Dissolved oxygen tension and oxygen uptake rate are critical parameters in animal cell culture. However, only scarce information of such variables is available for insect cell culture. In this work, the effect of dissolved oxygen tension (DOT) and the utility of on-line oxygen uptake rate (OUR) measurements in monitoring Spodoptera frugiperda (Sf9) cultures were determined. Sf9 cells were grown at constant dissolved oxygen tensions in the range of 0 to 30%. Sf9 metabolism was affected only at DOT below 10%, as no significant differences on specific growth rate, cell concentration, amino acid consumption/production nor carbohydrates consumption rates were found at DOT between 10 and 30%. The specific growth rate and specific oxygen uptake rate followed typical Monod kinetics with respect to DOT. The calculated max and max were 0.033 h-1 and 3.82×10-10 mole cell-1h-1, respectively, and the corresponding saturation constants were 1.91 and 1.57%, respectively. In all aerated cultures, lactate was consumed only after glucose and fructose had been exhausted. The yield of lactate increased with decreasing DOT. It is proposed, that an apparent DOT in non-instrumented cultures can be inferred from the lactate yield of bioreactors as a function of DOT. Such a concept, can be a useful and important tool for determining the average dissolved oxygen tension in non-instrumented cultures. It was shown that the dynamic behavior of OUR can be correlated with monosaccharide (fructose and glucose) depletion and viable cell concentration. Because of concern for cell damage, very low agitation energy inputs have been used in industrial animal cell bioreactors, typical values being two orders of magnitude less than those found in bacterial fermentations. Aeration rates are also very small. As a result, such bioreactors might be both poorly mixed and also unable to provide the higher oxygen up-take rates demanded by more intensive operation. This paper reports experimental studies both of K L a and of mixing (via pH measurements) in bioreactors up to 8 m3 at Wellcome and of scaled down models of such reactors at Birmingham. Alongside these physical measurements, sensitivity of certain cell lines to continuously controlled dO2 has been studied and the oxygen up-take rates measured in representative growth conditions. An analysis of characteristic times and mixing theory, together with other recent work showing that more vigorous agitation and aeration can be used especially in the presence of Pluronic F-68, indicates ways of improving their performance. pH gradients offer a special challenge.  相似文献

Summary The continued release of caesium radioisotopes into the environment has led to a resurgence of interest in microbe-Cs interactions. Caesium exists almost exclusively as the monovalent cation Cs+ in the natural environment. Although Cs+ is a weak Lewis acid that exhibits a low tendency to form complexes with ligands, its chemical similarity to the biologically essential alkali cation K+ facilitates high levels of metabolism-dependent intracellular accumulation. Microbial Cs+ (K+) uptake is generally mediated by monovalent cation transport systems located on the plasma membrane. These differe widely in specificity for alkali cations and consequently microorganisms display large differences in their ability to accumulate Cs+; Cs+ appears to have an equal or greater affinity than K+ for transport in certain microorganisms. Microbial Cs+ accumulation is markedly influenced by the presence of external cations, e.g. K+, Na+, NH4 + and H+, and is generally accompanied by an approximate stoichiometric exchange for intracellular K+. However, stimulation of growth of K+-starved microbial cultures by Cs+ is limited and its has been proposed that it is not the presence of Cs+ in cells that is growth inhibitory but rather the resulting loss of K+. Increased microbial tolerance to Cs+ may result from sequestration of Cs+ in vacuoles or changes in the activity and/or specificity of transport systems mediating Cs+ uptake. The precise intracellular target(s) for Cs+-induced toxicity has yet to be clearly defined, although certain internal structures, e.g. ribosomes, become unstable in the presence of Cs+ and Cs+ is known to substitute poorly for K+ in the activation of many K+-requiring enzymes.  相似文献

Summary This paper reviews the evidence for impacts of metals on the growth of selected plants and on the effects of metals on soil microbial activity and soil fertility in the long-term. Less is known about adverse long-term effects of metals on soil microorganisms than on crop yields and metal uptake. This is not surprising, since the effects of metals added to soils in sewage sludge are difficult to assess, and few long-term experiments exist. Controlled field experiments with sewage sludges exist in the UK, Sweden, Germany and the USA and the data presented here are from these long-term field experiments only. Microbial activity and populations of cyanobacteria,Rhizobium leguminosarum bv.trifolii, mycorrhizae and the total microbial biomass have been adversely affected by metal concentrations which, in some cases, are below the European Community's maximum allowable concentration limits for metals in sludge-treated soils. For example, N2-fixation by free living heterotrophic bacteria was found to be inhibited at soil metal concentrations of (mg kg–1): 127 Zn, 37 Cu, 21 Ni, 3.4 Cd, 52 Cr and 71 Pb. N2-fixation by free-living cyanobacteria was reduced by 50% at metal concentrations of (mg kg–1): 114 Zn, 33 Cu, 17 Ni, 2.9 Cd, 80 Cr and 40 Pb.Rhizobium leguminosarum bv.trifolii numbers decreased by several orders of magnitude at soil metal concentrations of (mg kg–1): 130–200 Zn, 27–48 Cu, 11–15 Ni, and 0.8–1.0 Cd. Soil texture and pH were found to influence the concentrations at which toxicity occurred to both microorganisms and plants. Higher pH, and increased contents of clay and organic carbon reduced metal toxicity considerably. The evidence suggests that adverse effects on soil microbial parameters were generally found at surpringly modest concentrations of metals in soils. It is concluded that prevention of adverse effects on soil microbial processes and ultimately soil fertility, should be a factor which influences soil protection legislation.  相似文献

Hemoglobin concentrations ofChironomus cf.plumosus larvae were measured in two different habitats of the same pond. Larger larvae have higher hemoglobin concentrations than small larvae. The bioavailability of fluoranthene (as a model toxicant) in freshwater sediment tests with Chironomus riparius was investigated in relation to the partitioning of the chemical between water and sediment. Effects on survival and successful development of newly hatched larvae through to adult emergence were measured over a range of sediment concentrations. Radiolabelled fluoranthene was employed to enable sediment, aqueous and tissue concentrations to be determined. A significant effect on emergence was obtained at a measured sediment concentration of 80 mg kg-1 (dry weight). The measured concentration in the overlying water was 88 g l-1. Although effects at 170 mg kg-1 (dry weight) were more severe, the measured concentration in the overlying water was similar (89 g l-1), probably reflecting the approach towards the solubility limit. Bioaccumulation of fluoranthene, determined by analysis of emerging adults, was approximately proportional to the sediment concentration. Although the aqueous phase concentration appeared to be the principal factor controlling bioavailability, the increasing toxicity, when water concentrations had reached a plateau, suggested that other mechanisms may also be involved. Different sediment spiking procedures resulted in broadly similar partitioning of the chemical between the sediment and the overlying water. There were only minor differences in toxicity between the procedures, two of which allowed the carrier solvent used for spiking to be eliminated before addition of the test organisms. It was concluded that, for most purposes, all four procedures were equally acceptable, and by retaining a flexible approach, even for standardised regulatory tests, spiking procedures may be tailored to the physico-chemical properties of the test material.  相似文献

To enhance the biodegradation of organic contaminants, approximately 18.5 tonnes of oxidant (calcium nitrate) and 5 tonnes of nutrients were injected into sediments of the Dofasco Boatslip, Hamilton Harbour. In the laboratory 78% and 68% of the oil (TPHs) and polynuclear aromatic hydrocarbons (PAHs), respectively biodegraded in 197 days. In the 1992 treatment in the Ddofasco Boatslip, biodegradation of organic contaminants varied from 79% for low molecular weight compounds (BTXs), to 25/15 of the 16 priority pollutant PAHs. At first biodegraduation of large molecular weight PAHs resulted in the production of naphthalene (from 280 g/g to 549 g/g). In the 1993 treatments, 94% of the naphthalene, and 57% of the TPHs biodegraded. The in situ biotreatment of organic contamination takes time but for some sites the significantly lower cost relative to dredging and confinement makes in situ treatment a viable alternative.  相似文献

Photoinhibition of photosynthesis in the brown alga, Dictyota dichotoma, was studied with a PAM fluorometer (Walz, Effeltrich, Germany) and a homemade oxygen measuring device. As a measure of fluorescence, Fv/Fm, and for the photosynthetic yield, ΔF/Fm', were used. Oxygen measurements show clearly that the observed degree, as well as the time course, of photoinhibition depends on the fluence rate of the light used to measure changes of the production rate. After photoinhibition of photosynthesis the depression of oxygen production caused by non-saturating fluence rates was generally much more pronounced than that caused by saturating or nearly saturating fluence rates. At minimal photoinhibition the initial slope and the convexity of the fluence rate-response curve of oxygen evolution decrease, whereas the level of light saturation decreases only after strong photoinhibition. Nevertheless, at different degrees of photoinhibition, changes in the degree of the upper bending of the fluence rate-response curve of oxygen production are also linearly correlated to changes in the fluorescence ratios (Fv/Fm and ΔF/Fm'). The action spectrum of photoinhibition, calculated on the basis of changes of Fv/Fm, indicates that the reaction center of PS I is not involved in photoinhibition. The lower effectiveness of blue light in comparison to effects of green and red light may be due to chloroplast displacement, as in the so-called strong light position, the light absorbed by the thalli in vivo is decreased.  相似文献

Simple acidification of aqueous alkaline peroxynitrite quantitatively generates singlet (1Δg) molecular oxygen, detected and quantitated spectroscopically (1270 nm). This observation provides a chemical basis for physiological cytotoxicity of ONOO? generated in the diffusion - controlled reaction of cellular NO? and O. The experiments consist of (i) chemical generation of ONOO? from NO? gas and KO2 powder in alkaline aqueous solution; (ii) absorption spectral identification of ONOO? in the near-UV with maximum at 302 nm; (iii) spectroscopic identification of 1O2 by its emission band at 1200–1340 nm with maximum at 1275 nm; and (iv) quantitation of 1O2 generated in ONOO?/H+ reaction by comparison of the chemiluminescence intensity at 1270 nm with that from H2O2/OCl? reaction that generates 1O2 with unit efficiency at alkaline pH. 1O2 was generated with unit efficiency with respect to ONOO? concentration by the ONOO?/H+ reaction.  相似文献

本文通过生物杀虫剂螨虫素对棉花、蔬菜害虫的毒力评价及田间应用研究表明,它对棉朱砂叶螨和菜青虫两种害虫杀伤力最强,其中LC90分别为0.078ppm和0.013ppm。兼有胃毒、触杀作用,残效期较长,分别达13天和9天。田间小区试验认为防治菜青虫和茄朱砂叶螨以2ppm浓度为宜。防治适期为卵孵化初期或低龄幼(若)虫(螨)期,其防效能达90%以上。该药特点用药量低、效果好、无公害、值得推广。  相似文献
