We used the polymerase chain reaction (PCR) and direct DNA sequencing to study genetic variation within and among populations of Atlantic cod, Gadus morhua , in the western North Atlantic. In a 307 bp region of the mitochondrial cytochrome b gene, 24 variable nucleotide positions define 24 genotypes, which differ by from one to six nucleotide substitutions. Greenland cod ( G. ogac ) differs from the most similar G. morhua genotype by an additional 12 nucleotide substitutions. Silent transitions dominate both intra- and interspecific comparisons, however four nucleotide substitutions within morhua result in amino acid replacements. Direct sequencing of DNA reveals substantially more of the genetic variation that exists within and between species than do previous indirect methods based on restriction fragment length polymorphisms, and thus has far greater potential to quantify such differences as may exist among fish stocks. Preliminary experiments also indicate that automation of DNA sequencing provides an efficient, rapid, and accurate means for detection of genetic variation in natural populations offish.  相似文献

Oviposition by a parasitoid wasp in an already parasitized host (= superparasitism) may result in larval competition and the loss of offspring. Consequently, in solitary species, the decision to superparasitize should be based on the probability of offspring survival. Females of Aphidius ervi Haliday and A. smithi Sharma & Subba Rao (Hymenoptera: Aphidiidae), two parasitoids of the pea aphid, discriminated between conspecific-and heterospecific-parasitized hosts. Both species showed partial preference for aphids previously attacked by A. smithi, a result predicted from the fact that A. ervi is superior to A. smithi in larval competition under most conditions. Females of A. smithi selectively attacked conspecific-parasitized pea aphids when given a choice between these and self-parasitized hosts. Results indicated that females of both species responded to an external pheromone-like marker to discriminate between selfconspecific-and heterospecific-parasitized aphids. In addition, A. smithi may use internal cues to recognize, and avoid oviposition in, aphids previously parasitized by A. ervi.
Résumé L'oviposition par une guêpe parasite dans un hôte qui a déjà été parasité (= superparasitisme) pourrait conduire à une compétition entre larves et à une perte de progéniture. Par conséquent, dans les espèces solitaires, la décision de superparasiter devrait être basée sur la probabilité de survie de la progéniture. Les femelles de Aphidius ervi Haliday et de A. smithi Sharma & Subba Rao (Hymenoptera: Aphidiidae), deux parasites du puceron du pois, ont discriminé entre les hôtes parasités par des individus de la même espèce et ceux parasités par des individus d'une espèce différente. Les deux espèces de parasite ont montré une préférence partielle pour les pucerons préalablement parasités par A. smithi, résultat qui était prévisible du fait que, dans la majorité des cas, A. ervi est supérieur à A. smithi dans la compétition entre larves. En plus, A. smithi pourrait utiliser des signaux internes pour reconnaître et éviter l'oviposition dans les pucerons préalablement parasités par A. ervi.

Time-related changes in the patterns of host discrimination may have a complex form depending on whether or not hosts are marked following first parasitisation, the persistence of chemicals in these marks, and changes in the host as a result of being parasitised. Here we present an experimental design capable of detecting temporal patterns in host discrimination of solitary parasitoids. This method is tested against experimental data from Aphidius ervi attacking the aphid, Acyrthosiphon kondoi. The results demonstrate that there are statistically significant changes in the patterns of egg distribution in aphid hosts held for intervals between 1 and 48 h between parasitisation. These changes appear to be attributable to increasing oviposition restraint by the superparasitising female as the time interval between attacks is increased. The form of this relationship is not significantly different to a sigmoid function and the rate of change of restraint was greatest between 6–8 h after the first oviposition.  相似文献

Summary The natural abundance13C/12C ratios (as δ13C) of organic matter of marine macroalgae from Fife and Angus (East Scotland) were measured for comparison with the species' ability to use CO2 and HCO 3 - for photosynthesis, as deduced from previously published pH-drift measurements. There was a clear difference in δ13C values for species able or unable to use HCO 3 - . Six species of Chlorophyta, 12 species of Phaeophyta and 8 species of Rhodophyta that the pH-drift data suggested could use HCO 3 - had δ13C values in the range -8.81‰ to -22.55‰. A further 6 species of Rhodophyta which the pH-drift data suggested could only use CO2 had δ13C values in the range -29.90‰ to-34.51‰. One of these six species (Lomentaria articulata) is intertidal; the other five are subtidal and so have no access to atmospheric CO2 to complicate the analysis. For these species, calculations based on the measured δ13C of the algae, the δ13C of CO2 in seawater, and the known13C/12C discrimination of CO2 diffusion and RUBISCO carboxylation suggest that only 15–21% of the limitation to photosynthesisin situ results from CO2 diffusion from the bulk medium to the plastids; the remaining 79–85% is associated with carboxylation reactions (and, via feedback effects, down-stream processes). This analysis has been extended for one of these five species,Delesseria sanguinea, by incorporating data onin situ specific growth rates, respiratory rates measured in the laboratory, and applying Fick's law of diffusion to calculate a boundary layer thickness of 17–24 μm. This value is reasonable for aDelesseria sanguinea frondin situ. For HCO 3 - -using marine macroalgae the range of δ13C values measured can be accommodated by a CO2 efflux from algal cells which range from 0.306 of the gross HCO 3 - influx forEnteromorpha intestinalis13C=-8.81‰) in a rockpool to 0.787 forChondrus crispus13C=-22.55‰). The relatively high computed CO2 efflux for those HCO 3 - -users with the more negative δ13C values implies a relatively high photon cost of C assimilation; the observed photon costs can be accommodated by assuming coupled, energy-independent inorganic carbon influx and efflux. The observed δ13C values are also interpreted in terms of water movement regimes and obtaining CO2 from the atmosphere. Published δ13C values for freshwater macrophytes were compared with the ability of the species to use CO2 and HCO 3 - and again there was an apparent separation in δ13C values for these two groups. δ13C values obtained for marine macroalgae for which no pH-drift data are available permit predictions, as yet untested, as to whether they use predominantly CO2 or HCO 3 -  相似文献

A photosynthate labelling method is presented which takes advantage of the natural difference in carbon-isotope composition () which exists between atmospheric CO2 (-8) and commercially available compressed CO2. Carbon dioxide with -4.0 and –27.9%., respectively, has been used for labelling. A plant growth cabinet served as the labelling compartment. CO2-free air was continuously injected at a rate of up to 54m3·h–1. Dilution of cabinet CO2 by CO2-free air was counterbalanced by addition of CO2 with known constant . Since the labelling-cabinet atmosphere was continuously exchanged at a high rate, photosynthetic carbon-isotope discrimination was fully expressed. In order to study the distribution of carbon acquired by the plant during a defined growth period, the of CO2 was modified by replacing, for example, atmospheric CO2 by CO2 with –27.9%. and the weight and 5 of plant carbon pools was monitored over time. In such an experiment the change of CO2 was followed by a rapid change of the of sucrose in mature flag-leaf blades of wheat (Triticum aestivum L.). The 5 of sucrose stabilized near –51%., indicating complete exchange by current photosynthate. In contrast 83% of the total carbon in mature flag-leaf blades was not exchanged after 14 d continuous labelling. Differential labelling of pre- and post-anthesis photosynthate indicated that 13% of grain carbon originated from pre-anthesis photosynthesis. Carbon-isotope discrimination and its consideration in experimentation and labelling data evaluation are discussed in detail. A field experiment was conducted to investigate if carbon isotope (13C) discrimination () measured at the vegetative stage of spring wheat (Triticum turgidum L. var. durum) is related with the yield and water use efficiency (WUE) at ripening. A line source sprinkler irrigation system exposed the wheat genotypes to different watering regimes, from rainfed to full irrigation and thereby increased the range in yield and WUE attainable in the four genotypes studied. The results indicated that values measured at the late stem elongation stage 60 days after planting (DAP), showed strong positive correlation with total dry matter yield (r=0.732***), and a highly significant negative correlation with WUE (r=–0.755***) measured at ripening 105 DAP. The data suggest that the imprints of measured at vegetative growth stage persists throughout the entire growth period, until maturity. Subject to confirmation from additional studies in other crops and locations, early measurements of may prove a useful tool for rapid and early screening of cultivars, for high yield and high WUE.  相似文献

The carbon isotope composition of terrestrial C4 plants depends on the primary carboxylation of phosphoenolpyruvate (PEP) and on the diffusion of CO2 to the carboxylation sites, but is also influenced by the final carboxylation of ribulose-1,5-bisphosphate (RuBP). Several models have been used for reproducing this complex situation. In the present review, a particular model is applied as a means to interpret the effects of environmental and genetically determined factors on carbon isotope discrimination during C4 photosynthesis. As a new feature, the model considers four types of limitation of the overall CO2 assimilation rate. Both carboxylation reactions are assumed to be limited by either maximum enzyme activity or maximum substrate regeneration rate. The model is applied to experimental data on the effects of CO2, irradiance and water stress on short-term discrimination by leaves of several C4 species measured simultaneously with CO2 gas exchange characteristics. In particular, different patterns of the influence of low irradiances on carbon isotope discrimination are interpreted as due to variations in that irradiance at which a transition from limitation by PEP regeneration rate and RuBP carboxylase activity to limitation by the regeneration rates of both substrates occurs. After discussing literature data on the effects of environmental conditions on carbon isotope discrimination by C4 plants seasonal and developmental changes in carbon isotope composition, studies on the systematic and geographic distribution of C4 plants, evolutionary and genetical aspects, and some ecological implications are reviewed.  相似文献

Carbon isotope discrimination in C3–C4 intermediates is determined by fractionations during diffusion and the biochemical fractionations occurring during CO2 fixation. These biochemical fractionations in turn depend on the fractionation by Rubisco in the mesophyll, the amount of CO2 fixation. These biochemical fractionations in turn depend on the fractionation by Rubisco in the mesophyll, the amount of CO2 fixation occurring in the bundle sheath, the extent of bundle-sheath leakiness and the contribution which C4-cycle activity makes to the CO2 pool there. In most instances, carbon isotope discrimination in C3–C4 intermediates is C3-like because only a small fraction of the total carbon fixed is fixed in the bundle sheath. In particular, this must be the case for Flaveria intermediates which initially fix substantial amounts of CO2 into C4-acids. In C3–C4 intermediates that refix photorespiratory CO2 alone, it is possible for carbon isotope discrimination to be greater than in C3-species, particularly at low CO2 pressures or at high leaf temperatures. Short-term measurements of carbon isotope discrimination and gas exchange of leaves can be used to study the photosynthetic pathways of C3-C4 intermediates and their hybrids as has recently been done for C3 and C4 species.  相似文献

ATP-dependent Sr2+ transport was examined in vitro using basolateral membrane (BLM) vesicles isolated from rat renal cortex to clarify the discrimination mechanisms between strontium (Sr) and calcium (Ca) in renal tubules during reabsorption. ATP-dependent Sr2+ uptake and Ca2+ uptake were observed in renal BLM vesicles and were inhibited by vanadate. Hill plots indicate similar kinetic behavior for Ca2+ and Sr2+ uptake. The apparentK m andV max of ATP-dependent Sr2+ uptake were both higher than those for Ca2+ uptake. ATP-dependent Sr2+ uptake by BLM vesicles diminished in the presence of 0.1 μM Ca2+ and was more markedly inhibited by 1 μM Ca2+. Hill plots of Sr2+ uptake data with and without 0.1 μM Ca2+ showed that the cooperative behavior of Sr2+ uptake was not changed by Ca2+. In the presence of 0.1 μM Ca2+, the affinity of the transport system for Sr2+ and the velocity of Sr2+ uptake in the BLM were both decreased. However, the rate of Ca2+ uptake was not diminished by Sr2+ concentrations of <1.6 μM. These results suggest that Ca2+ is preferentially transported in the renal cortex BLM when Ca2+ and Sr2+ are present at the same time.  相似文献

A circular dichroism-based detection system presents several advantages in the HPLC analysis of chiral compounds because of the selective monitoring of optically active molecules. Its use allows reliable determination of enantiomeric excesses and elution order. To this end, the application of empirical, semiempirical, and nonempirical methods to get stereochemical information from the CD signal is reported. Furthermore, recording the CD spectra on line and evaluation of the dissymetry factor make the CD detection very powerful in characterizing the stereochemistry of chiral eluates.  相似文献
