Hydrologic pathways through soil affect element leaching by determining the relative importance of biogeochemical processes such as sorption and decomposition. We used stable hydrogen isotopes of water (δD) to examine the influence of flowpaths on soil solution chemistry in a mature spruce–hemlock forest in coastal Oregon, USA. Soil solutions (50 cm depth, n = 13) were collected monthly for 1 year and analyzed for δD, major ions and dissolved organic carbon (DOC) and nitrogen (DON). We propose that the variability of δD can be used as an index of flowpath length and contact time. Throughfall variability in δD was much greater than soil solution variability, illustrating that soil solution integrates the variation in inputs. Lysimeters with greater variation in δD presumably have a greater proportion of flow through rapid flowpaths such as macropores. The variation in soil solution δD for individual lysimeters explained up to 53% of the variation in soil solution chemistry, and suggests that flowpaths influence leaching of some constituents. 2 = 0.51 and 0.37, respectively), perhaps because transport via macropores reduces interaction of DOM with the soil matrix. In contrast, nitrate concentrations were highest in lysimeters with a small variation in δD, where long contact time and low DOC concentrations may yield higher net nitrification. Our results demonstrate the utility of stable isotopes to link flowpaths and soil solution chemistry, and illustrate how the spatial complexity of soils can influence ecosystem-level nutrient losses. 相似文献

Summary Plant yield of mycorrhizal and non-mycorrhizalFestuca rubra L. was linearly decreased with increasing zinc concentrations in nutrient solution. In all cases, non-mycorrhizal plant growth was significantly greater than that of mycorrhizal plants. Zinc and phosphorus concentrations of root and shoot of mycorrhizal plants were greater in all zinc treatments while mycorrhizal plants showed equal or lower tolerance indices to zinc than non-mycorrhizal plants. A solvent system that extracts a maximum number of metabolites belonging to diverse chemical classes from complex biofluids, such as plasma, may offer useful inputs to understand the metabolic and physiological state of an individual. The present study compared seven solvent systems for extraction of metabolites from plasma. The extracts were analyzed by mass spectrometry (MS) and MS/MS (MS2) using a quadrupole time-of-flight liquid chromatography/MS system in positive and negative modes of ionization. Metabolites with molecular mass below 400 were identified using Human Metabolome Database MS2 and MS search interfaces. The acetone/isopropanol (2:1) system yielded promising results in positive ionization mode, as the maximum number of MS and MS2 features was detected in the extract. It was found to be superior in extraction of various classes of metabolites, especially organic acids, nucleosides and nucleoside derivatives, and heterocyclic molecules. Glycerophosphocholines in the mass range of 400–700 were found to be efficiently extracted by the methanol/chloroform/water (8:1:1) system. In negative mode as well, the maximum number of MS2 features was detected in methanol/chloroform/water and acetone/isopropanol extracts. The consumption of in-situ chemical oxidation (ISCO) oxidant by soil oxidizable matter (OM), termed the soil oxidant demand (SOD), is an essential factor when designing treatments for successful remediation at an ISCO site. This study aims to assess the impact of different oxidants on SOD and the soil itself, using the Taguchi experimental design. Five oxidation systems, including persulfate (PS), hydrogen peroxide (HP), permanganate (PM), Fe2+ activated PS and Fe2+ activated HP, and four factors including oxidant concentration, activator concentration, reaction time, and pH were investigated. The results of the Taguchi analysis in this study show that oxidant concentration had the greatest effect on the SOD. Other factors also affected the SOD and the optimum conditions for achieving a lower SOD were determined using the Taguchi design method. Additionally, original and oxidized soils were analyzed using a scanning electron microscope equipped with an X-ray energy dispersive spectrometer to determine the surface morphology and chemical composition of the samples. Antifreeze proteins (AFPs) adsorb to ice crystals and inhibit their growth, leading to non-colligative freezing point depression. Crops like spring wheat, that are highly susceptible to frost damage, can potentially be made frost tolerant by expressing AFPs in the cytoplasm and apoplast where ice recrystallisation leads to cellular damage. The protein sequence for HPLC-6 α-helical antifreeze protein from winter flounder was rationally redesigned after removing the prosequences in the native protein. Wheat nuclear gene preferred amino acid codons were used to synthesize a recombinant antifreeze gene, rAFPI. Antifreeze protein was targeted to the apoplast using a Murine leader peptide sequence from the mAb24 light chain or retained in the endoplasmic reticulum using C-terminus KDEL sequence. The coding sequences were placed downstream of the rice Actin promoter and Actin-1 intron and upstream of the nopaline synthase terminator in the plant expression vectors. Transgenic wheat lines were generated through micro projectile bombardment of immature embryos of spring wheat cultivar Seri 82. Levels of antifreeze protein in the transgenic lines without any targeting peptide were low (0.06–0.07%). The apoplast-targeted protein reached a level of 1.61% of total soluble protein, 90% of which was present in the apoplast. ER-retained protein accumulated in the cells at levels up to 0.65% of total soluble proteins. Transgenic wheat line T-8 with apoplast-targeted antifreeze protein exhibited the highest levels of antifreeze activity and provided significant freezing protection even at temperatures as low as −7°C. 相似文献

Different methods for estimating the non-fertilizer N supply (NFNS) of mineral grassland soils were compared. NFNS was defined as the N uptake on unfertilized plots. The potential mineralization rate (0–12 weeks), macroorganic matter and active microbial biomass (determined by the substrate-induced respiration method; SIR) were correlated positively with NFNS. The difference between the actual soil organic N or microbial N content (determined by the fumigation incubation method) and their contents under equilibrium conditions ( org. N and MB-N), however, gave the best estimations of NFNS. For field conditions the best estimation for NFNS was: NFNS (kg N ha–1 yr–1)=132.3+42.1× org. N (g kg–1 soil; r=0.80). This method is based on the observation that, under old grassland swards, close relationships exist between soil texture and the amounts of soil organic N and microbial N. These relationships are assumed to represent equilibrium conditions as under old swards under constant management, the gain in soil organic N and microbial N equals the losses. Soils under young grassland and recently reclaimed soils contained less soil organic N and microbial N. In such soils the amounts of organic N and microbial N increase with time, which is reflected in a lower NFNS. The annual accumulation of organic and microbial N gradually becomes smaller until organic N, microbial N and NFNS reach equilibrium. Biochemical analysis of enantioselective short-chain alcohol dehydrogenase from the hyperthermophilic archaeon Thermococcus sibiricus (TsAdh319) revealed unique polyextremophilic properties of the enzyme – half-life of 1 h at 100 °C, tolerance to high salt (up to 4 M) and organic solvents (50% v/v) concentrations. To elucidate the molecular basis of TsAdh319 polyextremophilicity, we determined the crystal structure of the enzyme in a binary complex with 5-hydroxy-NADP at 1.68 Å resolution. TsAdh319 has a tetrameric structure both in the crystals and in solution with an intersubunit disulfide bond. The substrate-binding pocket is hydrophobic, spacious and open that is consistent with the observed promiscuity in substrate specificity of TsAdh319. The present study revealed an extraordinary number of charged residues on the surface of TsAdh319, 70% of which were involved in ion pair interactions. Further we compared the structure of TsAdh319 with the structures of other homologous short-chain dehydrogenases/reductases (SDRs) from thermophilic and mesophilic organisms. We found that TsAdh319 has the highest arginine and aspartate + glutamate contents compared to the counterparts. The frequency of occurrence of salt bridges on the surface of TsAdh319 is the highest among the SDRs under consideration. No differences in the proline, tryptophan, and phenylalanine contents are observed; the compactness of the protein core of TsAdh319, the monomer and tetramer organization do not differ from that of the counterparts. We suggest that the unique thermostability of TsAdh319 is associated with the rigidity and simultaneous “resilience” of the structure provided by a compact hydrophobic core and a large number of surface ion pairs. To understand the ecology and environmental tolerances of newly hatched larvae of the amphidromous fish Sicyopterus japonicus during their downstream migration, the salinity tolerance of eggs, 0-15 day old larvae, and adults, and the temperature tolerance, specific gravity and phototaxis of hatched larvae were examined. Tolerances of adults were measured as survival after a 24 h challenge in freshwater (FW), brackish water (1/3 SW) and seawater (SW). The survival rate of adult S. japonicus was 100% in FW and 1/3 SW, while none survived in SW. Hatching success of eggs (30 eggs each) was significantly higher in FW (mean: 73%) and 1/3 SW (73%) than in SW (19%). Tolerance of newly hatched larvae to salinity and temperature was investigated in different combinations of salinities (FW, 1/3 SW and SW) and temperatures (18, 23 and 28 °C). Larval survival was significantly different in each salinity and temperature. Survival rate was significantly higher in 1/3 SW than in FW and higher in SW than in FW at 23 °C and 28 °C. At the latter part of the experiment, there was no survival in FW and at 28 °C. Survival was higher in lower temperatures, but larval development did not occur in FW. Specific gravity of newly hatched larvae was 1.036 at 28 °C and 1.034 at 23 °C. When exposed to a light source on one side of an aquarium, larval distribution was not affected. Our results indicated larval S. japonicus are more adapted to brackish water and seawater than freshwater, while the adults and eggs are more adapted to freshwater and brackish water than seawater. Ground beetle activity and species richness was monitored using pitfall traps in a plot trial system on a farm in northern England where the effects of organic and conventional fertility and crop protection management were separated within different crop types between 2005 and 2008. As well as analyses on species activity the beetles were split into small, medium‐sized and large groups, and into groups of herbivores and specific Collembola feeders. Crop type had significant effects on the activity of the 20 most abundant species and all groups, generally with most in beans and winter barley and least in vegetables and spring barley. Most significant reactions to crop protection and fertility management were in cereals and grass/clover. Activity of small species was highest in conventionally crop‐protected cereals but not in vegetables, with more medium‐sized and herbivorous species in organic plots, but there was little influence of crop protection management on large and Collembola feeding species. However, large species were significantly more active in organically fertilised cereals and grass/clover, but not in vegetables, and there were more Collembola feeders in conventional cereals but not in grass/clover. Small species were more abundant in conventionally fertilised grass/clover but there were more in organic cereals and vegetables. These inconsistent activity reactions to management were also observed with individual species but most preferred organically managed plots. There were few significant crop protection : fertility management interactions. Species richness was also significantly affected by crop type and where management had an influence, more species were found in organically managed plots. Constrained ordination emphasised that ground beetle activity was influenced more by crops than by management. Given the diverse nature of organic crop rotations, crop type should be considered a major influence in any environmental manipulation aimed at increasing ground beetle activity for provision of ecosystem services. 相似文献
