Androgenic compounds induce an interaction between the NH2- and COOH-terminal regions (N–C interaction) of androgen receptor (AR). We describe a rapid yeast bioassay for androgenic and anti-androgenic compounds based on androgen-dependent β-catenin-enhanced N–C interaction. The bioassay was also effective at detecting compounds that inhibit the N–C interaction in ways that do not involve binding to the ligand-binding domain. 相似文献

A recombinant strain of Salmonella enterica serovar Typhimurium (mutant in propionate-activation activity) was metabolically engineered to control the composition of poly(3-hydroxybutyrate-co-3-hydroxy- valerate) (PHBV), a polyhydroxyalkanoate copolymer with commercially desirable properties. A gene (prpE) encoding propionyl-CoA synthetase was placed under the control of the IPTG-inducible taclacUV5 promoter (P(taclacUV5)) while the polyhydroxyalkanoate synthesis operon (phaBCA) from Acinetobacter sp. RA3849 was coexpressed under the control of the arabinose-inducible araBAD promoter (P(BAD)). S. enterica, harboring both constructs, was grown in medium containing a fixed substrate concentration and the composition of the copolymer was varied between 2 mol% and 25 mol% 3-hydroxyvalerate by controlling the IPTG level in the medium. This \"dial-a-composition\" system should find application in cases where the substrate concentration of a feedstream for PHBV bioplastic production is not adjustable. 相似文献

Leaks and isotopic disequilibria represent potential errors and artefacts during combined measurements of gas exchange and carbon isotope discrimination (Δ). This paper presents new protocols to quantify, minimize, and correct such phenomena. We performed experiments with gradients of CO2 concentration (up to ±250 μmol mol?1) and δ13CCO2 (34‰), between a clamp‐on leaf cuvette (LI‐6400) and surrounding air, to assess (1) leak coefficients for CO2, 12CO2, and 13CO2 with the empty cuvette and with intact leaves of Holcus lanatus (C3) or Sorghum bicolor (C4) in the cuvette; and (2) isotopic disequilibria between net photosynthesis and dark respiration in light. Leak coefficients were virtually identical for 12CO2 and 13CO2, but ~8 times higher with leaves in the cuvette. Leaks generated errors on Δ up to 6‰ for H. lanatus and 2‰ for S. bicolor in full light; isotopic disequilibria produced similar variation of Δ. Leak errors in Δ in darkness were much larger due to small biological : leak flux ratios. Leak artefacts were fully corrected with leak coefficients determined on the same leaves as Δ measurements. Analysis of isotopic disequilibria enabled partitioning of net photosynthesis and dark respiration, and indicated inhibitions of dark respiration in full light (H. lanatus: 14%, S. bicolor: 58%). 相似文献

A general method for constructing low-rank tensor product smooths for use as components of generalized additive models or generalized additive mixed models is presented. A penalized regression approach is adopted in which tensor product smooths of several variables are constructed from smooths of each variable separately, these \"marginal\" smooths being represented using a low-rank basis with an associated quadratic wiggliness penalty. The smooths offer several advantages: (i) they have one wiggliness penalty per covariate and are hence invariant to linear rescaling of covariates, making them useful when there is no \"natural\" way to scale covariates relative to each other; (ii) they have a useful tuneable range of smoothness, unlike single-penalty tensor product smooths that are scale invariant; (iii) the relatively low rank of the smooths means that they are computationally efficient; (iv) the penalties on the smooths are easily interpretable in terms of function shape; (v) the smooths can be generated completely automatically from any marginal smoothing bases and associated quadratic penalties, giving the modeler considerable flexibility to choose the basis penalty combination most appropriate to each modeling task; and (vi) the smooths can easily be written as components of a standard linear or generalized linear mixed model, allowing them to be used as components of the rich family of such models implemented in standard software, and to take advantage of the efficient and stable computational methods that have been developed for such models. Millions of dollars of limited state cleanup funds are spent each year in New Hampshire to identify, sample, excavate, and treat thousands of tons of contaminated soil. Cost analyses of numerous sites indicated that soil remediation costs alone reach upwards of $300,000.00 per site. The New Hampshire Department of Environmental Services “Interim Policy for Management of Soils Contaminated from Spills/Releases of Virgin Petroleum Products”; (DES, 1989, 1991) set conservative remediation goals based on total petroleum hydrocarbons in 1989 using the Leaching Potential Analysis method (California Luft Manual, 1989). A current review of available literature and several case histories indicated that chemical‐specific soil cleanup levels may be more appropriate for establishing remedial goals. New chemical‐specific soil cleanup guidelines using a risk‐based approach have been developed. These new guidelines are conservatively based using two principal considerations: (1) an assumed soil exposure scenario that estimated the human health risks associated with potential long‐term exposure to site soils via ingestion, inhalation and dermal contact and (2) the estimated fate and transport of chemicals of concern in the soil unsaturated zone. The first consideration assumed a total cancer risk that did not exceed 1 × 10‐6. The second consideration employed the use of the SEasonal SOIL Compartment (SESOIL) model which simultaneously models water transport, sediment transport, and pollutant fate (US EPA, 1981). Several state soil standards from Oregon, Wisconsin, Massachusetts, and other states were extensively reviewed in order to develop a level of confidence that use of the SESOIL model was appropriate. A series of “sensitivity”; analyses was also performed in order to evaluate the response of the model to changes in various input parameters unique to New Hampshire's hydrogeologic conditions. Generic soil cleanup guidelines were developed for 24 petroleum‐based volatile and semivolatile chemicals of concern to be applied statewide. Site‐specific soil cleanup guidelines will be allowed if it can be demonstrated that insertion of site‐specific data into the model will not adversely affect groundwater quality. As a result of the above processes, timely and much more cost‐effective remediation will be achieved while still maintaining a high degree of protection of the groundwater quality and human health. 相似文献

The colonization of Aedes aegypti and Anopheles dirus was performed using out‐of‐date human blood from a blood bank as a nutritional supply dispensed from a common artificial feeder. Preserved human blood was collected and used for feeding on days 5, 15, and 25 after date of expiration and dispensed from a common artificial feeder to rear the mosquitoes. Ae. aegypti had a feeding rate of 78.7, 62, and 18% at the respective intervals while An. dirus had a rate of 80, 56.8, and 7.3% on the same respective days. Direct feeding on live hamsters resulted in a rate of 96 and 90% for Ae. aegypti and An. dirus, respectively. Although egg production rates decreased from the day 5 feeding to the day 25 feeding, all of the developmental stages resulting from An. dirus fed at day 5 and 15 showed insignificant differences when compared with direct feeding on the blood of a hamster. 相似文献

Redox changes within neurones are increasingly being implicated as an important causative agent in brain ageing and neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS), Parkinson's disease (PD) and Alzheimer's disease (AD). Cells have developed a number of defensive mechanisms to maintain intracellular redox homeostasis, including the glutathione (GSH) system and antioxidant enzymes. Here we examine the effects of N-acetyl-L-cysteine (NAC) on beta-amyloid (A beta) secretion and tau phosphorylation in SHSY5Y neuroblastoma cells after exposure to oxidative stress inducing/cytotoxic compounds (H(2)O(2), UV light and toxic A beta peptides). A beta and tau protein are hallmark molecules in the pathology of AD while the stress factors are implicated in the aetiology of AD. The results show that H(2)O(2), UV light, A beta 1-42 and toxic A beta 25-35, but not the inactive A beta 35-25, produce a significant induction of oxidative stress and cell cytotoxicity. The effects are reversed when cells are pre-treated with 30 mM NAC. Cells exposed to H(2)O(2), UV light and A beta 25-35, but not A beta 35-25, secrete significantly higher amounts of A beta 1-40 and A beta 1-42 into the culture medium. NAC pre-treatment increased the release of A beta 1-40 compared with controls and potentiated the release of both A beta 1-40 and A beta 1-42 in A beta 25-35-treated cells. Tau phosphorylation was markedly reduced by H(2)O(2) and UV light but increased by A beta 25-35. NAC strongly lowered phospho-tau levels in the presence or absence of stress treatment. 相似文献

Poly(sarcosine) displayed on polymeric micelle is reported to trigger a T cell‐independent type2 reaction with B1a cells in the mice to produce IgM and IgG3 antibodies. In addition to polymeric micelle, three kinds of vesicles displaying poly(sarcosine) on surface were prepared here to evaluate the amounts and avidities of IgM and IgG3, which were produced in mice, to correlate them with physical properties of the molecular assemblies. The largest amount of IgM was produced after twice administrations of a polymeric micelle of 35 nm diameter ( G1 ). On the other hand, the production amount of IgG3 became the largest after twice administrations of G3 (vesicle of 229 nm diameter) or G4 (vesicle of 85 nm diameter). The augmented avidity of IgG3 after the twice administrations compared with that at the single administration was the highest with G3 . These differences in immune responses are discussed in terms of surface density of poly(sarcosine) chains, nanoparticle size, hydrophobic component of poly(L‐lactic acid) or (Leu‐ or Val‐Aib)n, and membrane elasticity of the nanoparticles.
