【目的】 在我国南方尤其是西南地区,光叶紫花苕(Vicia villosa Roth.)作为重要的青饲和绿肥两用豆科作物被广泛种植,有助于提高土壤氮素和后茬作物的产量品质。接种有益微生物是促进豆科作物生物固氮和生长的重要措施之一。为此,本文研究了一株自主分离获得的白腐真菌¾¾撕裂蜡孔菌(Careporia lacerata HG2011)对光叶紫花苕结瘤固氮和生长的影响,并揭示其潜在机制。【方法】 采用微生物培养、植物培养和田间试验,研究C. lacerata磷铁活化能力、代谢产物构成、与根瘤菌Rhizobium sophorae S3的相互作用,及其对光叶紫花苕结瘤、生长、产量、品质和土壤有效磷铁的影响。【结果】 C. lacerata和根瘤菌之间无拮抗作用。液相色谱-质谱(liquid chromatography-mass spectrometry, LC-MS)分析发现,C. lacerata发酵液含有氨基酸、有机酸和类黄酮等化感物质,能增强根瘤菌的趋化性并促进生物膜形成。此外,C. lacerata还能释放生长素、赤霉素、水杨酸和铁载体,活化难溶性有机和无机磷。在植物培养试验中,单独接种C. lacerata或根瘤菌均能促进光叶紫花苕生长,但以共接种处理效果最佳。C. lacerata定殖于光叶紫花苕根际,导致根长、根系表面积和结瘤数显著增加。田间试验发现,接种C. lacerata显著提高了光叶紫花苕单株根瘤数、根瘤质量和固氮酶活性,以及土壤有效磷铁含量和磷酸酶活性,产量比常规施肥处理增加12.15%且品质无显著变化。【结论】 C. lacerata能够在光叶紫花苕根际定殖,通过分泌化感物质、生长素和活化土壤磷铁等机制促进结瘤固氮和生长发育。C. lacerata易于培养,菌剂制备成本低廉,施用简便,对提高豆科作物产量品质具有一定应用价值。  相似文献

转运蛋白是一类膜蛋白,可介导生物膜内外化学物质的跨膜转运及信号交换。有机酸转运蛋白在微生物有机酸代谢的跨膜转运过程中发挥重要作用,根据转运蛋白有机酸转运的方向不同可以分为摄取转运蛋白和外排转运蛋白。在微生物代谢中,有些有机酸可以作为能源直接参与体内代谢,有些是能量转换过程中的重要中间产物;摄取转运蛋白的过表达,可以促进微生物细胞获取能源物质,高效的生产目标产物;有机酸摄取转运蛋白敲除或外排转运蛋白表达,有利于底盘细胞外排更多目标产物,进而促进有机酸的生物合成。研究有机酸转运蛋白的结构和功能,有助于解析微生物细胞有机酸生物合成及利用的机制,对于提高工业微生物对有机酸的利用及生物合成具有重要作用。本文综述了微生物有机酸转运蛋白分类和结构、转运方式和转运功能等方面,重点综述了转运蛋白在有机酸生产中的应用,为工业微生物有机酸的高效生物合成及未来发展提供参考。  相似文献

Abstract. To document the relationship between a plant's position in the canopy and its leaf nutrient content, leaf nitrogen and phosphorus were determined for 30 species growing in mature evergreen lowland rain forest at La Selva Biological Station, Costa Rica. Species that grow either in the understory, midstory, or the canopy were selected. Species were further separated into three life forms: self-supporting monocots, self-supporting dicots, and climbers. Mass-based nutrient concentrations were expected to decrease with stature, as has been reported in studies of other forests. In fact, mass-based nitrogen and phosphorus did not vary significantly among the three adult-stature classes, although area-based values differed greatly: canopy plants averaged 60 % more nitrogen and 90 % more phosphorus per unit leaf area than understory plants. Differences in leaf characteristics were evident among the three life forms. Most notably, area-based phosphorus and leaf specific mass were lowest in climbers, intermediate in self-supporting dicots, and highest in self-supporting monocots. These results support the characterization of climbers as investing in inexpensive structures, perhaps in order to gain competitive advantage in light capture by allocating resources to maximize elongation rates.  相似文献

The purpose of this article is to describe the treatment of a 45-year-old male with a hyperfunctional voice disorder by a biofeedback therapist and a speech-language pathologist. The interdisciplinary approach to the treatment of this voice disorder involved the combined use of traditional voice therapy techniques and EMG biofeedback procedures together with cognitive behavioral therapy. Voice therapy was facilitated through the use of a computer-based, speech-monitoring system. The remediation of this voice disorder was attributed to the collaborative efforts of two professionals representing diverse professional training and treatment protocols. The results showed reductions in muscle activity in the infrahyoid and laryngeal areas as well as improved use of proper breathing and voicing onset behaviors. Follow-up at 10 and 15 months posttreatment intervals indicated that the client had retained all target voice skills and the tension reduction/biofeedback skills. Results suggest that interdisciplinary, collaborative efforts using biofeedback and voice therapy can prove beneficial in the treatment of hyperfunctional voice disorders.Note: Neither author has been compensated in any way for the use of the CAFET System nor does either have any financial interest in the company.  相似文献

Group-hunting is ubiquitous across animal taxa and has received considerable attention in the context of its functions. By contrast much less is known about the mechanisms by which grouping predators hunt their prey. This is primarily due to a lack of experimental manipulation alongside logistical difficulties quantifying the behaviour of multiple predators at high spatiotemporal resolution as they search, select, and capture wild prey. However, the use of new remote-sensing technologies and a broadening of the focal taxa beyond apex predators provides researchers with a great opportunity to discern accurately how multiple predators hunt together and not just whether doing so provides hunters with a per capita benefit. We incorporate many ideas from collective behaviour and locomotion throughout this review to make testable predictions for future researchers and pay particular attention to the role that computer simulation can play in a feedback loop with empirical data collection. Our review of the literature showed that the breadth of predator:prey size ratios among the taxa that can be considered to hunt as a group is very large (<100 to >102). We therefore synthesised the literature with respect to these predator:prey ratios and found that they promoted different hunting mechanisms. Additionally, these different hunting mechanisms are also related to particular stages of the hunt (search, selection, capture) and thus we structure our review in accordance with these two factors (stage of the hunt and predator:prey size ratio). We identify several novel group-hunting mechanisms which are largely untested, particularly under field conditions, and we also highlight a range of potential study organisms that are amenable to experimental testing of these mechanisms in connection with tracking technology. We believe that a combination of new hypotheses, study systems and methodological approaches should help push the field of group-hunting in new directions.  相似文献

Summary The rate of ethanolic fermentation of high gravity wheat mashes bySaccharomyces cerevisiae was increased by nitrogen sources such as ammonium sulfate or arginine. This stimulation was mediated through increased proliferation of cells. Large quantities of proline, however, were excreted by the yeast into the medium when arginine was added as a nutrient supplement. The amount of proline excreted was proportional to the concentration of arginine supplied. Nitrogen sources such as ammonium sulfate or lysine enhanced the production of proline from arginine and its excretion into the medium. Results show that the stimulation of very high gravity fermentation by arginine is not merely through provision of a source of nitrogen but also because it serves as a precursor for the production of proline, a compound which may play a significant role in alleviating the effects of osmotic stress.  相似文献

Aim

Coastal fishes have a fundamental role in marine ecosystem functioning and contributions to people, but face increasing threats due to climate change, habitat degradation and overexploitation. The extent to which human pressures are impacting coastal fish biodiversity in comparison with geographic and environmental factors at large spatial scale is still under scrutiny. Here, we took advantage of environmental DNA (eDNA) metabarcoding to investigate the relationship between fish biodiversity, including taxonomic and genetic components, and environmental but also socio-economic factors.

Methods

We analysed fish eDNA in 263 stations (samples) in 68 sites distributed across polar, temperate and tropical regions. We modelled the effect of environmental, geographic and socio-economic factors on α- and β-diversity. We then computed the partial effect of each factor on several fish biodiversity components using taxonomic molecular units (MOTU) and genetic sequences. We also investigated the relationship between fish genetic α- and β-diversity measured from our barcodes, and phylogenetic but also functional diversity.

Results

We show that fish eDNA MOTU and sequence α- and β-diversity have the strongest correlation with environmental factors on coastal ecosystems worldwide. However, our models also reveal a negative correlation between biodiversity and human dependence on marine ecosystems. In areas with high dependence, diversity of all fish, cryptobenthic fish and large fish MOTUs declined steeply. Finally, we show that a sequence diversity index, accounting for genetic distance between pairs of MOTUs, within and between communities, is a reliable proxy of phylogenetic and functional diversity.

Main conclusions

Together, our results demonstrate that short eDNA sequences can be used to assess climate and direct human impacts on marine biodiversity at large scale in the Anthropocene and can further be extended to investigate biodiversity in its phylogenetic and functional dimensions.  相似文献

The CO2 enrichment effects (300–650 µmol mol-1) on mineral concentration (N, P, K, Ca, Mg, Mn, Fe, Zn), absolute total mineral contents per individual and of whole stands of four herbaceous (Trifolium repens L.,Trifolium pratense L.,Lolium perenne L.,Festuca pratensis HUDS.) and two woody species (Acer pseudo-platanus L.,Fagus sylvatica L.) were investigated.In general, the mineral concentration of the plant tissues decreased (all six species: N>Ca>K>Mg) with the exception of P. Mn and Fe were only determined for the tree species. Both decreased in concentration (Mn>Fe). Zn was only analysed forTrifolium pratense andFestuca pratensis and decreased significantly in the grass.Despite of decreases in concentrations of as much as 20% in some cases there were increases in absolute amounts per individual and, therefore, in the whole vegetation up to 25% because of the enhanced dry matter accumulation at elevated CO2 supply.Dedicated to Prof. Dr. R.
