The culture filtrate proteins (CFPs) from Mycobacterium tuberculosis have been shown to induce protective immune responses in human and animal models, making them a promising source of candidate targets for tuberculosis drugs, vaccines, and diagnostics. The constituents of the M. tuberculosis CFP proteome are complex and vary with growth conditions. To effectively profile CFPs, gel‐based prefractionation is usually performed before MS analysis. In this study, we describe a novel prefractionation approach by which the proteome is divided into seven partially overlapping fractions by biomimetic affinity chromatography (BiAC) using a six‐column cascade. The LC‐MS/MS analysis of individual fractions identified a total of 541 CFPs, including 61 first‐time identifications. Notably, ~1/3 (20/61) of these novel CFPs are membrane proteins, among which nine proteins have 2–14 transmembrane domains. In addition, ~1/4 (14/61) of the CFPs are basic proteins with pI values greater than 9.0. Our data demonstrate that biomimetic affinity chromatography prefractionation markedly improves protein detection by LC‐MS/MS, and the coverage of basic and hydrophobic proteins in particular is remarkably increased.  相似文献

We present a simple and effective algorithm RDOCK for refining unbound predictions generated by a rigid-body docking algorithm ZDOCK, which has been developed earlier by our group. The main component of RDOCK is a three-stage energy minimization scheme, followed by the evaluation of electrostatic and desolvation energies. Ionic side chains are kept neutral in the first two stages of minimization, and reverted to their full charge states in the last stage of brief minimization. Without side chain conformational search or filtering/clustering of resulting structures, RDOCK represents the simplest approach toward refining unbound docking predictions. Despite its simplicity, RDOCK makes substantial improvement upon the top predictions by ZDOCK with all three scoring functions and the improvement is observed across all three categories of test cases in a large benchmark of 49 non-redundant unbound test cases. RDOCK makes the most powerful combination with ZDOCK2.1, which uses pairwise shape complementarity as the scoring function. Collectively, they rank a near-native structure as the number-one prediction for 18 test cases (37% of the benchmark), and within the top 4 predictions for 24 test cases (49% of the benchmark). To various degrees, funnel-like energy landscapes are observed for these 24 test cases. To the best of our knowledge, this is the first report of binding funnels starting from global searches for a broad range of test cases. These results are particularly exciting, given that we have not used any biological information that is specific to individual test cases and the whole process is entirely automated. Among three categories of test cases, the best results are seen for enzyme/inhibitor, with a near-native structure ranked as the number-one prediction for 48% test cases, and within the top 10 predictions for 78% test cases. Summary From two lines of Medicago sativa characterized by a high regeneration capability, calli resistant to culture filtrate of Fusarium oxysporum f. sp. medicaginis have been selected. In these calli regeneration capability was greatly reduced and only one plant per callus was recovered. Regenerated plants have been evaluated for resistance to culture filtrate and for in vivo resistance to the pathogen. Three plants out of eight were resistant to the fungus and a high correlation between resistance to culture filtrate and in vivo resistance was observed.Research work supported by C.N.R., Italy. Effective protection against Mycobacterium tuberculosis may be achieved in experimental animals by immunization with proteins secreted by tuberculous bacilli in the extracellular milieu during growth. In this study, monoclonal antibodies were raised against Mycobacterium bovis bacillus Calmette-Guérin (BCG) culture filtrate proteins or live BCG, in an attempt to identify novel mycobacterial secretion antigens: the localization of the antigens recognized by the monoclonal antibodies within the mycobacterial cell was studied and interspecies reactivity was also investigated. The monoclonal antibodies obtained recognized proteins of molecular mass ranging from 5 to 82 kDa, with a prevailing frequency in the 30 kDa region. Three of the monoclonal antibodies recognized proteins present only in culture filtrates, one reacted with a cytoplasmic antigen, while the remaining antibodies recognized components which were mainly associated with the cell wall and the cytoplasmic membrane. The chemical nature and possible identity of the antigens was checked. Three monoclonal antibodies are likely to react with novel mycobacterial antigens of 5, 42 and 82 kDa, respectively.  相似文献

【背景】马铃薯干腐病是一种由镰刀菌引起的田间和储藏期都普遍发生的病害,主要引起块茎腐烂,致使马铃薯品质和产量降低,严重影响其食用价值和经济价值。【目的】发掘有效的生防菌株以控制马铃薯干腐病,并探究其抑菌作用。【方法】从甘肃定西地区马铃薯根际土壤中分离到109株细菌,以硫色镰刀菌(Fusarium sulphureum)为靶标菌,采用平板对峙法筛选拮抗菌,并通过形态学、生理生化特征及16S r RNA基因序列分析对拮抗菌株进行鉴定。检测拮抗菌无菌发酵液对F.sulphureum菌丝生长、孢子萌发、马铃薯块茎损伤接种病斑直径、干腐病发病率及对绿豆种子发芽的影响。【结果】筛选到一株对马铃薯干腐病有较强抑制作用的菌株YL11,经鉴定其为假单胞菌属(Pseudomonas sp.)菌株。YL11菌株无菌发酵液对F.sulphureum菌丝生长、孢子萌发、马铃薯块茎病斑扩展、干腐病发病率、毒素活性均有显著抑制作用。20%无菌发酵液对F.sulphureum菌落生长的抑制率达到87.3%;75%无菌发酵液可完全抑制孢子萌发;无菌发酵液浸泡能有效抑制马铃薯干腐病病斑的扩展,14 d时对病斑扩展的抑制率达到67.1%;90 d后干腐病的发生率降低了68.4%;同时降低了F.sulphureum毒素的活性。【结论】拮抗菌株YL11能显著抑制F.sulphureum的生长,对马铃薯干腐病有较强的生物防治效果,具有潜在的应用价值。  相似文献

Mycobacterial polypeptides from 2 kDa to 14 kDa may be involved in host response to infection and be useful for new diagnostics and vaccines. Tris-tricine SDS-polyacrylamide gel electrophoresis separation of proteins in tuberculin purified protein derivative (PPD) and in 6-8-week culture filtrates of Mycobacterium tuberculosis, Mycobacterium bovis BCG, and other Mycobacterium species demonstrated as many as 10 low molecular mass bands. Common and distinct bands were observed among different species and PPD. These low molecular mass culture filtrate proteins may represent potential diagnostic reagents and vaccines for Mycobacterium tuberculosis.  相似文献

Seven genotypes of sunflower, including populations and hybrids, showing differential susceptibility to Alternaria leaf and stem blight were crossed to CMS 234A with pollen selection and without pollen selection. The pathogen culture filtrate was used as selective pressure at stylar tissue by applying it 1 h before pollination. Distilled water applied to stigmas and styles served as control. Two sets of seven hybrids, one set from selective and the other from non-selective fertilization, were evaluated for reaction to Alternaria leaf and stem blight during the rainy season under natural epiphytotic conditions. Selection for resistant pollen on the stigmatic surface resulted in a corresponding increase in progeny resistance. The study demonstrates the transmission of the selected trait from the pollen generation to progeny. Further, it was observed that the effect of pollen selection was high in the progenies of moderately resistant parents compared to progenies of highly susceptible parents. The effect of successive pollen selection was studied by backcrossing the progeny derived through selective fertilization to the fertile parent using selective fertilization. Successive pollen selection further improved disease resistance of progeny. However, the improvement was not very great. Hence, repeated cycles of selection are required to achieve a useful level of resistance in the case of sunflower, since resistance is polygenetically controlled. Summary The present investigation deals with the stabilization of rifamycin-B oxidase activity in the culture filtrate ofCurvularia lunata using various methods. It was found that rifamycin-B oxidase activity in the culture filtrate was stable up to 5 days at 5°C, degrading thereafter due to microbial contamination. The stabilization of enzyme activity was carried out by (i) concentration of culture filtrate and (ii) lyophilization, the activity remaining intact for 24 and 75 days, respectively. In another method, the enzyme activity was preserved by addition of 25–30% glycerol to the culture filtrate and the storability of enzyme activity then increased up to 90 days at 5°C. The conversion of rifamycin-B to rifamycin-S using the stabilized rifamycin-B oxidase and fresh culture filtrate were comparable when run under similar conditions. The recovery of rifamycin-S powder from these experiments was not affected in any way in the presence of glycerol. Therefore, the present method of preservation of rifamycin-B oxidase may find industrial application for commerical production of rifamycin-S, which is an important intermediate for the synthesis of an antituberculosis drug, rifamycin.  相似文献
