本试验以黄瓜和水稻幼苗为材料,研究了光照和黑暗条件下低温对植物叶绿素蛋白质复合体的影响。SDS—PAGE电泳结果表明:5℃及12h 280μmol m~(-2)S~(-1)处理2d,Chl-蛋白质复合体的降解明显大于5℃暗低温处理;低温与光照对P700-CPa_1的影响大于LHCP。叶绿素荧光测定表明;5℃及12h 280μmol m~(-2)s~(-1)的处理对PSⅡ的影响亦大于暗低温处理。由此认为:低温与光对植物叶绿体的PSⅠ和PSⅡ都有明显的影响,其机理可能与常温下高光强引起的光抑制相类似;不同的是低温下中等光强就能引起光抑制。因此,在光照低温下往往加剧植物冷害的发生。  相似文献

The effect of cell density (1–4.5 g L-1) and light intensity (44 and 82 mol m-2 s-1) on fatty acid composition andeicosapentaenoic acid (EPA, 20:5 3) production was studied ina semi-continuous culture of Monodus subterraneus grown in a helicaltubular photobioreactor (`Biocoil') under laboratory conditions. Under lowlight, the highest proportion of EPA (31.5% of total fatty acids) and EPAcontent (3.5% of dry weight), biomass productivity (1.3 g L-124 h-1) and EPA productivity (44 mg L-1 24 h-1)occurred at optimal cell density of about 1.7 g L-1. Cell densityhad no effect on the total fatty acid (TFA) content and was maintained atca. 11% of dry weight. Under high light, the highest proportion ofEPA to fatty acids (31.8%), the total fatty acids content (13.4%) andEPA content (4.3% of dry weight) occurred at cell density of about 3.4gL-1. But the highest biomass productivity (1.7 g L-124 h-1) and EPA productivity (56 mg L-1 24 h-1) wereobtained at a cell density of 1.6 and 2.6g L-1, respectively. Ourresults suggest that manipulating the cell density and light intensity canmodify the composition of fatty acid and production of eicosapentaenoicacid (EPA) in M. subterraneus.  相似文献

A comprehensive analysis of the quaternary features of distantly related homo‐oligomeric proteins is the focus of the current study. This study has been performed at the levels of quaternary state, symmetry, and quaternary structure. Quaternary state and quaternary structure refers to the number of subunits and spatial arrangements of subunits, respectively. Using a large dataset of available 3D structures of biologically relevant assemblies, we show that only 53% of the distantly related homo‐oligomeric proteins have the same quaternary state. Considering these homologous homo‐oligomers with the same quaternary state, conservation of quaternary structures is observed only in 38% of the pairs. In 36% of the pairs of distantly related homo‐oligomers with different quaternary states the larger assembly in a pair shows high structural similarity with the entire quaternary structure of the related protein with lower quaternary state and it is referred as “Russian doll effect.” The differences in quaternary state and structure have been suggested to contribute to the functional diversity. Detailed investigations show that even though the gross functions of many distantly related homo‐oligomers are the same, finer level differences in molecular functions are manifested by differences in quaternary states and structures. Comparison of structures of biological assemblies in distantly and closely related homo‐oligomeric proteins throughout the study differentiates the effects of sequence divergence on the quaternary structures and function. Knowledge inferred from this study can provide insights for improved protein structure classification and function prediction of homo‐oligomers. Light brown apple moth, Epiphyas postvittana (Walker), is a newly invasive pest in California. Habrobracon gelechiae Ashmead is an indigenous North American ectoparasitoid of lepidopterans. Using E. postvittana as the host, we determined H. gelechiae host stage preference for oviposition and suitability for larval growth: the parasitoid attacked second to fifth instars, but preferred older (third to fifth) host larvae for oviposition, laying more eggs per clutch on the largest (fifth instar). Offspring survival was better on larger (third to fifth instars) hosts. Adult survival and fecundity and immature temperature development were studied at three key temperatures. At 12°C, H. gelechiae failed to oviposit. At 22 and 30°C, H. gelechiae had, respectively, egg to adult developmental times of 15.4 and 8.7 days, adult female survival of 54.8 and 27.2 days, lifetime parasitism of 39.9 and 21.8 hosts and 181 and 151 eggs per female, an intrinsic rate of increase of 0.107 and 0.165, and mean generation time 30.7 and 18.9 days. We studied intraguild interactions with the endoparasitoid Meteorus ictericus Nees, and found that H. gelechiae did not discriminate against hosts that were parasitised by M. ictericus. Neither the paralysing venom injected by H. gelechiae or the presence of the ectoparasitoid's eggs or larvae arrested M. ictericus development. We also conducted a field release of H. gelechiae on two plant species commonly infested by E. postvittana and showed that H. gelechiae was able to locate and attack moth larvae on both hosts.  相似文献

Oleic acid metabolism can be considered to be an indicator of growth photoregulation in cowpea ( Vigna unguiculata Westphal cv. M53) epicotyls. Phosphatidylcholine and phosphatidylethanolamine are the two lipid classes concerned in the photoregulation of oleic acid accumulation. The incorporation of radioactive precursors in internodes of whole plants has shown that there is de novo synthesis of these phospholipids during the light dependent growth process.
The variations in oleic acid content were used to study the photocontrol of elongation in segments excised from the apical part of the epicotyl. In this system, as in whole plants, oleic acid was the only fatty acid showing significant variation related to the light/dark treatments. Differences in photoresponse between excised intenode segments and internodes in whole plants are discussed.  相似文献

The kinetics of agglomeration of proteins precipitating in a viscous solution was measured by light scattering. The resulting transitory maximum was linearly proportional to the mass of protein over three orders of magnitude. The change in scattering intensity is described as a change in scattering symmetry due to a continuous in particle size.This method is both fast (minutes) and sensitive (30 ng protein) and is independent of the chemical composition of the different protein species and is barely influenced by size and shape of the proteins.By solubilising the protein in an alkaline dedecyl sulfate solution this method can be applied to all types of biological samples (e.g. tissue homogenates proteins) and also to all types of biological preparations containing detergents as well as urea, sucrose, salts and lipids.  相似文献
