Localized infection of a plant can be mapped by a sequence of images capturing chlorophyll fluorescence transients in actinic light. Choice of the actinic light protocol co-determines fluorescence contrast between infected leaf segment and surrounding healthy tissue. Frequently, biology cannot predict with which irradiance protocol, in which fluorescence image of the sequence, and in which segment of the image there will be the highest contrast between the healthy and infected tissue. Here, we introduce a new technique that can be applied to identify the combination of chlorophyll fluorescence images yielding the highest contrast. The sets of the most contrasting images vary throughout the progress of the infection. Such specific image sets, stress-revealing fluorescence signatures, can be found for the initial and late phases of the infection. Using these signatures, images can be divided into segments that show tissue in different infection phases. We demonstrate the capacity of the algorithm in an investigation of infection of the model plant Arabidopsis thaliana by the bacterium Pseudomonas syringae. We show that the highest contrast is found with transients elicited by fluctuating, harmonically modulated irradiance with long periods.  相似文献

The monsoon affected mountains of the southern Arabian Peninsula harbour in climatically favoured refugia vegetation elements of palaeo-African origin. To understand better the temporal and spatial differentiation of these refugia, chloroplast variation in Justicia areysiana Deflers (Acanthaceae), a shrub species endemic to the Yemeni and Omani mountains close to the Arabian Sea, was studied using PCR-RFLP and chloroplast microsatellite diversity. Eleven haplotypes were characterized and show a distinct geographical distribution pattern with a deep split between populations from south Yemeni fog oases and those from Hawf Mountains/Dhofar region in east Yemen and south Oman. Very limited haplotype diversity within populations (hS = 0.15) and a high level of population differentiation (GST = 0.81) demonstrate the strong genetic isolation of populations from each other. Past oscillations between humid and arid periods connected with glacial and interglacial episodes in the Pleistocene and Holocene are considered responsible for the observed patterns of genetic variation.  © 2005 The Linnean Society of London, Botanical Journal of the Linnean Society , 2005, 148 , 437–444.  相似文献

The present paper concerns with ion homeostatic reactions in view of stimulus-secretion coupling of the beta-cell, including Ca2+ fluxes of the endoplasmatic reticulum (ER). A steady state of cytosolic sodium and potassium ion concentrations ([Na+]c and [K+]c, respectively), and of the membrane potential (Delta c phi) can be attained only, if the flux through the electrogenic Na-K pump (JNaK) is balanced electrically, and if JNaK is rather high (about 25% of total ATP consumption at 10 mM glucose). Metabolically caused changes of cellular pH are unlikely, because, on the one hand, CO2 can rapidly leave the cell through cellular membranes, and because ATP cycling cannot produce nor consume protons. A slight decrease of pHc during cellular activity is caused mainly by an increased Ca-H exchange flux through the plasma membrane Ca2+ pump (J PMCA), which might be overcome, however, by H+ transport into secretory granules. The present simulations show that the conductance of ATP-sensitive K+ channels (K ATP) is highly susceptible to changes of [Mg2+]c. As a physical link between the Ca2+ filling state of the ER and the initiation of a depolarising, Ca2+ release-activated current (I CRAN), a metabolite (inositol 1,4,-diphosphate (IP2)) of the inositol 1,4,5-triphosphate (IP3) cycle is introduced. Sufficient ATP for insulin secretion is made available during glucose activation by [IP2] inhibition of a parallel [ATP]c consuming flux through protein biosynthesis (J Pbs). This leads to fast oscillations with a triphasic patterns of [Ca2+]c oscillations. Slow oscillations are initiated by including a Ca2+ leak current through highly uncoupled SERCA3 pumps. Both types of oscillations may superimpose yielding compound bursting and mixed oscillations of [Ca2+]c.  相似文献

In contradiction with field observations, theory predicts that the number of coexisting plankton species at equilibrium cannot exceed the number of limiting resources, which is called the "paradox of the plankton". Recently, Huisman & Weissing (1999 , 2000 ) showed, in a model study, that the number of coexisting species may exceed the number of limiting resources when internal system feedback induces oscillations or chaos. In this paper, we use the term "supersaturated coexistence" for this phenomenon. On the basis of these findings, they claimed that the paradox of the plankton is solved. We investigated the prerequisites for supersaturated coexistence in the same model. Our results indicate that supersaturated coexistence is a rare phenomenon in parameter space, requires a very precise parameterization of the community members and is sensitive to the introduction of new species and the removal of the present species. This raises the question of whether supersaturated coexistence is likely to occur in nature. We conclude that the claim by Huisman & Weissing (1999 , 2000 ) is premature.  相似文献

We treat synchronization of metabolic oscillations in two cellsand in ensembles of cells adsorbed at the liquid-solid interface.(i) Synchronization of oscillations in two cells is assumedto occur via perturbation of the metabolite concentration nearone cell due to the metabolite diffusion flux from another cell.This direct channel of synchronization may be important ifthe distance between two cells is comparable with the cell diameter.The corresponding coupling coefficient is found to be proportionalto the metabolite diffusion coefficient and inversely proportionalto the cell radius and the distance between the cells.(ii) In the case of ensembles of adsorbed cells, synchronizationof oscillations is considered to be indirect, i.e., to occur viathe metabolite concentration formed outside the cells nearthe interface due to metabolite diffusion from the cells. 目的和方法 :4 0 0~ 5 0 0 μm大鼠水平脑切片含有封闭的EC 海马环路。强直电刺激 (60Hz ,2s)海马Schaeffer侧支诱发癫痫放电 ,全细胞记录CA1胞体层单个神经元电活动 ,同步记录相应树突区细胞外场电位 ,探讨单个神经元膜电位振荡特性与细胞外癫痫电活动之间的关系。结果 :①强直电刺激诱发CA1神经元膜电位后放性振荡行为呈宽频特征 (3~ 10 0Hz)。以θ节律多见 ,跟随在刺激引起的膜电位去极化或超极化偏移 (paroxysmaldepolarizingorhyperpolaringshift,PDSorPHS)之后 ,振荡波的上升支和下降支分别由膜电位去极化 超极化或超极化 去极化成分构成 ;②逐渐增强的IPSP构成了膜电位振荡的起搏成分 ,继而反弹形成锋电位和阈下振荡 ,与细胞外癫痫样电活动同步 ,并促成癫痫放电由紧张性向阵挛性形式转变 ;③发现了电偶联电位 (spikelets)以及细胞之间的染料偶联现象。结论 :单个神经元作为振荡器可以启动群体神经元超同步化癫痫样电活动 ;缝隙连接可能参与了膜电位振荡的启动与场电位癫痫样电活动的同步作用。  相似文献

Using a generalSaccharomyces cerevisiae as a model strain, continuous ethanol fermentation was carried out in a stirred tank bioreactor with a working volume of 1,500 mL. Three different gravity media containing glucose of 120, 200 and 280 g/L, respectively, supplemented with 5 g/L yeast extract and 3 g/L peptone, were fed into the fermentor at different dilution rates. Although complete steady states developed for low gravity medium containing 120 g/L glucose, quasi-steady states and oscillations of the fermented parameters, including residual glucose, ethanol and biomass were observed when high gravity medium containing 200 g/L glucose and very high gravity medium containing 280 g/L glucose were fed at the designated dilution rate of 0.027 h−1. The observed quasi-steady states that incorporated these steady states, quasi-steady states and oscillations were proposed as these oscillations were of relatively short periods of time and their averages fluctuated up and down almost symmetrically. The continuous kinetic models that combined both the substrate and product inhibitions were developed and correlated for these observed quasi-steady states.  相似文献

多年生灌木沙棘和中间锦鸡儿是黄土高原生态重建的重要物种,设计人工模拟水分胁迫实验,测量沙棘和中间锦鸡儿蒸腾作用的各种指标,研究其蒸腾特性对水分胁迫的适应方式。结果表明,同等水分处理条件下,中间锦鸡儿单叶水平上的蒸腾速率高于沙棘。沙棘和中间锦鸡儿的蒸腾速率日进程在晴天、阴雨天和生长发育的不同阶段明显不同。夜间蒸腾占全天蒸腾的比例相当大,夜间蒸腾在不同物种之间、不同供水量之间存在明显差异,而且越干旱的环境比例越大。两种植物气孔阻力的季节变化格局在不同水分处理间大体相似。沙棘的昼夜蒸腾节律在各种水分处理条件下都表现出明显的气孔振荡现象,而中间锦鸡儿没有。叶片温度、光合有效辐射和气孔阻力是各种水分条件下沙棘和中间锦鸡儿蒸腾作用的共同的限制因子,相对于沙棘,中间锦鸡儿还更多地受到空气相对湿度的影响。  相似文献

恐惧作为个体应对内外界危险因素形成的自我保护机制的一部分,在生物体的生存中发挥着重要作用.但过度的恐惧不仅对个体生存无益,反而易引发创伤后应激障碍、焦虑等精神疾病,严重影响个体生活质量.临床上通常采用基于行为学研究结果的暴露疗法对恐惧相关疾病进行治疗,然而在患者处于治疗环境之外的时候,上述症状经常会复发.因此,解析恐惧记忆相关神经环路内信息处理的神经机制,对于理解这些疾病的发生发展,寻求切实有效的治疗方案至关重要.大量研究表明与恐惧记忆消退相关的脑区主要涉及杏仁核、内侧前额叶和海马.在恐惧消退的过程中,这3个脑区表现出特定的神经振荡模式,而且这些活动也具有同步性,构成了恐惧记忆成功消退的神经基础.未来可利用基于神经神经振荡的无创性脑刺激手段干预恐惧记忆消退的神经环路,以促进恐惧记忆的消退并避免复发,为恐惧相关障碍的临床治疗提供重要的科学依据.  相似文献

Increases in cytosolic free Ca2+ ([Ca2+]cyt) are common to many stress-activated signalling pathways, including the response to saline environments. We have investigated the nature of NaCl-induced [Ca2+]cyt signals in whole Arabidopsis thaliana seedlings using aequorin. We found that NaCl-induced increases in [Ca2+]cyt are heterogeneous and mainly restricted to the root. Both the concentration of NaCl and the composition of the solution bathing the root have profound effects on the magnitude and dynamics of NaCl-induced increases in [Ca2+]cyt. Alteration of external K+ concentration caused changes in the temporal and spatial pattern of [Ca2+]cyt increase, providing evidence for Na+-induced Ca2+ influx across the plasma membrane. The effects of various pharmacological agents on NaCl-induced increases in [Ca2+]cyt indicate that NaCl may induce influx of Ca2+ through both plasma membrane and intracellular Ca2+-permeable channels. Analysis of spatiotemporal [Ca2+]cyt dynamics using photon-counting imaging revealed additional levels of complexity in the [Ca2+]cyt signal that may reflect the oscillatory nature of NaCl-induced changes in single cells.  相似文献
