Amyloid‐like peptides are an ideal model for the mechanistic study of amyloidosis, which may lead to many human diseases, such as Alzheimer disease. This study reports a strong second harmonic generation (SHG) effect of amyloid‐like peptides, having a signal equivalent to or even higher than those of endogenous collagen fibers. Several amyloid‐like peptides (both synthetic and natural) were examined under SHG microscopy and shown they are SHG‐active. These peptides can also be observed inside cells (in vitro). This interesting property can make these amyloid‐like peptides second harmonic probes for bioimaging applications. Furthermore, SHG microscopy can provide a simple and label‐free approach to detect amyloidosis. Lattice corneal dystrophy was chosen as a model disease of amyloidosis. Morphological difference between normal and diseased human corneal biopsy samples can be easily recognized, proving that SHG can be a useful tool for disease diagnosis. 相似文献

The properties of the system maintaining the upright posture were compared in different states of the oculomotor system: during target fixation and horizontal fast and slow pursuit (0.1 and 0.01 Hz), recording the trajectories of the center of pressure in the frontal and the sagittal planes. Methods of nonlinear analysis were applied to assess the similarity in pairwise comparisons. The overall similarity of the frontal plane dynamics proved to be higher than that of the sagittal plane dynamics. However, differences were revealed in fast pursuit versus slow pursuit or fixation in the frontal but not in the sagittal plane. Such differences may reflect the different inertia of the oculomotor and the balance control systems. In general, the results are consistent with the current notions on the two orthogonal subsystems of postural control. 相似文献

Thioredoxins (TRXs) are distributed ubiquitously in prokaryotic and eukaryotic organisms. Plants have the most complex forms
of TRXs. The functional roles of such TRXs have been studied in abiotic stress but their roles in plant defense responses
against biotic stresses have been less well studied. Here, we identified an h-type TRX gene from pepper, CaTRXh1, and characterized its possible effect on Type II nonhost resistance, which entails localized programmed cell death in response
to nonhost pathogens. Peptide sequences of CaTRXh1 showed a high degree of similarity with TRXhs from tobacco and Arabidopsis thaliana. Southern blot analyses revealed that CaTRXh1 was present as a single copy in the pepper genome. Intriguingly, leaf infiltration by Xanthomonas axonopodis pv. glycines 8ra, eliciting a visible type II nonhost hypersensitive response (HR), and its type III secretion-system null mutant 8–13,
eliciting a type I nonhost non-HR, both induced CaTRXh1 at a level similar to that of pathogenesis-related protein 4, an HR marker gene in pepper. More surprisingly, expression
of CaTRXh1 was significantly increased when X. axonopodis pv. vesicatoria race 3 infiltrated the leaf of a pepper cultivar containing a resistance gene, but not with infiltration of a susceptible
pepper cultivar. Taken together, our study suggests that the expression of CaTRXh1 has a critical role in HR-mediated active defense responses in pepper.

Electrophysiological and behavioral responses of the wingless tea aphid, Toxoptera aurantii (Boyer), to 14 synthetic volatiles identified from tea shoots, their partial (GLV mixture) and full (ACB mixture) blends, and fresh young tea leaves, buds, tender stems, adult tea leaves and tea aphid-damaged young leaves (ADYL) were studied by using an electroantennography (EAG) and a four-arm olfactometer. ACB elicited the largest EAG responses. Major volatile components, Z-3-hexen-1-ol, E-2-hexenal, n-hexanol, methyl salicylate and benzylalcohol, from the tea shoots were strongly EAG active. All the 4 tested tea shoot tissues also elicited significant EAG responses, with the young tea leaves being the strongest, followed by buds, tender stems and adult tea leaves. Surprisingly, ADYL elicited a weakly negative EAG response. In the olfactory assays, the fresh and tender tea leaves, as well as the individual major volatile components, e.g. Z-3-hexenyl acetate, methyl salicylate, E-2-hexen-1-ol and Z-3-hexen-1-ol, from the tender shoots (EAG-active) were all attractive. This result might indicate that the wingless tea aphids may use tea shoot volatiles as kairomone to find their optimal feeding sites, e.g. fresh tender tea shoots. 相似文献

Despite the ubiquity of nonlinear functional relationships in nature we tend to characterize mechanisms in science using more tractable linear functions. In demographic modeling, transfer function analysis is used to calculate the nonlinear response of population growth rate to a theoretical perturbation of one or more matrix elements. This elegant approach is not yet popular in ecology. Inconveniently, using transfer function without care can produce erroneous results without warning. We used a large matrix projection model database to explore the potential pitfalls to be avoided in using transfer function analysis. We asked a fundamental population control question, what matrix element perturbation would be needed to reach a minimum goal of replacement population growth? We then tracked instances in which transfer function yields erroneous output and explored these cases in detail to measure how frequently it occurs. We developed a phylogenetically-corrected mixed effects logistic regression model in a Bayesian framework to test the effect of species traits and the identity of matrix elements on the probability that transfer function yields errors. We found in 16% of cases the transfer function yielded erroneous outcomes. These errors were more likely when perturbing demographic stasis and also for shrubs more than any other life form. Errors in transfer function analysis were often due to perturbing matrix elements beyond their biological limits, even when this is still mathematically correct. To use transfer function analysis properly in demographic modeling and avoid erroneous results, input must be carefully selected to include only a biologically admissible set of perturbations. 相似文献

Numerous ring-width chronologies from different species have recently been developed in diverse tropical forests across South America. However, the temporal and spatial climate signals in these tropical chronologies is less well known. In this work, annual growth rings of Amburana cearensis, a widely distributed tropical tree species, were employed to estimate temporal and spatial patterns of climate variability in the transition from the dry Chiquitano (16–17°S) to the humid Guarayos-southern Amazon (14–15°S) forests. Four well-replicated chronologies (16–21 trees, 22–28 radii) of A. cearensis were compared with temperature and precipitation records available in the region. The interannual variations in all four A. cearensis tree-ring chronologies are positively correlated with precipitation and negatively with temperature during the late dry-early wet season, the classic moisture response seen widely in trees from dry tropical and temperate forests worldwide. However, the chronologies from the dry Chiquitano forests of southern Bolivia reflect the regional reduction in precipitation during recent decades, while the chronologies from the tropical lowland moist forests in the north capture the recent increase in precipitation in the southern Amazon basin. These results indicate that A. cearensis tree growth is not only sensitive to the moisture balance of the growing season, it can also record subtle differences in regional precipitation trends across the dry to humid forest transition. Comparisons with previously developed Centrolobium microchaete chronologies in the region reveal a substantial common signal between chronologies in similar environments, suggesting that regional differences in climate are a major drivers of tree growth along the precipitation gradient. The difficulty of finding A. cearensis trees over 150-years old is the main limitation involved in the paleoclimate application of this species. The expansion of monocultures and intensive cattle ranching in the South American tropics are contributing to the loss of these old growth A. cearensis trees and the valuable records of climate variability and climate change that they contain. 相似文献

Antibiotics regulate various physiological functions in cyanobacteria and may interfere with the control of cyanobacterial blooms during the application of algaecides. In this study, Microcystis aeruginosa was exposed to H2O2 and glyphosate for 7 d in the presence of coexisting mixed antibiotics (amoxicillin, spiramycin, tetracycline, ciprofloxacin, and sulfamethoxazole) at an environmentally relevant concentration of 100 ng · L?1. The mixed antibiotics significantly (P < 0.05) alleviated the growth inhibition effect of 15–45 μM H2O2 and 40–60 mg · L?1 glyphosate. According to the increased contents of chlorophyll a and protein, decreased content of malondialdehyde, and decreased activities of superoxide dismutase and glutathione S‐transferase, antibiotics may reduce the toxicity of the two algaecides through the stimulation of photosynthesis and the reduction in oxidative stress. The presence of coexisting antibiotics stimulated the production and release of microcystins in the M. aeruginosa exposed to low concentrations of algaecides and posed an increased threat to aquatic environments. To eliminate the secondary pollution caused by microcystins, high algaecide doses that are ≥45 μM for H2O2 and ≥60 mg · L?1 for glyphosate are recommended. This study provides insights into the ecological hazards of antibiotic contaminants and the best management practices for cyanobacterial removal under combined antibiotic pollution conditions. 相似文献
