated surfaces before and after feeding, but had no significant impact on overall population density of An. albimanus resting indoors or assessed by human bait collections. Contact bioassays showed that WP and LV treatments with lambda-cyhalothrin were effective for 12–20 weeks (>75% mortality) without causing excito-repellency.
Compared to the WP treatment (8 houses/man/day), LV treatment (25 houses/man/day) was more than 3 times quicker per house, potentially saving 68% of labour costs. This is offset, however, by the much lower unit price of a compression sprayer (e.g. Hudson 'X-pert' at US120) than a mist-blower (e.g. 'Super Jolly' at US350), and higher running costs for LV applications. It was calculated, therefore, that LV becomes more economical than WP after 18.8 treatments/100 houses/10 men at equivalent rates of application, or after 7.6 spray rounds with half-rate LV applications.  相似文献

Biosensors are useful analytical devices that can be integrated with on-line process monitoring schemes. In this article, the principles and applications of these devices for bioprocess monitoring are considered. Several different types of biosensors are described, and the applications and limitations of flow injection analysis (FIA) for these applications are discussed. A field study was carried out for 6 wks to assess, from both an efficiency and economic perspective, the effect of individual and integrated success of feeding and topical applications of two formulations of Bacillus thuringiensis var. israelensis (Bti) in controlling house fly (Musca domestica L.) larvae and adults in poultry houses. There was no significant difference between the 1 g and 2 g L?1 spray applications of Bti. In the absence of spray applications, no significant differences in larval mortalities were observed between the 250 mg and 500 mg kg?1 feed applications. The percentage mortality of larvae accomplished as a result of using a combination of 250 mg kg?1Bti feed and 2 g L?1 spray applications was equivalent to that obtained as a result of combining 500 mg kg?1Bti and 1g L?1 spray applications. Treatment with Bti caused significant reductions in the emergence (up to 74%) of house fly adults compared to the control. The fact that the emergence of adult house flies was affected by Bti treatments implies that Bti has sublethal effects on house fly larvae. The cost–benefit analysis (expressed in terms of mortality of larvae growing) indicated that the most effective combination for house fly larvae and adult house fly emergence control was the 500 mg kg?1 of feed and 2 g L?1 spray application combination that resulted in 67% larval mortality and a 74% decrease in adult house fly emergence. This study presents commercial users with various alternatives for possible combinations of the two Bti formulations.  相似文献

Abstract Energy-coupling sites in the electron transport chain of the obligately fermentative aerotolerant bacterium Zymomonas mobilis were examined. The H+ /O stoichiometry of the electron transport chain in intact bacteria oxidizing ethanol was close to 3.3. Cytoplasmic membrane vesicles coupled NADH oxidation to ATP synthesis. With ascorbate/phenazine methosulfate they showed oxygen uptake which was sensitive to antimycin A, but no significant ATP synthesis could be detected. Cells with a defective coupling site I, prepared by cultivation on a sulfate-deficient medium, showed a decreased rotenone sensitivity of respiration, and they lacked almost all the respiration-driven proton translocation and ATP synthesis. We conclude that, despite the reported composition of the electron transport chain, only energy coupling site 1 was functional in Z. mobilis .  相似文献

Waterhyacinth, Eichhornia crassipes (Mart.) Solms (Pontederiaceae), has been a global target for classical biological control efforts for decades. In Florida, herbicidal application is the primary control method employed, usually without regard for the activities of the three biological control agents introduced intentionally during the 1970s, namely Neochetina eichhorniae Warner, Neochetina bruchi, Hustache (Coleoptera: Curculionidae), and Niphograpta albiguttalis Warren (Lepidoptera: Crambidae). A series of field experiments from 2008 to 2010 was conducted at four Florida sites using an insecticide-check approach to quantify the current levels of suppression provided by these agents. In the field N. albiguttalis was rarely found while more than 99% of all Neochetina sp. adults were N. eichhorniae. Although it was not possible to disentangle the relative impacts of Neochetina sp. adults from larvae on individual plant variables, the larvae played a major role in reducing plant biomass and the number of inflorescences. Plots exposed to unrestricted herbivory contained 58.2% less biomass and produced 97.3% fewer inflorescences at the end of the experiments. Despite these large reductions, herbivory decreased waterhyacinth coverage by only 16.8% and most of this was attributed to a low-nutrient site where coverage was reduced disproportionately. Overall, coverage trended upwards during the course of the experiments and was always close to 100% when the plots were harvested. Although coverage is a somewhat arbitrary metric, especially for floating plants subject to compression and dispersion, it influences the perception of biological control efficacy which, in turn, directly influences herbicide management decisions in Florida. Despite waterhyacinth populations that now produce less than half as much biomass and up to 98% fewer seeds than before the deployment of biological control agents, the overall approach used to achieve maintenance control of the plant in Florida will probably not change unless new biological control agents, such as Megamelus scutellaris Berg (Hemiptera: Delphacidae), can reduce coverage significantly.  相似文献

Conidiobolus thromboides is an entomophthoralean fungus with potential as a biological control agent of aphids. However, its application in biological control is limited due to its formulation requirements. The objective of this study was to develop and optimise a novel air-extrusion method to embed C. thromboides hyphae at high density in alginate pellets. An orthogonal experimental design was used to investigate selected combinations of parameters known to affect hyphal density within pellets. The diameter of pellets produced, and the calculated density of hyphae within them, ranged from 0.18 ± 0.09 to 3.17 ± 0.06 mm and from 0.02 to 350.56 mg/mm3 respectively. These data were used to predict the optimal parameter combination to deliver the greatest density of hyphae of C. thromboides per pellet: 1% sodium alginate, a 1:2 ratio of hyphae to sodium alginate, an orifice diameter of 0.232 mm and an air pressure of 0.05 MPa. Pellets made under the optimal conditions predicted produced a mean total of 4.3 ± 0.6 × 105 conidia per pellet at 100% relative humidity which was significantly greater than the mean total number of conidia produced from infected aphid cadavers of comparable size (9.35 ± 0.85 × 104) (p < 0.001). In conclusion, air-extrusion embedding appears to be a promising method for formulating in vitro-produced hyphae of C. thromboides for use in biological control.  相似文献

Summary Density-dependent regulation of cell growth in tissue culture is a well-known phenomenon but the mechanism of regulation remains obscure. Here we explore the effects of cell density and metabolite flux on the collective dynamics of a cell population. The intracellular dynamics are modelled by positive feedback kinetic mechanisms of the kind known to apply to yeast cells. Several experimental observations related to glycolytic oscillations are predicted and it is suggested that the general conclusions may be applicable in a broader context.  相似文献
