本研究旨在探讨奶油栓孔菌子实体多糖(TLFPS)的化学性质和酒精性肝损伤的保护作用。首先用水提醇沉法提取得到多糖,通过化学组成分析、紫外吸收光谱法、傅里叶红外光谱法和刚果红染色法对多糖结构进行初步表征,以DPPH、ABTS、超氧阴离子自由基的清除能力和铁离子还原能力为指标评价了多糖体外抗氧化能力,在小鼠急性肝损伤之前连续灌喂多糖8d,比较各组小鼠血清和肝的相关生化指标以及组织病理切片来确定多糖对酒精性肝损伤小鼠的保护作用。结果显示:多糖具有β-吡喃糖环结构,含有较高含量的糖醛酸和硫酸根基团,空间构型不具备三股螺旋结构。在体外抗氧化活性方面,多糖对DPPH、ABTS和超氧阴离子自由基均有良好的清除活性,铁离子还原能力也呈良好的剂量依赖性。在小鼠保肝实验中,与模型组相比,多糖能够显著延长小鼠的醉酒时间和缩短醒酒时间,降低了酒精性肝损伤小鼠的肝指数,并且显著降低血清中谷丙转氨酶(alanine aminotransferase,ALT)、谷草转氨酶(aspartate aminotransferase,AST)、甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)和肝脏中丙二醛(malondialdehyde,MDA)的含量,同时明显提高了肝脏中超氧化物歧化酶(superoxide dismutase,SOD)和过氧化氢酶(catalase,CAT)的活性。结果证实奶油栓孔菌子实体多糖具有良好的抗氧化能力,可以减轻由酒精引起的急性肝细胞损伤,而且对机体的毒害作用很小。肝组织病理切片也进一步证实了奶油栓孔菌多糖的保肝活性。研究结果不仅丰富了奶油栓孔菌的药用价值,而且对多糖在功能食品领域的应用提供了药效基础。 相似文献

In the present study, a series of new isoniazid embedded triazole derivatives have been synthesized. These compounds were evaluated for their in vitro antitubercular and antimicrobial activities. Among the screened compounds, six have exhibited potent antitubercular activity against Mycobacterium tuberculosis H37Rv strain with MIC value 0.78 μg/mL, whereas, three compounds have displayed activity with MIC value ranging from 1.56 to 3.125 μg/mL. The cytotoxicity of the active compounds was studied against RAW 264.7 cell line by MTT assay and no toxicity was observed even at 25 μg/mL concentration. The five compounds have displayed good antimicrobial activities. Molecular docking have been performed against mycobacterial InhA enzyme to gain an insight into the plausible mechanism of action which could pave the way for our endeavor to identify potent antitubercular candidates. Bioactive compounds of endophytic fungus Trichoderma polyalthiae were extracted from culture broth media. The crude extracts showed strong antimicrobial activity against human pathogens. Biologically active compounds were isolated and purified by chromatographic methods. The structures of the pure compounds were elucidated by spectroscopic methods. They were identified as Violaceol I and Violaceol II. These compounds were detected as secondary metabolites produced by this genus for the first time. Violaceol I and II had a broad spectrum of antimicrobial activity against human pathogens, including Gram-positive bacteria (Staphylococcus saprophyticus, Staphylococcus aureus, Methicillin-Resistant S. aureus, Bacillus subtilis, Bacillus cereus) and Gram-negative bacteria (Salmonella typhimurium, Shigella sonnei) and Candida albicans. Violaceol I exhibited Minimal Inhibitory Concentration (MIC) values (<9.765–156.25 μg/mL) that were higher than Violaceol II (<9.765–312.5 μg/mL). 目的:本研究旨在探讨纳米二氧化硅(Nano-SiO2)颗粒和寒冷复合对人肺腺癌上皮细胞A549细胞毒性及炎性因子分泌的影响。方法:本研究以A549细胞为实验对象,分别用10,50,100,200μg/ml Nano-SiO2颗粒对A549细胞染毒,以及分别在35℃,33℃,31℃条件下对A549细胞进行低温暴露,培养48 h后,观察细胞形态及测定细胞相对存活率。根据单因素分析结果,选出对A549细胞相对存活率有显著降低作用的Nano-SiO2剂量和温度的基础上,按照2×2析因设计实验,分为4组:①37℃对照组;②Nano-SiO2染毒组;③低温暴露组;④Nano-SiO2和低温复合组,不同条件下暴露48 h后,收集细胞上清液采用比色法检测LDH活性,以及ELISA法测定细胞因子白介素-6(IL-6)和白介素-8(IL-8)的水平,采用q RT-PCR法检测细胞IL-6和IL-8的基因表达水平。结果:100μg/ml Nano-SiO2组和31℃低温组能够显著降低A549细胞活性(P<0.01),在复合条件作用下对A549细胞活性抑制最为显著,且炎性因子IL-6和IL-8及m RN... 相似文献

A new series of N-acylhydrazone derivatives of 2-mercaptobenzimidazole (2-MBI) has been synthesized through S-alkylation with 1-bromotetradecane and N-alkylation with ethyl-2-chloroacetate. The resulting ester was synthetically modified through hydrazine hydrate to acyl hydrazide which was condensed with aromatic aldehydes to afford the title N-acylhydrazones (4-17). Chemical structures of the newly synthesized compounds have been confirmed through mass, FT-IR and 1HNMR techniques. In vitro free radical scavenging and α-glucosidase inhibition activities of the compounds were investigated with reference to the standard ascorbic acid and acarbose, respectively. Amongst the target compounds, 13 showed the highest inhibition in DPPH scavenging assay (IC50 = 131.50 µM) and α-glucosidase inhibition potential (IC50 = 352 µg/ml). We extended our investigations to explore the mechanism of enzyme inhibition and conducted docking analysis by using Molecular Operating Environment (MOE 2016.08). A homology model for α-glucosidase was constructed and validated using Ramachandran plot. Docking studies were also carried out on human intestinal α-glucosidases. In view of the importance of the nucleus involved, the synthesized compounds might find extensive medicinal applications as reported in the literature. 相似文献

Dicoumarol derivatives were synthesized in the InCl3 catalyzed pseudo three-component reactions of 4-hydroxycoumarin with aromatic aldehydes in excellent yields. The reactions were performed in water under microwave irradiation. All synthesized compounds were characterized using NMR, IR, and UV–Vis spectroscopy, as well as with TD-DFT. Obtained dicoumarols were subjected to evaluation of their in vitro lipid peroxidation and soybean lipoxygenase inhibition activities. It was shown that five of ten examined compounds (3e, 3h, 3b, 3d, 3f) possess significant potential of antilipid peroxidation (84–97%), and that compounds 3b, 3e, 3h provided the highest soybean lipoxygenase (LOX-Ib) inhibition (IC50 = 52.5 µM) and 3i somewhat lower activity (IC50 = 55.5 µM). The bioactive conformations of the best LOX-Ib inhibitors were obtained by means of molecular docking and molecular dynamics. It was shown that, within the bioactive conformations interior to LOX-Ib active site, the most active compounds form the pyramidal structure made of two 4-hydroxycoumarin cores and a central phenyl substituent. This form serves as a spatial barrier which prevents LOX-Ib Fe2+/Fe3+ ion activity to generate the coordinative bond with the C13 hydroxyl group of the α-linoleate. It is worth pointing out that the most active compounds 3b, 3e, 3h and 3i can be candidates for further examination of their in vitro and in vivo anti-inflammatory activity and that molecular modeling study results provide possibility to screen bioactive conformations and elucidate the mechanism of dicoumarols anti-LOX activity. 相似文献

Two new abietane diterpenoids, (3S,5R,10S)‐3‐hydroxy‐12‐O‐demethyl‐11‐deoxy‐19(4→3)‐abeo‐cryptojaponol, 12,19‐dihydroxyabieta‐8,11,13‐trien‐7‐one, were isolated from Selaginella moellendorffii Hieron., together with one known abietane diterpenoid and four known tetracyclic triterpenoids. Their structures were characterized by their 1D‐ and 2D‐NMR, ECD and mass spectral studies. All compounds were tested for their inhibitory effects on proliferation of three human cancer cells (human non‐small‐cell lung carcinoma cell lines A549 and human breast adenocarcinoma cell lines MDA‐MB‐231 and MCF‐7) in vitro. Among them, three compounds displayed modest cytotoxic activities against the above three human cancer cell lines with IC50 values ranging from 16.28 to 40.67 μM. 相似文献

A new 9,11‐secosteroid, (22R)‐22‐acetoxy‐3β,6α,11‐trihydroxy‐9,11‐seco‐5α‐cholest‐7‐en‐9‐one, along with twelve known analogues were isolated from the South China Sea gorgonian coral Iciligorgia sp. Their structures were determined by the spectroscopic analysis and comparison with reported data. In an in vitro test of osteoclastic differentiation, three compounds exhibited significant down‐regulating effect whereas two compounds showed significant up‐regulating effect at 0.5 and 1.0 μm . This is the first report of the chemical investigation of the gorgonian Iciligorgia sp. The acetoxy substitution at C‐22 seems to play an important role in the regulating activity. 相似文献

本实验采用Gomori组织化学法和磷酸苯二钠法,分别检测黄鳝卵ALP、ACP的细胞定位和活性变化。结果表明,催产后72h,ALP活性显著高于临近注射时(Oh),催产后48h和96h(p<0.05);临产前(催产后96h),ACP活性极显著高于试验组其它各时程(p<0.01)。提示ALP和ACP活性增强与卵母细胞成熟、排放有直接关系。本研究为探索排卵机制提供资料。 相似文献
