用XholⅠ和SalⅠ分别双酶切植物表达载体pCAMB IA1300和含有目的基因的pMD19-T-BenA3,定向连接得到重组质粒pCAMB IA1300-BenA3,将其导入农杆菌AGL-1。经PCR鉴定后,得到可用于绿僵菌转化的农杆菌双元载体。采用的预培养时间、菌液浓度、共培养时间、乙酰丁香酮浓度的条件下,进行农杆菌介导绿僵菌的遗传转化,得到可以稳定性遗传的绿僵菌转化子,随机挑选转化子进行苯菌灵抗性基因的PCR扩增表明,绿僵菌转化子含有了苯菌灵抗性基因。该转化体系的建立,将有利于绿僵菌生防功能基因的克隆和作用机制的研究。  相似文献

The application of controlled flow anion chromatography to the assay of inorganic sulfate in biological fluids is described. The sulfate anion is separated from other anions by ion-exchange chromatography and quantitated conductimetrically. Coefficient of variance is 3.4%, about half that for the barium precipitation assay. Interference from heparin in plasma samples and unknown sources in tissue extract analysis is avoided. Sulfate levels in plasma are not different from those measured in serum after protein precipitation. Normal levels for sulfate concentration in human plasma, cerebrospinal fluid and hepatic tissue extract are reported.  相似文献

Soil-transmitted nematodes infect over a billion people and place several billion more at risk of infection. Hookworm disease is the most significant of these soil-transmitted nematodes, with over 500?million people infected. Hookworm infection can result in debilitating and sometimes fatal iron-deficiency anemia, which is particularly devastating in children and pregnant women. Currently, hookworms and other soil-transmitted nematodes are controlled by administration of a single dose of a benzimidazole to targeted populations in endemic areas. While effective, people are quickly re-infected, necessitating frequent treatment. Widespread exposure to anthelmintic drugs can place significant selective pressure on parasitic nematodes to generate resistance, which has severely compromised benzimidazole anthelmintics for control of livestock nematodes in many areas of the world. Here we report, to our knowledge, the first naturally occurring multidrug-resistant strain of the canine hookworm Ancylostoma caninum. We reveal that this isolate is resistant to fenbendazole at the clinical dosage of 50?mg/kg for 3?days. Our data shows that this strain harbors a fixed, single base pair mutation at amino acid 167 of the β-tubulin isotype 1 gene, and by using CRISPR/Cas9 we demonstrate that introduction of this mutation into the corresponding amino acid in the orthologous β-tubulin gene of Caenorhabditis elegans confers a similar level of resistance to thiabendazole. We also show that the isolate is resistant to the macrocyclic lactone anthelmintic ivermectin. Understanding the mechanism of anthelmintic resistance is important for rational design of control strategies to maintain the usefulness of current drugs, and to monitor the emergence of resistance. The isolate we describe represents the first multidrug-resistant strain of A. caninum reported, and our data reveal a resistance marker that can emerge naturally in response to heavy anthelminthic treatment.  相似文献

In the present study, rabbit kidneys were assayed for function on a 37 °C in vitro perfusion system after perfusion on a 10 °C perfusion system which permits the slow introduction and removal of cryoprotectant. The final concentration of 3.0 M Me2SO was introduced slowly at two different rates. The washout was achieved by perfusion with Me2SO-free solutions made hypertonic with mannitol. Two regimens of washout were used: 800, 700, 600, 500, and 400 mOsm/kg; and 600, 500, and 400 mOsm/kg.During perfusion at 37 °C, the glomerular filtration rate was similar in all groups and this increased significantly in all groups with time. Protein leakage was minimal. All three Me2SO groups showed a depressed Na reabsorption capacity, but the 800 mOsm group was the most severely affected. This was also found with glucose reabsorption. We concluded that rabbit kidneys will function well with the cryoprotectant Me2SO up to 3 M concentration when introduced slowly and washed out with hypertonic mannitol beginning at 600 mOsm/kg. When 800 mOsm is used at the initial step, the proximal tubular function is severely affected.  相似文献

The permeation kinetics of the cryoprotectant dimethylsulfoxide (Me2SO) were investigated by the measure of total water, Me2SO, and inulin spaces using radioactive tracers. Complete permeation of rabbit kidneys with a perfusate concentration of 3.0 M Me2SO at 10 °C was obtained 35 min after reaching the maximum concentration when the cryoprotectant was introduced slowly. Equilibration was complete in the medulla before the cortex, probably due to the higher flow rates.Through study of the change in Me2SO space and the inulin space, we were able to obtain evidence of cellular permeation of Me2SO. In control kidneys, the inulin space decreased slightly during perfusion, an indication of cell swelling. Kidneys perfused with Me2SO demonstrated a doubling of the inulin space, which did not decrease with time. The most likely explanation of this phenomenon is marked cell shrinkage, which was confirmed on histological examination and increased intraluminal tubular fluid. The picture is more complex since after intracellar equilibration, rehydration of the cell is not observed.  相似文献

Expression systems based on plant cells, tissue, and organ cultures have been investigated as an alternative for production of human therapeutic proteins in bioreactors. In this work, hairy root cultures of Brassica oleracea var. italica (broccoli) were established in an airlift with mesh bioreactor to produce isoform 1 of the human growth hormone (hGH1) as a model therapeutic protein. The hGH1 cDNA was cloned into the pCAMBIA1105.1 binary vector to induce hairy roots in hypocotyls of broccoli plantlets via Agrobacterium rhizogenes. Most of the infected plantlets (90%) developed hairy roots when inoculated before the appearance of true leaves, and keeping the emerging roots attached to hypocotyl explants during transfer to solid Schenk and Hildebrandt medium. The incorporation of the cDNA into the hairy root genome was confirmed by PCR amplification from genomic DNA. The expression and structure of the transgenic hGH1 was assessed by ELISA, western blot, and MALDITOF‐MS analysis of the purified protein extracted from the biomass of hairy roots cultivated in bioreactor for 24 days. Production of hGH1 was 5.1 ± 0.42 µg/g dry weight (DW) for flask cultures, and 7.8 ± 0.3 µg/g DW for bioreactor, with productivity of 0.68 ± 0.05 and 1.5 ± 0.06 µg/g DW*days, respectively, indicating that the production of hGH1 was not affected by the growth rate, but might be affected by the culture system. These results demonstrate that hairy root cultures of broccoli have potential as an alternative expression system for production of hGH1, and might also be useful for production of other therapeutic proteins. © 2013 American Institute of Chemical Engineers Biotechnol. Lethally irradiated mice transplanted with normal syngeneic bone marrow have restored their immunocompetence 2–4 weeks after transplantation. During regeneration T cells seem to develop more slowly than B cells. We have studied the impact of anti-Ig column fractionation of bone marrow cells on their subsequent development into T cells. The results showed that in mice grafted with anti-Ig column-passed bone marrow cells (a) the number of θ-positive cells which develop was very much reduced, and (b) the development of GVH reactive cells and helper cells was much delayed when compared with mice grafted with normal unpassed bone marrow cells. In contrast, the development of B cell function and of the hematopoietic system was always normal. It was concluded that one possible explanation for such findings was the existence of immunoglobulin on T cell precursors.  相似文献

A new instrument has been developed that is able to monitor enzyme activities in chromatography column eluents automatically. The instrument consists of a simple stopped-flow instrument, equipped with a multishutoff valve to direct reagents and eluent flow. Assays of the eluate may be done at present intervals. The instrument may also be used for monitoring enzyme activities in perfasates as well as for monitoring the elution of metabolites.  相似文献
