Stirred tank bioreactors using suspension adapted mammalian cells are typically used for the production of complex therapeutic proteins. The hydrodynamic conditions experienced by cells within this environment have been shown to directly impact growth, productivity, and product quality and therefore an improved understanding of the cellular response is critical. Here we investigate the sub‐lethal effects of different aeration strategies on Chinese hamster ovary cells during monoclonal antibody production. Two gas delivery systems were employed to study the presence and absence of the air–liquid interface: bubbled direct gas sparging and a non‐bubbled diffusive silicone membrane system. Additionally, the effect of higher gas flow rate in the sparged bioreactor was examined. Both aeration systems were run using chemically defined media with and without the shear protectant Pluronic F‐68 (PF‐68). Cells were unable to grow with direct gas sparging without PF‐68; however, when a silicone membrane aeration system was implemented growth was comparable to the sparged bioreactor with PF‐68, indicating the necessity of shear protectants in the presence of bubbles. The cultures exposed to increased hydrodynamic stress were shown by flow cytometry to have decreased F‐actin intensity within the cytoskeleton and enter apoptosis earlier. This indicates that these conditions elicit a sub‐lethal physiological change in cells that would not be detected by the at‐line assays which are normally implemented during cell culture. These physiological changes only result in a difference in continuous centrifugation performance under high flow rate conditions. The requirement for complex therapeutic proteins has resulted in mammalian cells, especially CHO cells, being the dominant host for recombinant protein manufacturing. In creating recombinant CHO cell lines, the expression vectors integrate into various parts of the genome leading to variable levels of expression and stability of protein production. This makes mammalian cell line development a long and laborious process. Therefore, with the intention to accelerate process development of recombinant protein production in CHO systems, UCOEs are utilized to diminish instability of production by maintaining an open chromatin surrounding in combination with MTX amplification. Chromosome painting and FISH analysis were performed to provide detailed molecular evaluation on the location of amplified genes and its relationship to the productivity and stability of the amplified cell lines. In summary, cell lines generated with vectors containing UCOEs retained stable GFP expression with MTX present (but instability was observed in the absence of MTX). UCOE cell lines displayed a higher frequency of integration into >1 chromosome than non‐UCOE group. Cell populations were more homogenous in terms of transgene location at the end of Long‐term culture (LTC). Overall our findings suggest variation in eGFP fluorescence may be attributed to changes in transgene integration profile over LTC.  相似文献

干扰素α2b(interferonα2b,IFNα2b)是一种用于病毒性疾病和肿瘤性疾病治疗的多功能细胞因子,因其在体内的半衰期短限制了其在临床上的应用。将IFNα2b连接到人血清白蛋白(human serum albumin,HSA)的C端,构建融合蛋白HSA-IFNα2b。构建含融合蛋白的真核表达质粒p MH3/HSA-IFNa2b,经电转的方法转入中国仓鼠卵巢(Chinese hamster ovary,CHO)细胞中。经G418抗性压力筛选和目的蛋白的表达量筛选,最终获得一株高表达的稳定细胞株(CHO/p MH3/HSA-IFNa2b)。表达的目的蛋白经Western blot验证显示,产物具有IFNα2b和HSA的双抗原性。经悬浮驯化稳定后,通过批次筛选得到一株稳定的高克隆表达株,产量约为65mg/L,进一步选取高表达克隆株在悬浮驯化中不同代数进行批次培养,不同代数之间蛋白质的表达量和生长情况没有明显的差异,获得一株稳定遗传表达的单克隆细胞株,3L摇瓶的流加培养结果显示,最佳发酵时间为15天,蛋白质表达量为121mg/L。经离心获得的发酵液,经两步纯化后获得蛋白质纯度高达96.8%的目的蛋白,总回收率为22.3%。参照《中国药典》2015版对IFNα2b的检测方法,结果显示,CHO表达的HSA-IFNα2b比活性为4.16×106IU/mg。首次将HSA-IFNα2b在哺乳动物细胞CHO中构建表达,表达获得高活性的HSA-IFNα2b融合蛋白。  相似文献

Oxidation of biopharmaceuticals is a major product quality issue with potential impacts on activity and immunogenicity. At Eli Lilly and Company, high tryptophan oxidation was observed for two biopharmaceuticals in development produced in Chinese hamster ovary cells. A switch from historical hydrolysate‐containing media to chemically defined media with a reformulated basal powder was thought to be responsible, so mitigation efforts focused on media modification. Shake flask studies identified that increasing tryptophan, copper, and manganese and decreasing cysteine concentrations were individual approaches to lower tryptophan oxidation. When amino acid and metal changes were combined, the modified formulation had a synergistic impact that led to substantially less tryptophan oxidation for both biopharmaceuticals. Similar results were achieved in shake flasks and benchtop bioreactors, demonstrating the potential to implement these modifications at manufacturing scale. The modified formulation did not negatively impact cell growth and viability, product titer, purity, charge variants, or glycan profile. A potential mechanism of action is presented for each amino acid or metal factor based on its role in oxidation chemistry. This work served not only to mitigate the tryptophan oxidation issue in two Lilly biopharmaceuticals in development, but also to increase our knowledge and appreciation for the impact of media components on product quality. © 2015 American Institute of Chemical Engineers Biotechnol. 目的:探讨多囊卵巢综合征(PCOS)患者外周血白细胞计数和血脂代谢变化的临床意义。方法:随机抽取2012年1月~2014年1月期间来院就诊未经药物治疗的PCOS患者48例作为研究对象,将体重指数(BMI)≥23的患者26例作为PCOS超重组,将BMI23的22例患者作为PCOS体重正常组,选择同期来院就诊的非PCOS妇女20例作为对照组,所有患者均于月经期空腹静脉抽血3~4m L,检测患者的血浆C反应蛋白(CRP)、血常规及血脂代谢指标。结果:PCOS超重组的甘油三酯(TG)、低密度脂蛋白(LDL-C)、载脂蛋白B(Apo-B)、外周白细胞总数、淋巴细胞、中性粒细胞、单核细胞、嗜酸性细胞、CRP指标均较对照组、PCOS体重正常组高,而高密度脂蛋白(HDL-C)、载脂蛋白A(Apo-A)水平较对照组、PCOS体重正常组低,差异均有统计学意义(P0.05);PCOS体重正常组淋巴细胞、CRP指标较对照组高,差异均有统计学意义(P0.05)。结论:慢性炎症是PCOS的主要病理表现,其中体重超重的PCOS患者主要表现为血脂代谢异常及白细胞升高,而体重正常的PCOS患者主要表现为淋巴细胞升高。  相似文献

The one‐third of plant viruses are seed transmitted, and this has significant economic consequences. Tobacco rattle virus (TRV), belonging to the genus Tobravirus and family Virgaviridae, has one of the widest host range of any known plant viruses. TRV infects vegetative organ and effects seed and pollen development that results in a decrease in crop yield. The mechanisms by which Tobravirus is transmissible to seeds are still poorly understood. The presence of the virus in pollen grains and inside ovaries is linked with seed transmission and can have effects on virus particles' transport during the pollination and fertilization process. This paper focuses on the significant impact of TRV on pepper and tobacco anthers and ultrastructure changes in ovaries. The presence of two types of TRV particles in ovary wall parenchyma and vascular tissues as well as in placenta cells was demonstrated via ultrastructural analysis. For the first time, the regular inclusion of virus particles was reported in both ovule integuments and nucellus parenchyma cells. Immunolocalization of TRV capsid proteins indicated the deposition of TRV CP epitope in ovary vascular bundles and in placenta cells. Moreover, the presence of virus particles was demonstrated inside pepper seeds in endothelium and integument parenchyma layers as well as on the embryo cell wall. Virus particles were found not only on the surface of pollen grains but also inside pepper pollen protoplasts in mature anthers. Also, this is the first time where TRV particles are reported in both differentiated endothecium cells and the remaining tapetum cells. Moreover, the detection of TRV capsid protein epitope in tobacco and pepper vascular anther tissues as well as in tapetum and endothecium cells was correlated with TRV distribution in infected anthers. Demonstrated analyses indicated that pollen grains and ovaries with ovules as well as could be a natural source of TRV transmission.  相似文献

Li)为研究对象,对植株不同部位的花序数量、花序正常发育率、每花序单花数量和果穗干质量,以及植株不同部位和花序不同部位的生物量投入比、座果率、结籽率、种子投影面积和种子千粒质量进行测定;在此基础上,对供试5种甘草属( Glycyrrhiza Linn.)植物的繁殖资源分配模式和种子生产策略进行分析。结果表明:同一植株内,光果甘草、乌拉尔甘草、黄甘草和蜜腺甘草的花序数量、花序正常发育率、每花序单花数量和果穗干质量从植株下部到上部总体上依次递减,而胀果甘草植株不同部位间这4项指标总体上无显著差异。同一植株内,胀果甘草植株中部的生物量投入比和座果率均较高,但其生物量投入比、座果率和结籽率在植株不同部位间均无显著差异;而供试另4种植物的生物量投入比、座果率和结籽率从植株下部到上部总体上依次递减。同一花序内,胀果甘草花序中部的生物量投入比明显高于花序上部和下部,座果率从花序下部到上部依次递减,结籽率则在花序不同部位间无显著差异,而供试另4种植物的生物量投入比、座果率和结籽率从花序下部到上部总体上依次递减。供试5种植物的种子投影面积和种子千粒质量在植株不同部位间和花序不同部位间均无显著差异。综合研究结果显示:在资源竞争、结构效应和花粉限制的影响下,供试5种甘草属植物存在2种不同的资源分配模式和种子生产格局。其中,光果甘草、乌拉尔甘草、黄甘草和蜜腺甘草通过减少对晚发育的花或果实的资源投入来保障早发育的花或果实获得较多的资源,达到繁殖成功的目的;而胀果甘草则采取对花和果实随机败育的方式减小资源竞争的压力,这2种繁殖资源分配模式和种子生产策略对提高甘草属植物的繁殖成功率具有重要作用。  相似文献

Standard feeds are imbalanced in term of n-6/n-3 polyunsaturated fatty acids (PUFA) ratio, with a low proportion of the latter. The reproductive system appears to be strongly affected by administration of n-3 PUFA, and ingredients rich in α-linolenic acid (ALA; i.e. vegetable sources) or EPA and DHA acids (i.e. fish oil) can be included in animal diets to balance PUFA intake. The aim of this study was to evaluate the effect of dietary supplementation with flaxseed (ALA) or fish oil (EPA and DHA) on PUFA metabolism in rabbit does. A total of 60 New Zealand White female rabbits were assigned to three experimental groups: control group, FLAX group fed 10% extruded flaxseed and FISH group fed 3% fish oil. Blood, milk, liver and ovaries were collected from the does to assess the lipid composition; furthermore, FADS2 gene expression was assessed in liver and ovary tissues. Reproductive performance of does was also recorded. The fertility rate and number of weaned rabbits improved with n-3 dietary supplementation: does at first parity showed the lowest reproductive results, but the administration of n-3 reduced the gap between primiparous and multiparous does. Feed consumption and milk production were not affected by the feeding regime. The fatty acid composition of milk, plasma, liver and ovaries were widely influenced by diet, showing higher concentrations of n-3 long-chain PUFA (LCP) in does fed with n-3 enriched diets. FISH diet resulted in the highest n-3 LCP enrichment, whereas in the FLAX group, this increase was lower. Blood and milk showed low levels of LCP, whereas liver and ovaries were the main sites of n-3 LCP synthesis and accumulation. Accordingly, although the liver is the main metabolic centre for LCP synthesis, ovaries also have a prominent role in LCP generation. FADS2 expression in liver and ovary tissue was downregulated by FISH administration. In conclusion, the enrichment of diets with n-3 PUFA could be an effective strategy for improving the reproductive performance of does.  相似文献
