In vitro induction of tetraploidy in mulberry (Morus alba L.)   总被引:6,自引:0,他引:6  
A high frequency of tetraploidy was induced in mulberry (Morus alba L.) through apical bud treatment under in vitro conditions. Apical buds from in vitro-grown plants were treated with three different concentrations (0.05, 0.1 and 0.2%) of colchicine in MS medium for 24 h. Tetraploidy at a frequency of 39.4±4.8% was obtained using 0.1% colchicine, whereas the frequency of tetraploidy was significantly reduced to 16.7±2.3% when 0.2% colchicine was used. Morphological, histological and cytological evidence indicated a phenotypic and genomic similarity of in vitro- with ex vitro-induced tetraploids. Rooting of tetraploids was on basal medium containing 2.6 μm NAA. The recovery of tetraploids was 80.8% more efficient using the in vitro method instead of the ex vitro method. The use of the same colchicine medium for up to 4 weeks with additional explants was found to be equally effective for the induction of tetraploidy. 用RT-PCR和RACE技术在NO3-诱导处理的小麦(Triticum aestivum L.)根中克隆到一个硝酸根转运蛋白基因的cDNA,命名为TaNRT2.3(GenBank登录号AY053452).序列分析表明,TaNRT2.3全长1 744 bp,其中含有1 521bp的ORF,编码507个氨基酸,具有12个跨膜区,属于MFS超基因家族中的NNP家族.TaNRT2.3与其他植物中已知的NRT2具有很高的同源性.Northern杂交表明:TaNRT2具有在根中表达的组织特异性,而在叶中未检测到.TaNRT2的表达受NO3-诱导,在含NH4 介质中不表达.NO3-在低浓度(5~200μmol/L)和高浓度(2.0 mmol/L)时均起作用.通过研究小麦在0.2 mmol/LNO3-条件下TaNRT2的表达水平及对NO3-的吸收效率,表明TaNRT2在小麦高效吸收NO3-方面起着重要的作用.分根实验表明植物中N循环本身可以作为吸收N的调节信号.  相似文献

从山葡萄(Vitis amurensis Rubr.)叶片中分离到长度为1216 bp的山葡萄Class Ⅲ几丁质酶基因VCH3的5’端非编码区(GenBank number AF441123),并发现有两个反向水杨酸(SA)顺式作用元件(TGACG)分别位于转录起始位点上游-1 181 bp和-293 bp处。为了鉴定VCH3启动子的功能,我们将该启动子的4个缺失片段(-1187 bp~ 7bp,-892 bp~ 7 bp,-589 bp~ 7 bp及-276 bp~ 7 bp)分别连接到β-葡糖苷酸酶基因(GUS)编码区的上游,构建成4个融合基因,并利用农杆菌介导叶盘转化法将这4个融合基因转入烟草(Nicotiana tobacum L.)栽培品种NC89中。SA处理的转基因烟草根系GUS酶活性的荧光检测结果表明:只含有TATA盒和CAAT盒而缺失了所有SA顺式作用元件的VCH3(-276)GUS表达盒对SA处理表现出较低程度的诱导性;只包含一个SA顺式作用元件的VCH3(-589)GUS或VCH3(-892)GUS表达盒表现出相似的较高水平的GUS酶活性;而包含两个SA顺式作用元件的全长启动子片段(-1187bp~ 7bp)驱动了最强的GUS酶活性,这说明VCH3启动子诱导的GUS酶活性的最大量表达需要两个SA顺式作用元件的协同作用。GUS酶的组织化学分析结果表明,在SA处理的转基因烟草根横切面中,VCH3启动子4个缺失片段驱动的GUS酶活性在维管组织中的表达活性均强于在外皮层和内皮层的表达  相似文献

Synthesis and secretion of bactericidal protein (cecropin) and lysozyme were induced by soluble peptidoglycan fragments (SPG) from Escherichia coli in a culture of fat body from Bombyx mori larvae. The rate of the secretion by fat body increased as a function of SPG concentration added to the culture medium. The induction of bactericidal activity was specific for peptidoglycan of a particular structure. Thus, SPG from Micrococcus luteus was 500-times less potent than E. coli SPG, and various glucans and peptides structurally related to peptidoglycan were all ineffective as elicitor. These results support the hypothesis that bacteria invading the haemocoel have to be partially degraded to generate peptidoglycan fragments as a signal molecule, which subsequently acts on a receptor on fat body cells and induces antibacterial protein synthesis.  相似文献

Few tropical species have been tested for their flowering response under controlled conditions. Hyptis brevipes Poit, is an annual herb, commonly found in wet margins of streams and ponds, being considered a weed for some perennial plantations in Brazil. Under experimental glasshouse conditions, this species proved to be an obligate short-day plant. Flowering was delayed when photoperiods longer than 8 h were given, the critical photoperiod being between 12 and 13 h. When both temperature and photoperiod were controlled, at 20°C a longer protoperiod (by almost 1 h) is still inductive compared to 25 and 30°C. The number of short-day cycles required for full induction is relatively high and dependent upon temperature; at 20°C or above, 10 cycles are adequate, but at 15°C, more short-day cycles are needed. The number of inflorescences formed as well as the floral index vary according to daylength × temperature × inductive cycle number, allowing flowering to be assessed quantitatively. Long days are inhibitory to flowering, either suppressing it completely (when symmetrically intercalated among 24 inductive cycles) or preventing the floral index from increasing.  相似文献

Leaf reflectance and fluorescence characteristics of soybean (Glycine max cv Bragg) are influenced strongly by Mn availability. This report evaluates the effects of leaflet choice, leaf age, and leaf nodal position on several spectral characteristics. Leaves were obtained from soybeans grown hydroponically under controlled environmental conditions with wide differences in Mn supply. The ratio of constant yield fluorescence (Fo) to variable yield fluorescence (Fv), the ratios of reflectance at 750 nm to 550 nm and that at 650 nm to 550 nm, the position of the "red edge" near 700 nm, and an index of leaf "yellowness" were measured periodically. Increasing leaf age caused increases in the "red edge" and in both reflectance ratios. Leaf "yellowness" and the fluorescence ratio Fo/Fv decreased with leaf age and increased with leaf nodal position, primarily in Mn deficient leaves. Effects arising from leaf choice were smaller than those caused by Mn deficiency.  相似文献

This study treated the isolation and passage of muscle-derived stem cells (MDSCs) from rat penile corpora cavernosa, detection of stem cell marker expression, observation of their self-renewal and continuous proliferation, and demonstration of their potential to differentiate into smooth muscle cells in co-culture. Muscle-derived stem cells from the rat penile corpora cavernosa were isolated and purified. The expression of stem cell markers Sca-1 and desmin was detected in PP6 cells, thus confirming that the main components of PP6 cells are MDSCs. The expression of Sca-1 and desmin occurred both in PP6 cells and cells at passages 3, 6, and 8, and there was no significant decrease in the expression level with increasing passage number. The growth curves indicated that the cell doubling time was approximately 48 h. The cells entered the stationary phase after approximately 7 days of culture. The proliferative activity of the cells at passage 8 remained unchanged. After 2 days of co-culture with smooth muscle cells, the DAPI-labeled MDSCs tended to exhibit smooth muscle cell morphology and expression of α-SMA was detected. MDSCs exist in the rat penile corpora cavernosa and possess the potential to differentiate into smooth muscle cells. This discovery serves as the basis in view of the potential use of endogenous stem cells for the treatment of erectile dysfunction (ED).  相似文献

Natural clones and clones from laboratory crossings of Daphnia pulex have been tested in their response to stimuli for male and ephippia formation. The clones are from ponds in Illinois, USA, an area with both obligate and cyclical clones of D. pulex. The progeny from two types of crosses were studied: 1) crosses between an obligate clone producing males and a cyclical clone producing only females, and 2) self-fertilization within a cyclical clone producing both sexes. Both the natural and the artificial clones showed great variation in response to environmental stimuli for males. Ephippia response seems more equal among the clones, but two of the artificial clones could hardly be stimulated to produce ephippia in the induction experiments. In crowded cultures, however, they produced a few. Crossings between cyclicals and obligates yielded mostly cyclical progeny. Progeny from the selfed clone had low survival and fecundity.  相似文献

Plant tissue culture technology offers a solution for meeting the increasing commercial demand on economically important plants such as rice, a widespread dietary staple. However, significant genotype-specific morphogenetic responses constitute a considerable on rice regeneration in plant biotechnology contexts. Aside from genotype dependency, the components of the nutrient media including gelling agents have an important impact on regeneration efficiency. The current study explores the effect of different gelling agents on various stages of rice regeneration in two Egyptian rice cultivars-Sakha104 and Giza178. Media solidified with varying concentrations of a variety of gelling agents (agar, bacto agar, gelrite and phytagel) were tested for their impact on the frequency of callus induction, shoot regeneration and rooting. The results indicated gellan gum (gelrite and phytagel) was superior to agar products (agar and bacto agar) for callus induction. By contrast, no significant differences were found between different gelling agents for shoot regeneration. Gellan gum and media solidified with bacto agar were found to lead to significantly higher root regeneration than agar. The Sakha104 cultivar showed better responses than Giza 178 for callus induction and similar performance to the Giza 178 cultivar for root regeneration irrespective of the gelling agent. This work provides insights into the impact of different gelling agents on the morphogenetic response of two rice cultivars and can be used to help maximize the frequency of rice regeneration.  相似文献

对烟草(Nicotiana tabacum L.)未授粉子房胚状体的诱导进行了研究,结果发现,胚状体是单细胞起源,起源于大孢子或卵细胞,接种发育早期的子房,胚状体起源于大孢子;接种发育晚期的子房,胚状体起源于卵细胞;接种发育中期的子种,胚状多起源于大孢子,少数起源于卵细胞,不同的基因型,蔗糖浓度及光照强度等对胚状体诱导率的影响亦有不同。  相似文献
