A seasonal study of the periphyton developed on wooden blocks suspended at five constant depths (0, 0.5, 1, 2, 5 m) in the Pawm-pawm arm of the Volta Lake is reported. The early colonizers at all depths were diatom species, they gradually declined in diversity and numbers and blue-green algae and green algae became more common. After one year the shallower (0–1 m) blocks were covered by a luxuriant growth of filamentous green algae though at lower levels diatoms predominated. Diversity and cell numbers of algae decreased with depth, this was most pronounced below 2 m. It appears that filamentous green algae eventually come to dominate wooden surfaces floating at relatively shallow depths in the Volta Lake. 相似文献

The regulation of the synthesis of alcohol oxidase, catalase, formaldehyde dehydrogenase and formate dehydrogenase was investigated in the methanol-utilizing yeast Hansenula polymorpha. The organism was found to synthesize immunologically identical alcohol oxidases during growth on glycerol and methanol. Growth on glycerol, however, was not dependent on the alcohol oxidase, as was shown with a mutant without alcohol oxidase protein. Similarly it was shown with a catalase activity negative mutant that high catalase activity during growth on glycerol was not a prerequisite for the utilization of this substrate, though absolutely required for growth on methanol.Experiments were conducted with mixed substrates to study the influence of methanol on alcohol oxidase synthesis. In batch cultures, growth on ribose plus methanol resulted in an enhanced rate of alcohol oxidase synthesis as compared to ribose alone. In continuous cultures, (D=0.1 h-1) addition of methanol to glycerol-, glucose-, or sorbose-limited cultures gave rise to increased alcohol oxidase activity of up to 20 U/mg, which is about by 2 times higher than the specific activity used for growth on methanol alone. The increase in specific activity of the dissimilatory enzymes on the mixed substrates is partly due to methanol per se, as was shown by a mutant unable to dissimilate or assimilate methanol. 相似文献

Summary Teeth of three macropod species, M. giganteus, W. bicolor and P. concinna, have been studied using the techniques of light microscopy, scanning- and transmission-electron microscopy and hardness measurement. Light microscope observations showed that the teeth of these species had a translucent enamel region close to the dentine and an outer opaque enamel region at the tooth's surface. These regions were not related to the presence or absence of tubules which are a characteristic feature of marsupial enamel. Hardness tests showed that the opaque enamel was softer than the translucent enamel. Scanning electron microscope observations revealed that there was no correlation between any particular prism packing or orientation and the opaque and translucent enamel regions. Transmission electron microscope observations showed that the translucent enamel region consisted of well defined prisms and well packed, lath-like crystals, whereas the opaque enamel was disrupted by voids (which ranged in size from enlarged micropores to about 2 m in diameter in extreme cases) between crystals and some randomly oriented, loosely packed crystals. This disruption within the opaque enamel region was more common at prism boundaries but pockets of disrupted enamel were also found within prisms and interprismatic regions. The opacity of the enamel was caused by scattering of light from the voids. The ultrastructure of the opaque enamel region indicated that this region was hypomineralized; hardness tests and polarized light microscope observations were consistent with these results. 相似文献

Biomass and activity of planktonic bacteria were investigated during a one year study in a shallow sandpit lake. The shallowness of the lake helped keep the water column homogeneous regarding bacterioplankton. Small free-living bacteria (0.03 µm3 cell–1) dominated the populations throughout the period studied. Fluorogenic substrates can detect enzymatic activity associated with cells. It is difficult, however, to detect activity within a single cell or in an organelle since hydrolytic substrates yield products that rapidly leak from the cell. Several new solutions are presented including trapping the fluorescent product in membranes, in cell organelles, or as a glutathione conjugate. Novel substrates also are described that directly yield highly fluorescent precipitates at the site of enzymatic activity. These can be used for detecting endogenous activity in cells or for enzyme-amplified histochemical detection. Some of these substrates can be used in live cells. 相似文献

We studied colonization dynamics and habitat preferences of macroinvertebrates associated with submerged woody debris in Louisiana coastal plain streams. In an in situ experiment, Branches of magnolia Magnolia grandiflora and water oak Quercus nigra were anchored over sand and gravel substrate at two sites in the Bogue Falaya River, St. Tammany Parish, Louisiana, U.S.A. Branches were removed from the units weekly for 4 weeks and biweekly for 1 month to assess colonization. Invertebrate numbers were highest at weeks three and six on magnolia over gravel habitat, although numbers of the most dominant taxa (Baetidae, Hydropsyche sp., Cernotina sp., Oecetis sp. and Hydroptilia sp.) peaked at different times during the study. Abundances of Hydropsyche sp. and Cernotina sp. were greatest on magnolia branches secured over gravel substrate, whereas Oecetis sp. abundances were greatest on oak branches secured over sand substrate.We also surveyed six streams in southeast Louisiana to examine the relative importance of wood, gravel and sand as substrate for lotic macroinvertebrates. In fall 1997 and spring 1998, we took water samples and measured in situ water quality characteristics at one site on each stream, and then collected submerged wood (surface area about 3051 cm2) and sand and gravel substrate (Surber samples totaling 2800 cm2 for each substrate) to determine the density and taxonomic composition of resident macroinvertebrates. Highest numbers of invertebrates were collected in fall and spring from gravel substrate (P=0.0001). Macronychus sp. were more abundant on wood than gravel or sand (P<0.0003) and Hydropsyche sp. was more abundant on fall wood samples (P=0.0001). Analyses of water quality parameters revealed associations between Hydropsyche sp. abundance and potassium (P=0.0070) and specific conductance (P=0.0080), although both parameters exhibited interactions with substrate type (P<0.0144). 相似文献

Abstract Populations within model seawater-displaced fuel tanks developed as a succession of eukaryotic species. Growth of filamentous fungi was preceded by the growth of several yeast species. More rapid initiation of growth by filamentous fungi was associated with extracellular metabolites produced by yeasts: both the pH flux towards more acidic conditions and the spore germination induction effect influenced population development. The relative binding affinity of each species for the insoluble hydrocarbon fuel is proposed as a major characteristic determining which species proliferated at the fuel-water interface. 相似文献

Candida lipolytica and Candida rugosa were isolated from blood samples from a patient with chronic myeloid leukemia (31 years old) and a patient with sickle cell disease (1-year-old), respectively. Isolates were grown for 48 h at 37 degrees C in either Sabouraud or tryptone soy broth (TSB). Peptidases were tested for using substrate sodium dodecyl sulfate-polyacrylamide gels with gelatin, casein, bovine serum albumin (BSA) or hemoglobin. Enzymography analyses were made on the following substrates: human albumin, IgG and human fibrinogen, which had been incubated with the concentrated supernatants. For C. lipolytica, a approximately 60-kDa gelatin-degrading serine proteolytic activity was found in the TSB supernantant as well as a metallopeptidase activity capable of hydrolysing human albumin, IgG and human fibrinogen. With C. rugosa, albumin, IgG and human fibrinogen substrates were degraded by an aspartyl-like peptidase activity. Supernatants from C. rugosa also showed three serine proteolytic activities towards gelatin (approximately 50 kDa, TSB), casein ( approximately 94 kDa, TSB) and BSA ( approximately 120-kDa, Sabouraud), in addition to a metallopeptidase capable of degrading casein ( approximately 110 kDa, Sabouraud). Little is known about peptidases of C. rugosa and C. lipolytica and this preliminary data may prove useful for future work on host-parasite relationship and antifungal agents. 相似文献
