A theoretical model dealing with endocytosis, exocytosis and caveolae invagination, describing plasmalemma homeostasis during cell growth and division, is proposed. It considers transmembrane pressure, membrane tension and mechanosensitivity of membrane processes. Membrane hydraulic conductivity and the flux of transmembrane nonvesicular transport are taken into account. The developed mathematical analysis operates with a formulated set of constitutive equations describing the mechanical state and kinetics of changes in an open dynamic membrane system. The standard version of a model with adjusted parameters was implemented, and predictions including a discussion on the effect of possible parameter modifications were presented. Computer simulations indicate big changes in the magnitude of membrane tension and elasticity, and in the number of membrane buddings in young cells and during mitosis. They also show the extent of cell growth inhibition resulting from a decrease in transmembrane transport or an increase in the exerted difference in osmotic pressure. Moreover, the simulations reveal that exocytosis regulated during mitosis may not be as important for cell growth, as sometimes presumed. Many patients suffer from chronic gastrointestinal diseases characterized by chronic inflammation, increased intestinal permeability and visceral pain in which there is no definitive treatment. Adult stem cells have recently been used in various disease states to contribute wound-healing processes. In the current study we investigated the ability of intra-colonic adult stem cells application to heal colonic inflammation in IL-10(-/-) mice with active colitis. The aims of this study were to determine whether intra-colonic infusion of adult colonic stem cells (CSCs) (local stem cell transplantation): (i) restores intestinal permeability; (ii) attenuates visceral hypersensitivity; (iii) heals murine colitis. IL-10(-/-) mice with active colitis were transplanted with adult stem cells. Mice received either a single intracolonic infusion of CSCs or colonic epithelial cells. Two weeks after transplantation, we measured visceral hypersensitivity and intestinal permeability and correlated these with histological improvement of colitis. IL-10(-/-) mice that received stem cell transplantation showed histopathologic evidence of recovery from colitis. Improvement in colitis as graded by pathology scores correlated with restoration of intestinal permeability and decreased visceral hypersensitivity. Intra-colonic administration of CSCs is a potential therapeutic method for treating refractory symptoms in patients with chronic gastrointestinal diseases associated with chronic inflammation and visceral hypersensitivity. This method may be safer and should have far fewer side effects than systemic stem cell administration. 相似文献

Computational studies of the potential biological impact of several energetic compounds were performed. The most commonly used explosives were considered in the present studies: trinitrotoluene (TNT), 2,4-dinitrotoluene (2,4-DNT), 2,4-dinitroanisole (DNAN), and 5-Nitro-2,4-dihydro-3H-1,2,4-triazol-3-one (NTO). The effect of such factors as ionic strength and presence of DMSO in the water solution on the structure of the membrane were considered using the POPC lipid bilayer as an example. Molecular dynamics (MD) simulations revealed that, even on a short-time scale, the influence of those additives is noticeable, and therefore those factors should always be taken into account. The MD and the COSMOmic approaches were used to elucidate the ability of the energetic compounds to penetrate the living cell. Calculated free energy profiles and partitioning coefficients revealed distributions of the compounds in the lipid bilayer as well as an overall ability to enter the cell. MD in this case provides a better representation of the free energy profile, while the COSMOmic approach works better to predict log(Klipw) values. The effect of the functional group was observed for the profiles that were obtained using the MD method. 相似文献

The structures of alkyl radicals generated in several methyl esters of fatty acids by irradiation with UV light were studied by the spin trapping technique. A spin trap, deuterated nitrosodurene, traps alkyl radicals in both saturated and unsaturated esters at the ambient temperature. The trapped radicals and their hyperfine splitting constants from several esters were as follows: pentadienyl radicals (aN= 13.8 ~ 14.0 G, aH = 5.9 ~ 6.0 G) from methyl linoleate, linolenate and docosahexaenoate; allyl radicals (aN = 13.9 G, aH = 6.8 G) and α-carbon radicals (aN = 13.3 G, aH = 10.0 G) from methyl oleate and elaidate; α-carbon radicals (aN = 13.3 ~ 13.4 G, aH = 9.6 ~ 10.0 G) and secondary alkyl radicals (aN = 13.9 G, aH = 6.8 ~ 7.2 G) from saturated esters. 相似文献

以山东密刺黄瓜 (Cucumis sativus L.)为试材 ,分别采用大棚土壤和露地土壤进行盆栽模拟试验 ,首次研究了不同浓度的苯丙烯酸对不同土壤栽培条件下黄瓜幼苗膜质过氧化作用的化感效应。结果表明 :不同浓度的苯丙烯酸对大棚土培幼苗不同时期的 SOD活性的化感效应均为抑制作用 ,并随着处理浓度的增加抑制作用增强。浓度大于 5 0 mg/kg时 ,对露地土培幼苗 6叶期 SOD活性的化感效应均为抑制作用。浓度大于 1 0 0 mg/kg时 ,对两种土培幼苗 CAT的化感效应均为抑制作用 ,并随着处理浓度的增加抑制作用增强。苯丙稀酸对不同土壤、不同时期黄瓜幼苗的 MDA均为促进作用 ,并随着处理浓度的增加促进作用增强。苯丙稀酸使黄瓜幼苗的电解质外渗率和伤害率增加 ,浓度大于 5 0 m g/kg时的大棚土培幼苗和浓度大于 1 0 0 m g/kg时的露地土培幼苗 ,电解质外渗率与对照及处理间均存在显著差异 (p <0 .0 5 ) ,伤害率与对照及处理间均存在极显著差异 (p <0 .0 1 )。大棚土培幼苗 MDA含量和电解质外渗率的化感强度均大于露地土培幼苗 相似文献

The existence of an extracellular signaling pathway that mediates nodule formation, a cell-mediated immune response, has been reported in Bombyx mori larvae. In this pathway, C-type lectins and the hemolymph serine proteinase BmHP-8 function in pathogen associated molecular pattern (PAMPs) recognition and signaling transduction. However, which molecule elicits the cellular response at the end of the pathway is unknown. Background
The epidermis forms a critical barrier that is maintained by orchestrated programs of proliferation, differentiation, and cell death. Gene mutations that disturb this turnover process may cause skin diseases. Human GASDERMIN A (GSDMA) is frequently silenced in gastric cancer cell lines and its overexpression has been reported to induce apoptosis. GSDMA has also been linked with airway hyperresponsiveness in genetic association studies. The function of GSDMA in the skin was deduced by dominant mutations in mouse gasdermin A3 (Gsdma3), which caused skin inflammation and hair loss. However, the mechanism for the autosomal dominance of Gsdma3 mutations and the mode of Gsdma3’s action remain unanswered.Results
We demonstrated a novel function of Gsdma3 in modulating mitochondrial oxidative stress. We showed that Gsdma3 is regulated by intramolecular fold-back inhibition, which is disrupted by dominant mutations in the C-terminal domain. The unmasked N-terminal domain of Gsdma3 associates with Hsp90 and is delivered to mitochondrial via mitochondrial importer receptor Tom70, where it interacts with the mitochondrial chaperone Trap1 and causes increased production of mitochondrial reactive oxygen species (ROS), dissipation of mitochondrial membrane potential, and mitochondrial permeability transition (MPT). Overexpression of the C-terminal domain of Gsdma3 as well as pharmacological interventions of mitochondrial translocation, ROS production, and MPT pore opening alleviate the cell death induced by Gsdma3 mutants.Conclusions
Our results indicate that the genetic mutations in the C-terminal domain of Gsdma3 are gain-of-function mutations which unmask the N-terminal functional domain of Gsdma3. Gsdma3 regulates mitochondrial oxidative stress through mitochondrial targeting. Since mitochondrial ROS has been shown to promote epidermal differentiation, we hypothesize that Gsdma3 regulates context-dependent response of keratinocytes to differentiation and cell death signals by impinging on mitochondria.Electronic supplementary material

Summary The effects of diltiazem, a drug which inhibits the calcium channels in cardiac muscle as well as the light-sensitive channels in photoreceptor cells, were studied on ionic fluxes in both membrane and intact cell preparations. Diltiazem nonselectively increased the ionic permeability to both anions and cations in photoreceptor rod outer segment and synaptic membrane vesicles as well as in intact erythrocytes. Under our conditions, the estimated threshold for the diltiazem effect varied between 12.5 and 200 m. In each case the concentration dependence exhibited the sigmoidal shape characteristic of positive cooperativity. The effect of diltiazem on ionic fluxes from phospholipid vesicles were strongly influenced by phospholipid composition and membrane charge. By contrast, diltiazem inhibited the efflux of86Rb from photoreceptor cells of intact aspartate-isolated retina, an effect opposite to that of diltiazem on ionic permeabilities in photoreceptor membrane vesicle preparations.These data raise serious doubts on the specificity of diltiazem as a calcium channel blocker or as a cGMP channel blocker when used at concentrations higher than 10 m. 相似文献

The use of bimolecular lipid membranes (BLM) as model membrane allows the analysis of the transport of mercury compounds across the lipidic barriers of biological membranes. The results of flux measurements show that two mercury compounds--HgCl2 and CH3HgCl--cross the BLM but the overall permeabilities are dependent on the pH of the aqueous media, and are not apparently influenced by the different phospholipid constituents of the bilayers. On the other hand, electrical measurements show that, function of the chemical speciation, the transport of this metal is done essentially in the neutral form. 相似文献

中国南方扬子地台震旦系陡山沱组产出丰富的微体化石,它们主要保存在磷块岩以及燧石结核和条带中。文章详细报道湖北峡东地区九龙湾剖面震旦系陡山沱组微体化石,描述以前未曾在峡东地区碳酸盐相燧石中发现的8属8种微体化石。研究九龙湾剖面陡山沱组大型带刺疑源类、微体多细胞藻类和动物胚胎化石的分布特征,发现Tianzhushania spinosa是最早出现的大型带刺疑源类分子,大冰期后微体生物的辐射是一个阶段性渐进的过程。同时.本项研究进一步证实华南扬子区陡山沱组碳酸盐相燧石和磷块岩地层中保存的微体化石面貌基本一致。 相似文献
