CdTe quantum dots, 3.28 nm in size, were synthesized using a one‐step method in an aqueous medium. The CdTe quantum dots were successfully employed as hybrid phosphors for white light‐emitting diode (LED) devices by combining them with yellow‐emitting YAG:Ce phosphor. The color‐rendering index value and International Commission on illumination coordinates for hybrid phosphor white LEDs were 75 and (x = 0.30, y = 0.29), respectively. Compared with conventional phosphors, semiconductor quantum dots have larger band gap energy and broader absorption features, and can be excited more efficiently by optical pumping sources. The results confirmed that the high color‐rendering index value of the white LED was due to the CdTe quantum dots introduced in the hybrid phosphor system. ZnS nanoparticles stabilized by a carbohydrate‐based matrix, hypromellose (hydroxypropyl methylcellulose) were prepared via a wet chemical method. The nanocomposite was characterized by X‐ray diffraction, transmission electon microscopy and Fourier transform infrared spectroscopy. X‐Ray diffraction patterns revealed a zinc blende structure. Thermogravimetric analysis suggested that polymer attached to the surface decomposes at 700 °C. Absorption measurements were carried out and calculation of the diameter polydispersity index (DPI) suggests the formation of monodisperse nanoparticles. The optical properties of the as‐prepared samples were studied by UV/vis spectroscopy and steady‐state photoluminescence (PL) spectroscopy. The PL studies indicate the applicability of these nanoparticles as biocompatible sensors or luminescence markers in future. Copyright © 2013 John Wiley & Sons, Ltd.  相似文献

Cu(II) ion‐doped NaCaAlPO4F3 phosphor has been synthesized using a solid state reaction method. The prepared sample is characterized by powder X‐ray diffraction, scanning electron microscope, optical absorption, electron paramagnetic resonance photoluminescence and Fourier transform infrared spectroscopy techniques. The crystallite size evaluated from x‐ray diffraction data is in nanometers. Scanning electron microscopy micrographs showed the presence of several irregular shaped particles. From optical absorption and electron paramagnetic resonance spectral data the doped Cu(II) ions are ascribed to distorted octahedral site symmetry. The synthesized phosphor exhibits emission bands in ultraviolet, blue and green regions under the excitation wavelength of 335 nm. The CIE chromaticity coordinates (x = 0.159, y = 0.204) also calculated for the prepared sample from the emission spectrum. Barium‐gadolinium‐titanate (BaGd2Ti4O12) powder ceramics doped with rare‐earth ions (Eu3+ and Tb3+) were synthesized by a solid‐state reaction method. From the X‐ray diffraction spectrum, it was observed that Eu3+ and Tb3+:BaGd2Ti4O12 powder ceramics are crystallized in the form of an orthorhombic structure. Scanning electron microscopy image shows that the particles are agglomerated and the particle size is about 200 nm. Eu3+‐ and Tb3+‐doped BaGd2Ti4O12 powder ceramics were examined by energy dispersive X‐ray analysis, Fourier transform infrared spectroscopy, photoluminescence and thermoluminescence (TL) spectra. Emission spectra of Eu3+‐doped BaGd2Ti4O12 powder ceramics showed bright red emission at 613 nm (5D07F2) with an excitation wavelength λexci = 408 nm (7F05D3) and Tb3+:BaGd2Ti4O12 ceramic powder has shown green emission at 534 nm (5D47F5) with an excitation wavelength λexci = 331 nm ((7F65D1). Multimodality imaging has emerged as a common technological approach used in both preclinical and clinical research. Advanced techniques that combine in vivo optical and μCT imaging allow the visualization of biological phenomena in an anatomical context. These imaging modalities may be especially useful to study conditions that impact bone. In particular, orthopaedic implant infections are an important problem in clinical orthopaedic surgery. These infections are difficult to treat because bacterial biofilms form on the foreign surgically implanted materials, leading to persistent inflammation, osteomyelitis and eventual osteolysis of the bone surrounding the implant, which ultimately results in implant loosening and failure. Here, a mouse model of an infected orthopaedic prosthetic implant was used that involved the surgical placement of a Kirschner-wire implant into an intramedullary canal in the femur in such a way that the end of the implant extended into the knee joint. In this model, LysEGFP mice, a mouse strain that has EGFP-fluorescent neutrophils, were employed in conjunction with a bioluminescent Staphylococcus aureus strain, which naturally emits light. The bacteria were inoculated into the knee joints of the mice prior to closing the surgical site. In vivo bioluminescent and fluorescent imaging was used to quantify the bacterial burden and neutrophil inflammatory response, respectively. In addition, μCT imaging was performed on the same mice so that the 3D location of the bioluminescent and fluorescent optical signals could be co-registered with the anatomical μCT images. To quantify the changes in the bone over time, the outer bone volume of the distal femurs were measured at specific time points using a semi-automated contour based segmentation process. Taken together, the combination of in vivo bioluminescent/fluorescent imaging with μCT imaging may be especially useful for the noninvasive monitoring of the infection, inflammatory response and anatomical changes in bone over time.  相似文献

We describe a multi-angle rotational optical imaging (MAROI) system for in vivo monitoring of physiopathological processes labeled with a fluorescent marker. Mouse models (brain tumor and arthritis) were used to evaluate the usefulness of this method. Saposin C (SapC)-dioleoylphosphatidylserine (DOPS) nanovesicles tagged with CellVue Maroon (CVM) fluorophore were administered intravenously. Animals were then placed in the rotational holder (MARS) of the in vivo imaging system. Images were acquired in 10° steps over 380°. A rectangular region of interest (ROI) was placed across the full image width at the model disease site. Within the ROI, and for every image, mean fluorescence intensity was computed after background subtraction. In the mouse models studied, the labeled nanovesicles were taken up in both the orthotopic and transgenic brain tumors, and in the arthritic sites (toes and ankles). Curve analysis of the multi angle image ROIs determined the angle with the highest signal. Thus, the optimal angle for imaging each disease site was characterized. The MAROI method applied to imaging of fluorescent compounds is a noninvasive, economical, and precise tool for in vivo quantitative analysis of the disease states in the described mouse models.  相似文献

Tissue clearing and subsequent imaging of transparent organs is a powerful method to analyze fluorescently labeled cells and molecules in 3D, in intact organs. Unlike traditional histological methods, where the tissue of interest is sectioned for fluorescent imaging, 3D imaging of cleared tissue allows examination of labeled cells and molecules in the entire specimen. To this end, optically opaque tissues should be rendered transparent by matching the refractory indices throughout the tissue. Subsequently, the tissue can be imaged at once using laser-scanning microscopes to obtain a complete high-resolution 3D image of the specimen. A growing list of tissue clearing protocols including 3DISCO, CLARITY, Sca/e, ClearT2, and SeeDB provide new ways for researchers to image their tissue of interest as a whole. Among them, 3DISCO is a highly reproducible and straightforward method, which can clear different types of tissues and can be utilized with various microscopy techniques. This protocol describes this straightforward procedure and presents its various applications. It also discusses the limitations and possible difficulties and how to overcome them.  相似文献

Disposable cell culture vessels are extensively used at small scales for process optimization and validation, but they lack monitoring capabilities. Optical sensors that can be easily adapted for use in small‐scale vessels are commercially available for pH, dissolved oxygen (DO), and dissolved carbon dioxide (DCO2). However, their use has been limited due to the contamination and compatibility issues. We have developed a novel solution to these problems for DO monitoring. Oxygen diffusion through permeable vessel wall can be exploited for noninvasive monitoring. An optical oxygen sensor can be placed outside the oxygen permeable vessel wall thereby allowing oxygen diffusing through the vessel wall to be detected by the sensor. This way the sensor stays separate from the cell culture and there are no concerns about contaminants or leachants. Here we implement this method for two cell culture devices: polystyrene‐made T‐75 tissue culture flask and fluorinated ethylene propylene (FEP)‐made Vuelife® cell culture bag. Additionally, mammalian and microbial cell cultures were performed in Vuelife® cell culture bags, proving that a sensor placed outside can be used to track changes in cell cultures. This approach toward noninvasive monitoring will help in integrating cell culture vessels with sensors in a seamless manner. © 2013 American Institute of Chemical Engineers Biotechnol.
