Addition of ATP to chloroplasts causes a reversible 25–30% decrease in chlorophyll fluorescence. This quenching is light-dependent, uncoupler insensitive but inhibited by DCMU and electron acceptors and has a half-time of 3 minutes. Electron donors to Photosystem I can not overcome the inhibitory effect of DCMU, suggesting that light activation depends on the reduced state of plastoquinone. Fluorescence emission spectra recorded at ?196°C indicate that ATP treatment increases the amount of excitation energy transferred to Photosystem I. Examination of fluorescence induction curves indicate that ATP treatment decreases both the initial (Fo) and variable (Fv) fluorescence such that the ratio of Fv to the maximum (Fm) yield is unchanged. The initial sigmoidal phase of induction is slowed down by ATP treatment and is quenched 3-fold more than the exponential slow phase, the rate of which is unchanged. A plot of Fv against area above the induction curve was identical plus or minus ATP. Thus ATP treatment can alter quantal distribution between Photosystems II and I without altering Photosystem II-Photosystem II interaction. The effect of ATP strongly resembles in its properties the phosphorylation of the light-harvesting complex by a light activated, ATP-dependent protein kinase found in chloroplast membranes and could be the basis of physiological mechanisms which contribute to slow fluorescence quenching in vivo and regulate excitation energy distribution between Photosystem I and II. It is suggested that the sensor for this regulation is the redox state of plastoquinone.  相似文献

Because of its physical properties, apolipoprotein B (apo B) has remained poorly characterized. In an attempt to elucidate apo B structure, the Fab fragments of 3 different monoclonal anti-human apo B antibodies were tested in a quantitative assay for their binding to human low density lipoprotein (LDL). In each case the assay gave a linear Scatchard plot with a maximum of 1 Fab fragment bound to a single LDL particle. This result favors an LDL model containing a single large Mr apo B protein, which is not composed of multiple, identical, small Mr subunits.  相似文献

  1. Most of the world's large rivers are dammed for the purposes of water storage, flood control, and power production. Damming rivers fundamentally alters water temperature and flows in tailwater ecosystems, which in turn affects the presence and abundance of downstream biota.
  2. We collaborated with more than 200 citizen scientists to collect 2,194 light trap samples across 2 years and more than 2,000 river km. Samples contained 16,222 net‐spinning caddisfly (Hydropsyche) individuals across six species. We used these data to model the distribution of Hydropsyche throughout the Colorado River Basin in the western U.S.A. to identify the roles of water temperature, flows, and species‐specific morphology in determining aquatic species distributions throughout a large arid watershed that has been heavily altered by damming.
  3. We predicted that water temperatures would determine Hydropsyche presence and abundance to a greater extent than diel variation in river stage associated with hydropower production. Among many species, adult female Hydropsychids are morphologically adapted to swim to deep‐water oviposition sites. We predicted that the presence of this ability would negate the otherwise deleterious effects of high stage change on caddisfly egg mortality.
  4. We found that distributions of the two most widespread species, Hydropsyche occidentalis and Hydropsyche oslari (92% of total Hydropsyche captured), were both predicted by water temperatures. However, we also found that the abundance of H. oslari decreased by as much as 10‐fold as diel stage change increased, despite the presence of female morphological adaptations for deep‐water oviposition. We found sexual dimorphism and evidence for deep‐water swimming adaptations in 5/6 species.
  5. Our results show that net‐spinning caddisflies have species‐specific responses to environmental variation and suggest that environmental flows designed to reduce diel stage change and destabilise water temperatures may improve habitat quality for these ubiquitous and important aquatic insects.

“Beyond GDP” initiatives flag the limits of the quantitative indicators of progress currently used for governance. Focusing on the quality assessment of quantitative information used for governance, we use some of the conceptual tools of theoretical ecology and evolutionary biology in order to identify the pre-analytical choices that determine the usefulness and pertinence of a model. Starting from the definition of a model as a formal representation of a specific and necessarily subjective observation, we show that the production of indicators is the final result of a series of decisions on what to observe and how. These choices, in turn, depend on the narrative, or set of narratives, adopted. Narratives provide causality and context to knowledge claims and are needed to select the indicators to be used for policy. Moving beyond the GDP debate requires reflexivity, that is, awareness of the key role that pre-analytical choices play in the definition of both the relevance of the chosen perceptions and narratives (determined by the normative stands of different actors – who defines wellbeing?), and the usefulness of the chosen models and data (determined by the pertinence of the resulting representation – how to measure wellbeing?). Reflexivity is essential in order to take into account the purposes for which different indicators were created and to define new purposes for the “beyond GDP” indicators.  相似文献

Aim To test predictions of the vicariance model, to define basic biogeographical units for Cerrado squamates, and to discuss previous biogeographical hypotheses. Location Cerrado; South American savannas south of the Amazon, extending across central Brazil, with marginal areas in Bolivia and Paraguay and isolated relictual enclaves in adjacent regions. Methods We compiled species occurrence records via field sampling and revision of museum specimens and taxonomic literature. All species were mapped according to georeferenced locality records, and classified as (1) endemic or non‐endemic, (2) typical of plateaus or depressions, and (3) typical of open or forested habitats. We tested predictions of the vicariance model using biotic element analysis, searching for non‐random clusters of species ranges. Spatial congruence of biotic elements was compared with putative areas of endemism revealed by sympatric restricted‐range species. Effects of topographical and vegetational mosaics on distribution patterns were studied according to species composition in biotic elements and areas of endemism. Results We recorded 267 Cerrado squamates, of which 103 (39%) are endemics, including 20 amphisbaenians (61% endemism), 32 lizards (42%) and 51 snakes (32%). Distribution patterns corroborated predictions of the vicariance model, revealing groups of species with significantly clustered ranges. An analysis of endemic species recovered seven biotic elements, corroborating results including non‐endemics. Sympatric restricted‐range taxa delimited 10 putative areas of endemism, largely coincident with core areas of biotic elements detected with endemic taxa. Distribution patterns were associated with major topographical and vegetational divisions of the Cerrado. Endemism prevailed in open, elevated plateaus, whereas faunal interchange, mostly associated with forest habitats, was more common in peripheral depressions. Main conclusions Our results indicate that vicariant speciation has strongly shaped Cerrado squamate diversity, in contrast to earlier studies emphasizing faunal interchange and low endemism in the Cerrado vertebrate fauna. Levels of squamate endemism are higher than in any other Cerrado vertebrate group. The high number of recovered endemics revealed previously undetected areas of evolutionary relevance, indicating that biogeographical patterns in the Cerrado were poorly represented in previous analyses. Although still largely undocumented, effects of vicariant speciation may be prevalent in a large fraction of Cerrado and Neotropical biodiversity.  相似文献

Aim This study investigates changes in bird communities between 1998 and 2008 in four savanna sites in Swaziland and the extent to which shrub encroachment is responsible for these changes. Location Swaziland, southern Africa. Methods Generalized estimated equations were used to estimate changes in bird species occurrence between 1998 and 2008. Remote sensing of aerial photographs/satellite images was used to assess vegetation changes during the same period. We assessed the role of shrub encroachment for bird communities by testing the relationship between change in species occurrence and species habitat using a general linear model. We also estimated species richness, colonization and extinction and used general linear models to test the effects of vegetation changes on these parameters. Results More than half of the bird species showed a significant change in occurrence between 1998 and 2008: 32 species increased and 29 decreased. Change in species occurrence was significantly explained by species habitat. Species significantly increasing were mainly associated with wooded savanna, whereas species significantly decreasing were mainly associated with open savanna. Species richness decreased significantly, and this decrease was significantly explained by shrub cover increase at the plot scale (from 24% to 44% on average). Extinction at the plot scale was significantly influenced by the loss of grass cover, while colonization at the plot scale was influenced by tree cover increase. Main conclusions This study represents the first evidence of temporal changes in bird communities owing to shrub encroachment in southern Africa. Despite its short time frame (10 years), this study shows dramatic changes in both vegetation structure and bird community composition. This confirms the general concern for southern African bird species associated with open savanna if current trends continue.  相似文献

Considering the significant evidential values of fingerprints in underwater criminal investigations and the need to visualise them using a user- and environmentally-friendly reagent, development of a novel, rapid and relatively greener nanobio-based reagent (NBR) is deemed beneficial. Lipase from the commercial Candida rugosa immobilised onto acid-functionalised multi-walled carbon nanotubes (NBR) was used as the safer and cheap lipid-sensing reagent to visualise groomed whole/split fingerprints on non-porous objects immersed in stagnant tap water for up to 30 days under a laboratory-controlled setting. Attenuated Total Reflectance – Fourier Transform Spectrometry, Field Emission Scanning Electron Microscopy and bioinformatics (molecular docking and molecular dynamics simulations) were employed to characterise and confirm the attachment of NBR onto the lipid constituents of wet fingerprints. Chromatographic results further confirmed the presence of n-hexadecanoic and octadecanoic acids on fingerprints up to 30 days of immersion. Thus, NBR may potentially be useful as the future state-of-the-art fingerprint visualisation technology.  相似文献

With reference to specific case studies in China, a new life, consisting of donated organs from different bodies by donation, can be called “donated life.” Donated life is a win-win action between altruism and egoism, that is, to save the lives of others and to regenerate the organs of donors or their relatives. Due to the emergence of this kind of life, traditional social ethics theories based on the marriage-related family find it difficult to difficult to explain the new realities. Primary open‐angle glaucoma is a leading cause of irreversible blindness, often associated with increased intraocular pressure. Extracellular vesicles (EVs) carry a specific composition of proteins, lipids and nucleotides have been considered as essential mediators of cell‐cell communication. Their potential impact for crosstalk between tissues responsible for aqueous humour production and out‐flow is largely unknown. The study objective was to investigate the effects of EVs derived from non‐pigmented ciliary epithelium (NPCE) primary cells on the expression of Wnt proteins in a human primary trabecular meshwork (TM) cells and define the mechanism underlying exosome‐mediated regulation that signalling pathway. Consistent with the results in TM cell line, EVs released by both primary NPCE cells and NPCE cell line showed diminished pGSK3β phosphorylation and decreased cytosolic levels of β‐catenin in primary TM cells. At the molecular level, we showed that NPCE exosome treatment downregulated the expression of positive GSKβ regulator‐AKT protein but increased the levels of GSKβ negative regulator‐PP2A protein in TM cells. NPCE exosome protein analysis revealed 584 miRNAs and 182 proteins involved in the regulation of TM cellular processes, including WNT/β‐catenin signalling pathway, cell adhesion and extracellular matrix deposition. We found that negative modulator of Wnt signalling miR‐29b was abundant in NPCE exosomal samples and treatment of TM cells with NPCE EVs significantly decreased COL3A1 expression. Suggesting that miR‐29b can be responsible for decreased levels of WNT/β‐catenin pathway. Overall, this study highlights a potential role of EVs derived from NPCE cells in modulating ECM proteins and TM canonical Wnt signalling.  相似文献
