The paper presents an analysis of locomotion and location in a vertical water column of 10Biomphalaria glabrata, under constant conditions of light, temperature, and food availability. Individual snails varied in distance traveled, and in the percentage of time spent in different areas of the water column. Distance covered ranged from 53 cm to 100 cm h–1 . Approximately 21% of locomotion was observed to be passive (floating up or sinking down). Snails spent approximately 58% of the time in the bottom section, 35% in the top section and only 7% in the middle region. The study provides baseline data that can be used to compare further studies ofBiomphalaria glabrata as various conditions are altered.  相似文献

Summary Objective, accurate, non-intrusive measurement of in vitro cell growth was realized through microcomputerized video image analysis. Recently-released video and digitizing hardware and software were incorporated into an analytical system which accurately quantified visual differences between cultures on a cell number or fresh mass basis. Sequential measurements during culture incubation further detected and quantified subtle changes in colony area and density resulting from growth. Each measurement was acquired rapidly, without encroaching on the in vitro environment, so cell growth was undisturbed. Custom software routines coordinated the quantification of this detailed record into precise cumulative growth curves.  相似文献

Abstract. The arrival of individually marked Glossina pallidipes Austen at a host odour source after their video-timed release from 30–75 m downwind was measured in the field in Zimbabwe. In the absence of odour, the proportion recaptured was <2% (= - random expectation); when synthetic ox odour was released, the probability of recapture at the source increased with proximity of release, from 6% at 75 m to 21% at 30 m (about twice this number arrived within ∼2 m of the source). There were two distinct distributions of recaptures: a 'fast' cohort which found the source within 40 s, and a 'slow' cohort which took from one to >20 min, with ∼50% of the flies in each cohort. The fastest flies probably reached the source in a single, mainly straight flight from take-off, at an overall average (straight line) displacement speed of 2.8-4.5 ms-1 (i.e. close to the preferred flight speed of ∼5 m s-1). The flies apparently maintained their ground speed largely independent of the wind speed they headed into. The 'slow' cohort had a constant probability of arrival at the source, presumably after losing and re-contacting the plume, and after having stopped at least once on the way. There were no marked correlations with wind parameters, although the probability of recapture increased slightly with the directness of the wind from the source, and the probability of 'slow' flight increased slightly with wind speed. It is inferred that a repeated sequence of anemotactic 'aim-then-shoot' orientation at take-off plus optomotor-steered in-flight correction of direction is used as a form of biassed random walk to bring the flies close to the odour source, rather than the use of moth-type anemotactic zigzagging.  相似文献

ABSTRACT. The responses of adult male Spodoptera littoralis (Boisd.) (Noctuidae) to components of the female sex pheromone were examined in a wind tunnel. The responses of male moths from both Egyptian and Cretan stocks to the primary 'attractant' (Z, E)-9,11 tetradecadienyl acetate (III), were compared with the responses to III combined with various percentages of the secondary components tetradecyl acetate (I), (Z)-9-tetradecenyl acetate (IIA), (E)-11-tetradecenyl acetate (IIB), IIA and IIB combined, and(Z, E)-9,12-tetradecadienyl acetate (IV). Analysis of the flight behaviour recorded on videotape showed that with one exception (IV), the secondary components increased the males' success in reaching the source and increased the length of time they spent at the source. In most cases, increasing the relative percentage of the secondary component caused a reduction in the number of males responding. The results are compared with those obtained in field trials with S.littoralis.  相似文献

Locomotor behavior and substrate use of cage-reared golden lion tamarins (Leontopithecus rosalia), newly released and free-ranging on the grounds of the National Zoological Park, were studied to determine if locomotion changed following exposure to naturalistic conditions. The animals employed a predominantly quadrupedal locomotor profile, incorporating leaping and vertical climbing to a lesser degree. There was no clear evidence of a change in locomotion due to the high degree of variability in these behaviors. The locomotor repertoire of the free-ranging group differed from that of groups occupying unenriched but relatively large conventional enclosures, indicating that whereas locomotion is plastic with respect to environment, substrate characteristics influence locomotor behavior and may promote stereotypical behavior. However, due to anatomical constraints, the locomotor repertoire tended to be less variable than substrate use. Similar behaviors were used in moving through a variety of habitat features in spite of strong associations between specific locomotor styles and substrate classes. © 1994 Wiley-Liss, Inc.  相似文献

Optical “clearing” is a cost saving method for preparing large numbers of whole, dissected or thickly sectioned cytological specimens such as plant ovules and ovaries. Minimal labor is required and specimens retain three-dimensional integrity. Previous development of high contrast stain-clearing methods using hemalum to impart contrast has facilitated analysis and photography under brightfield illumination for small ovules. The deep stain intensity of hemalum, however, often precludes adequate light transmission and contrast within internal focal planes, limiting the applicability of hemalum-based stain-clearing to small specimens. Having encountered this problem for nucelli of cotton (Gossypium barbadense L.), which are roughly 300 μm thick at fertilization, we have developed a modified stain-clearing system. The two key features of these new methods are the use of azure, C, which allows the intensity of staining to be readily regulated, and contrast manipulation via video signal and image processing. Intensity of azure C stain was readily controlled by modifying the staining and/or dehydration media to produce relatively low contrast specimens. Analysis was facilitated by indirect viewing on a video monitor using adjustments of sensitivity, exposure, and contrast of the charge-coupled device (CCD) camera. Digital processing provided further enhancement. Acceptable images were obtained from virtually all specimens. These methods, which combine low contrast (high transmittance) specimens with high contrast imaging, should facilitate data acquisition on reproduction, thus the developmental and genetic characterization of reproductive mutants. Other applications, e.g., in pathology and embryology, are readily envisioned.  相似文献

To develop and test a system for computer-assisted image analysis, repeated video recordings of reed canary-grass roots (Phalaris arundinacea L.) were made in an 18-window rhizotron. The images were digitized and processed using a Unix computer and the Khoros software development environment.Two image sizes, 126×95 mm and 61×46 mm, both comprising 650 × 490 pixels, were compared. Among image processing techniques used were median filtering, segmentation and skeletonization. Root area and length in both the topsoil and subsoil were estimated using the two image sizes. The resolution (image size) strongly affected the calculated root lengths. The results were compared with root length measurements obtained manually.Statistically significant differences in root length and area in the topsoil were detected between the sampling dates using the computer-assisted methods. Possible sources of error and methods for reducing them are discussed.  相似文献

Handheld and endoscopic optical‐sectioning microscopes are being developed for noninvasive screening and intraoperative consultation. Imaging a large extent of tissue is often desired, but miniature in vivo microscopes tend to suffer from limited fields of view. To extend the imaging field during clinical use, we have developed a real‐time video mosaicking method, which allows users to efficiently survey larger areas of tissue. Here, we modified a previous post‐processing mosaicking method so that real‐time mosaicking is possible at >30 frames/second when using a device that outputs images that are 400 × 400 pixels in size. Unlike other real‐time mosaicking methods, our strategy can accommodate image rotations and deformations that often occur during clinical use of a handheld microscope. We perform a feasibility study to demonstrate that the use of real‐time mosaicking is necessary to enable efficient sampling of a desired imaging field when using a handheld dual‐axis confocal microscope.  相似文献
