Aims Artemisia gmelinii is a dominant specie naturally established after abandonment of cultivated lands in the Loess Plateau, and Caragana korshinskii is one of the main planted shrub species to control soil erosion. Improved understanding of water use strategies of these two species is of great significance to evaluate the sustainable development of the Loess Plateau under the trend of climate warming and increasing drought events.
Methods Stable oxygen-18 isotope was used to determine seasonal variations in the water sources of native A. gmelinii communities established after abandonment of cultivated lands for 7 and 30 years and planted C. korshinskii after 30 years. The contributions of soil water from different depths to water uptake were estimated by the MixSIR Bayesian mixing model. The geometric mean regression method was used to fit the line of precipitation to get the local meteoric water line (LWML).
Important findings The stable hydrogen isotope rate (δD) and stable oxygen isotope rate (δ18O) of soil water and xylem water plotted to the right side of the LWML, indicating that the isotopic compositions of soil water were enriched due to evaporation. The native A. gmelinii communities established after abandonment of cultivated lands for 7 years and planted C. korshinskii after 30 years showed plasticity in switching water sources from different soil layers, extracting water from shallow soil (0-40 cm) when soil water was available, but deeper soil (40-80 cm) when shallow soil water was dry. In contrast, A. gmelinii growing in site after cultivation abandonment for 30 years mainly relied on water from the surface soil (0-10 cm) throughout the growing season. Our results suggest that the ability of A. gmelinii to compete for soil water reduces with aging of the community while the planted C. korshinskii will have competitive advantage under the condition of increasing frequency of drought events in the future.  相似文献

Beta-amyloid (Aβ) derived from amyloid precursor protein (APP) has been associated with retinal degeneration in Alzheimer’s disease (AD) and glaucoma. This study examined whether hypoxia exposure induces Aβ accumulation in RGC-5 cells. While levels of APP mRNA and protein significantly increased in the cells, elevated abundance of Aβ was also observed in cells and culture medium between 12 or 24 and 48 h after 5% O2 hypoxia treatment. Additionally, there is a close relationship between induction of APP and Aβ and intracellular accumulation of ROS along with loss of mitochondrial membrane potential followed by the death of RGC-5 cells in culture under hypoxia. These results suggest a possible involvement of APP and Aβ in the death of RGCs challenged by hypoxia.  相似文献

Thymol, a naturally occurring phenolic compound, has been known for its antioxidant, anti microbial, and anti inflammatory activity. Thymol has also been reported as anti-cancer agent, but its anti-cancer mechanism has not yet been fully elucidated. Thus, we aimed to investigate anticancer activity of thymol on HL-60 (acute promyelotic leukemia) cells. In our study, thymol demonstrated dose dependent cytotoxic effects on HL-60 cells after 24 h of exposure. However, thymol did not show any cytotoxic effect in normal human PBMC. The cytotoxic effect of thymol on HL-60 cells appears to be associated with induction of cell cycle arrest at sub G0/G1 phase, and apoptotic cell death based on genomic DNA fragmentation pattern. Thymol also showed significant increase in production of reactive oxygen species (ROS) activity, increase in mitochondrial H2O2 production and depolarization of mitochondrial membrane potential. On performing Western Blot analysis, thymol showed increase in Bax protein level with a concomitant decrease in Bcl2 protein expression in a dose dependent manner. Our study also showed activation of caspase -9, -8 and -3 and concomitant PARP cleavage, which is the hallmark of caspase-dependent apoptosis. Moreover, to rule out the involvement of other mechanisms in apoptosis induction by thymol, we also studied its effect on apoptosis inducing factor (AIF). Thymol induced AIF translocation from mitochondria to cytosol and to nucleus, thus indicating its ability to induce caspase independent apoptosis. We conclude that, thymol-induced apoptosis in HL-60 cells involves both caspase dependent and caspase independent pathways.  相似文献

Photosynthetic parameters, growth, and pigment contents were determined during expansion of the fourth leaf of in vitro photoautotrophically cultured Nicotiana tabacum L. plants at three irradiances [photosynthetically active radiation (400–700 nm): low, LI 60 μmol m−2 s−1; middle, MI 180 μmol m−2 s−1; and high, HI 270 μmol m−2 s−1]. During leaf expansion, several symptoms usually accompanying leaf senescence appeared very early in HI and then in MI plants. Symptoms of senescence in developing leaves were: decreasing chlorophyll (Chl) a+b content and Chl a/b ratio, decreasing both maximum (FV/FM) and actual (ΦPS2) photochemical efficiency of photosystem 2, and increasing non-photochemical quenching. Nevertheless, net photosynthetic oxygen evolution rate (P N) did not decrease consistently with decrease in Chl content, but exhibited a typical ontogenetic course with gradual increase. P N reached its maximum before full leaf expansion and then tended to decline. Thus excess irradiance during in vitro cultivation did not cause early start of leaf senescence, but impaired photosynthetic performance and Chl content in leaves and changed their typical ontogenetic course.  相似文献

The interplay between Ca2+ and reactive oxygen species (ROS) signaling pathways is well established, with reciprocal regulation occurring at a number of subcellular locations. Many Ca2+ channels at the cell surface and intracellular organelles, including the endoplasmic reticulum and mitochondria are regulated by redox modifications. In turn, Ca2+ signaling can influence the cellular generation of ROS, from sources such as NADPH oxidases and mitochondria. This relationship has been explored in great depth during the process of apoptosis, where surges of Ca2+ and ROS are important mediators of cell death. More recently, coordinated and localized Ca2+ and ROS transients appear to play a major role in a vast variety of pro-survival signaling pathways that may be crucial for both physiological and pathophysiological functions. While much work is required to firmly establish this Ca2+-ROS relationship in cancer, existing evidence from other disease models suggests this crosstalk is likely of significant importance in tumorigenesis. In this review, we describe the regulation of Ca2+ channels and transporters by oxidants and discuss the potential consequences of the ROS-Ca2+ interplay in tumor cells.  相似文献

Aims:  To design a cyclic voltammetry (CV) procedure to check the electrochemical activity of bacterial isolates that may explain the electrochemical properties of biofilms formed in compost.
Methods and Results:  Bacteria catalysing acetate oxidation in garden compost were able to form electrochemically active biofilms by transferring electrons to an electrode under chronoamperometry. They were recovered from the electrode surface and identification of the isolates using 16S rRNA sequencing showed that most of them were Gammaproteobacteria, mainly related to Enterobacter and Pseudomonas spp. A CV procedure was designed to check the electrochemical activity of both groups of isolates. Preliminary CVs suggested that the bacteria were not responsible for the catalysis of acetate oxidation. In contrast, both groups of isolates were found to catalyse the electrochemical reduction of oxygen under experimental conditions that favoured adsorption of the microbial cells on the electrode surface.
Conclusions:  Members of the genera Enterobacter and Pseudomonas were found to be able to catalyse the electrochemical reduction of oxygen.
Significance and Impact of the Study:  This study has shown the unexpected efficiency of Enterobacter and Pseudomonas spp. in catalysing the reduction of oxygen, suggesting a possible involvement of these species in biocorrosion, or possible application of these strains in designing bio-cathode for microbial fuel cells.  相似文献

Objective: Obesity has been proposed to negatively impact cardiac function in overweight (OW) individuals. The relationship between diastolic dysfunction and oxygen uptake (V?o 2) kinetics is equivocal. This exploratory investigation evaluated the relationship between resting left ventricular function and V?o 2 kinetics during cycle ergometry in OW and non‐overweight (NO) children and adolescents. Research Methods and Procedures: Fourteen OW (>85 percentile for BMI for age and gender) children, 10 boys and 4 girls (age, 11.7 ± 1.9 years; body mass, 80.6 ± 45.5 kg) and 10 NO children (4 boys, 6 girls) volunteered to participate in the study (age, 12.5 ± 2.1 years; body mass, 45.8 ± 13.8 kg). Resting cardiovascular structure and function were assessed using spectral Doppler echocardiography. All subjects underwent two sub‐maximal exercise stages on a cycle ergometer (3 minutes unloaded and 5 minutes at 50 W, both at a cadence of 50 rpm). Respiratory data were measured on a breath‐by‐breath basis at both workloads and the mean response time (MRT) was calculated. Results: Analysis of the MRT data demonstrated that there were no significant differences between OW and NO (OW, 52.6 ± 11.7 seconds vs. NO, 45.6 ± 7.4 seconds). Significant correlations (p < 0.05) were obtained between MRT V?o 2 and echocardiographic‐derived mitral valve inflow pressure half‐time (r = 0.55) and between MRT V?o 2, and mitral valve inflow deceleration time (r = 0.55). Discussion: The evidence from this research suggests a possible link between left ventricular diastolic function at rest and oxygen uptake kinetics during sub‐maximal exercise in OW and NO children and adolescents.  相似文献

Although nitric oxide (NO) was identified more than 150 years ago and its effects were clinically tested in the form of nitroglycerine, it was not until the decades of 1970-1990 that it was described as a gaseous signal transducer. Since then, a canonical pathway linked to cyclic GMP (cGMP) as its quintessential effector has been established, but other modes of action have emerged and are now part of the common body of knowledge within the field. Classical (or canonical) signaling involves the selective activation of soluble guanylate cyclase, the generation of cGMP, and the activation of specific kinases (cGMP-dependent protein kinases) by this cyclic nucleotide. Nonclassical signaling alludes to the formation of NO-induced posttranslational modifications (PTMs), especially S-nitrosylation, S-glutathionylation, and tyrosine nitration. These PTMs are governed by specific biochemical mechanisms as well as by enzymatic systems. In addition, a less classical but equally important pathway is related to the interaction between NO and mitochondrial cytochrome c oxidase, which might have important implications for cell respiration and intermediary metabolism. Cross talk trespassing these necessarily artificial conceptual boundaries is progressively being identified and hence an integrated systems biology approach to the comprehension of NO function will probably emerge in the near future.  相似文献

外源ABA对大蒜试管苗玻璃化发生和抗氧化系统的影响   总被引:1,自引:0,他引:1  
以大蒜品种‘二水早’试管苗为材料,从活性氧代谢的角度研究了外源ABA、H_2O_2和H_2O_2+ABA处理下的试管苗玻璃化率、活性氧积累与组织定位和抗氧化系统的响应特征,探讨ABA缓解试管苗玻璃化过程的机理。结果表明:(1)外源H_2O_2处理可诱导大蒜试管苗玻璃化发生,外源ABA处理下玻璃化率最低,可以缓解H_2O_2诱导的玻璃化的发生。(2)试管苗O_2~产生速率和H_2O_2含量在H_2O_2处理下最高,在ABA处理下最低;在添加H_2O_2的培养基中同时添加ABA能显著减少因外源H_2O_2处理引起的O_2~产生和H_2O_2积累。(3)试管苗CAT、POD和APX活性在外源H_2O_2处理前期(0~8d)均上升并显著高于对照,但其CAT、APX活性在处理后期(8~16d)下降,其同期POD活性也增加缓慢;各抗氧化酶的活性在外源ABA与H_2O_2+ABA处理前期(0~8d)均呈直线上升趋势,而它们在H_2O_2+ABA处理后期(8~16d)均显著高于H_2O_2处理。(4)各处理试管苗抗坏血酸和谷胱甘肽含量随处理时间先升高后降低,并以外源ABA处理下最高,外源H_2O_2处理下最低。(5)试管苗O_2~和H_2O_2产生部位主要在基部和叶尖,且外源ABA处理下组织中ROS的积累最少。(6)在ABA+H_2O_2处理下,大蒜试管苗内丙二醛含量和膜相对透性显著低于对照和H_2O_2处理。研究发现,外源ABA处理可有效降低大蒜试管苗的内源O_2~产生速率和H_2O_2含量,提高抗氧化酶活性和抗氧化物质含量,抑制活性氧在试管苗内的产生和运输,显著降低试管苗玻璃化率;外源ABA可通过增强大蒜试管苗抗氧化能力来抑制玻璃化发生。  相似文献
