Tremendous adaptability of insects is predominantly provided by fast tuning of physiological functioning of an organism to the permanently changing environmental conditions. One of the mechanisms of plasticity in insects is modulation of performance of their sense organs by neurohormones. Activity of at least three out of four receptor cells located in cockroach pheromonesensitive sensilla is modulated by octopamine. An increase of firing rate of pheromone receptor cells and a decrease of electroantennogram amplitude is accompanied by enhanced behavioral responses of male cockroaches to sex pheromone. The effect of octopamine on reception of a repellent (1,8-cineole, eucalyptol) by an insect is reported for the first time. Simultaneous modulation of responses of receptor cells located in sex specific sensilla to semantically different odorants indicates their cooperation in formation of insect behavior.  相似文献

The trace biogenic amines tyramine and octopamine are found in the nervous systems of animals ranging in complexity from nematodes to mammals. In insects such as Drosophila melanogaster, the trace amine octopamine is a well-established neuromodulator that mediates a diverse range of physiological processes, but an independent role for tyramine is less clear. Tyramine is synthesized from tyrosine by the enzyme tyrosine decarboxylase (TDC). We previously reported the identification of two Tdc genes in Drosophila: the peripherally-expressed Tdc1 and the neurally-expressed Tdc2. To further clarify the neural functions of the trace amines in Drosophila, we examined normal and cocaine-induced locomotor activity in flies that lack both neural tyramine and octopamine because of mutation in Tdc2 (Tdc2(RO54)). Tdc2(RO54) flies have dramatically reduced basal locomotor activity levels and are hypersensitive to an initial dose of cocaine. Tdc2-targeted expression of the constitutively active inward rectifying potassium channel Kir2.1 replicates these phenotypes, and Tdc2-driven expression of Tdc1 rescues the phenotypes. However, flies that contain no measurable neural octopamine and an excess of tyramine due to a null mutation in the tyramine beta-hydroxylase gene (TbetaH(nM18)) exhibit normal locomotor activity and cocaine responses in spite of showing female sterility due to loss of octopamine. The ability of elevated levels of neural tyramine in TbetaH(nM18) flies to supplant the role of octopamine in adult locomotor and cocaine-induced behaviors, but not in functions related to female fertility, indicates mechanistic differences in the roles of trace amines in these processes.  相似文献

We investigated whether octopamine (OA) is associated with the disappearance of cooperation in Polyrhachis moesta ant queens. Queens of P. moesta facultatively found the colony with genetically unrelated queens. The founding queens perform frequent food exchange with these non-related queens and partake in cooperative brood rearing, whereas single colony queens exclude non-related queens via aggressive behaviour. Thus, aggression is a factor that reduces cooperation. Given that aggression is generally associated with brain OA in insects, we hypothesized that OA controls the behavioural change in cooperation in the ant queen, via an increase in aggression. To test this hypothesis, we compared the amounts of OA and related substances in the brain between founding and colony queens, and observed the interaction of founding queens following oral OA administration. The brain OA levels in colony queens were significantly higher than those in founding queens. Oral administration of OA to founding queens caused significantly less trophallaxis and allogrooming behaviour than in the control founding queens, but with no significant increase in aggression. These results suggest that OA promotes the disappearance of cooperation in founding queens of P. moesta. This is the first study to reveal the neuroendocrine mechanism of cooperation in ant queens.  相似文献

Major metabolites of octopamine and tyramine in the Limulus nervous system are identified here as gamma-glutamyl octopamine and gamma-glutamyl tyramine. We show that these conjugates are normal products of amine metabolism in Limulus, and that they are normally present in octopamine-rich Limulus tissues. The synthesis of these conjugates is not restricted to nervous tissue, but the highest activity of gamma-glutamyl amine synthetase was measured in the CNS. Our interest in these molecules stems from our previous observations which showed that they were synthesized and stored in, and released from, the efferent fibers to Limulus eyes which modulate the sensitivity of the eyes to light. Here we provide direct evidence for the release of the conjugates from Limulus eyes in response to depolarization, and that gamma-glutamyl octopamine can increase the sensitivity of the lateral eye to light. Our observations lend support to the hypothesis that gamma-glutamyl octopamine may serve as an intercellular messenger in the Limulus visual system.  相似文献

【目的】为了探讨章鱼胺受体2基因(Octopamine receptor 2,OA2B2)在棉铃虫Helicoverpa armigera(Hübner)体内表达谱及激素对其调控作用,为今后研究Ha OA2B2受体基因在转录调控中的作用,开发新型杀虫剂提供一定的理论依据。【方法】本实验采用QRT-PCR技术、激素处理、RNAi介导的蜕皮激素相关核受体沉默等方法对Ha OA2B2受体基因在棉铃虫不同发育阶段及组织中的表达谱与激素调控进行探究。【结果】生物信息学分析表明,Ha OA2B2受体基因编码框长度1 191 bp,编码396个氨基酸。Ha OA2B2受体基因氨基酸序列与家蚕Bombyx mori一致性高达89%。Ha OA2B2受体基因在棉铃虫各时期均有表达,尤其在5龄第3天转录水平达到最高;Ha OA2B2受体基因在棉铃虫幼虫头部表达量最高。蜕皮激素可促进Ha OA2B2受体基因的表达。【结论】分析了该基因的序列特征和表达谱,蜕皮激素对其表达量的影响,为今后研究Ha OA2B2受体基因在转录调控中的作用以及开发新型杀虫剂提供一定的理论依据。  相似文献

Series of agonists and antagonists were examined for their actions on a Bombyx moriα‐adrenergic‐like octopamine receptor (OAR) stably expressed in HEK‐293 cells. The rank order of potency of the agonists was clonidine>naphazoline>tolazoline in Ca2+ mobilization assays, and that of the antagonists was chlorpromazine>yohimbine. These findings suggest that the B. mori OAR is more closely related to the class‐1 OAR in the intact tissue than to the other classes. N′‐(4‐Chloro‐o‐tolyl)‐N‐methylformamidine (DMCDM) and 2‐(2,6‐diethylphenylimino)imidazolidine (NC‐5) elevated the intracellular calcium concentration ([Ca2+]i) with EC50s of 92.8 µM and 15.2 nM, respectively. DMCDM and NC‐5 led to increases in intracellular cAMP concentration ([cAMP]i) with EC50s of 234 nM and 125 nM, respectively. The difference in DMCDM potencies between the cAMP and Ca2+ assays might be due to “functional selectivity.” The Ca2+ and cAMP assay results for DMCDM suggest that the elevation of [cAMP]i, but not that of [Ca2+]i, might account for the insecticidal effect of formamidine insecticides. © 2009 Wiley Periodicals, Inc.  相似文献

Both adipokinetic hormone and octopamine have a stimulating effect on lipid release from locust fat body in vitro, when incubated in diluted haemolymph. The presence of adipokinetic hormone results in the formation of the flight-specific haemolymph lipoprotein A+ accepting the increased amount of lipids released into the incubation medium. In contrast, interconversions of lipoproteins do not occur when octopamine is added to the incubation medium, which is in line with the expectations: the lipid-mobilizing effect of octopamine is a limited and short-term effect. When fat body tissue is incubated with isolated haemolymph protein fractions, the lipid-mobilizing effect of adipokinetic hormone only occurs when the incubation medium contains both lipoprotein, Ay and protein fraction C, resulting in the formation of lipoprotein A+. In similar control incubations with the hormone omitted, some lipoprotein A+ is also formed (concomitant with a slight amount of lipid released), though significantly less than in incubations with hormone. Besides a stimulating function on lipolytic processes in the fat body, adipokinetic hormone is suggested to influence haemolymph lipoprotein rearrangement. A possible counteracting function of another factor in the haemolymph is discussed.  相似文献
