Bird A.F. and Stynes B.A. 1981. The life cycle of Anguina agrostis: Development in the host plant. Internationaljournal for Parasitology11: 431–440. The growth and development of the infective second stage “dauer” larvae (DL2) of Anguina agrostis into adults have been followed under field conditions in rye grass (Lolium rigidum). Three moults were observed to occur during the parasitic phase of development. From the third (second parasitic) moult onwards, there was much more variability in the size of the female nematodes than in the males and sexual dimorphism became very pronounced. The transition from the DL2 to the second stage parasitic larva (PL2) is marked by the disappearance of the numerous lipid storage granules which are characteristic of the DL2, and the development in the PL2 of an intestine which becomes more pronounced in each succeeding stage, particularly in the adult female. Anguina agrostis is unusual among parasitic nematodes in that the DL2 has the thickest cuticle of all stages, including adults. The L4 and adult males have thicker cuticles than the females at the same stages of development. Moulting appears to involve resorption of the innermost basal zone of the shed cuticle as well as morphological and chemical changes to the epicuticle.

Smales L. R. The life history of Labiostrongylus eugenii, a nematode parasite of the Kangaroo Island Wallaby (Macropus eugenii): development and hatching of the egg and the free living stages. International Journal for Parasitology7: 449–456. Labiostrongylus eugenii (Trichonematidae) occurs in the stomach of the Kangaroo Island Wallaby. Egg morphology is similar to that of other strongyloids. When incubated at 25°C embryogenesis is completed in about 30 h. An incomplete moult occurs within the egg, and larvae hatch at a sheathed second-stage 4312–8312 h later. Development occurred at all temperatures between 2° and 37°C with an optimum about 25°C and an upper limit near 37°C. The hatching process is very rapid, taking about 2 min. It is signalled by increased larval activity followed by a change in shell permeability. The larva hatches at that pole of the shell which has become plastic. The sheathed second-stage larva measures 659.50 ± 22.54 μm by 27.98 ± 1.22 μm. Its internal structures are concealed by a mass of opaque granules which were demonstrated as neutral lipid by oil red O staining. A second incomplete moult at 3–4 days results in a doubly sheathed infective larva from which the lipid gradually disappears. The mouth never appears patent and the larvae neither feed nor grow but rather decrease in size with age. Optimal temperatures for larvae range between 15°–25°C with 37°C about the upper limit. The significance of this developmental pattern is discussed.

The duration of larval and pupal stages of two species of predatory flies was investigated at five constant temperatures. Ilione albiseta (Scopoli): Mean duration of first and second instars was shortest at 23°C and there was 100% survival of first instar larvae at all temperatures. Mean third instar larval duration decreased from 104 days at 17°C to 27.5 days at 26°C. Total percentage larval survival was greatest at 17°C and least at 26°C. Mean pupal duration decreased as temperature increased and this occurred also under outdoor conditions. Pherbellia cinerella (Fallén): As temperature increased mean larval duration decreased but the percentage of larvae pupating also decreased (100% at 14°C, 33.3% at 26°C). There was a trend for shorter mean pupal durations at higher temperatures.

The reproduction and development of the prosobranch gastropodAlaba vladivostokensis Bartsch were studied. In Vostok Bay,A. vladivostokensis reproduces in the warmest season at water temperatures of 19–23°C. The females lay egg masses in the shape of flat, coiled bands on eelgrass leaves and on sargosso thalluses. The total period of development from egg laying to larval settling takes 25–27 days. The reproduction and development patterns and larval morphology ofA. vladivostokensis are similar to that ofAustralaba picta (A. Adams), which inhabits the coast of Japan. It is suggested that the population ofA. vladivostokensis in Peter the Great Bay should considered as part of the wide geographical range ofA. picta.

The perinatal (prenatal and early neonatal) period is a critical stage for hypothalamic programming of sexual differentiation as well as for the development of energy and metabolic homeostasis. We hypothesized that neonatal treatment with antidiabetic drug biguanide metformin would positively modify regulation of growth hormone – IGF-1 – insulin signaling pathway slowing down aging and improving cancer preventive patterns in rodents. To test this hypothesis male and female 129/Sv mice were s.c. injected with metformin (100 mg/kg) at the 3rd, 5th and 7th days after birth. Metformin-treated males consumed less food and water and their body weight was decreased as compared with control mice practically over their entire lifespan. There were no significant differences in age-related dynamics of food and water consumption in females and they were heavier than controls. The fraction of mice with regular estrous cycles decreased with age and demonstrated a tendency to decrease in the females neonatally treated with metformin. Neonatal exposure to metformin practically failed to change the extent of hormonal and metabolic parameters in blood serum of male and female mice. In males, neonatal metformin treatment significantly increased the mean life span (+20%, P < 0.05) and slightly increased the maximum life span (+3.5%). In females, the mean life span and median in metformin-treated groups were slightly decreased (−9.1% and −13.8% respectively, P > 0.05) in comparison to controls, whereas mean life span of last 10% survivors and maximum life span were the same as in controls. Almost half (45%) of control male mice and 71.8% male mice neonatally exposed to metformin survived up to 800 d of age, the same age was achieved by 54.3% of mice in control female group and 30% of metformin-treated females (P < 0.03). Thus, neonatal metformin exposure slows down aging and prolongs lifespan in male but not in female mice.

The eruciform larvae of holometabolous insects are primarily characterized by bearing a varying number of abdominal prolegs in addition to three pairs of thoracic legs. However, whether the prolegs are evolutionarily homologous among different insect orders is still a disputable issue. We examined the embryonic features and histological structure of the prolegs of the scorpionfly Panorpa byersi Hua and Huang (Mecoptera: Panorpidae) and the Oriental armyworm Mythimna separata (Walker) (Lepidoptera: Noctuidae) to investigate whether the prolegs are homologous between these two holometabolous insect orders. In the scorpionfly, paired lateral process primordia arise on abdominal segments I–VIII (A1–A8) in line with the thoracic legs in early embryonic stages, but degenerate into triangular protuberances in later stages, and paired medial processes appear along the midventral line before dorsal closure and eventually develop into unjointed, cone‐shaped prolegs. Histological observation showed that the lumina of the prolegs are not continuous with the hemocoel, differing distinctly from that of the basic appendicular plan of thoracic legs. These results suggest that the prolegs are likely secondary outgrowths in Mecoptera. In the armyworm, lateral process primordia appear on A1–A10 in alignment with the thoracic legs in the early embryonic stages, although only the rudiments on A3–A6 and A10 develop into segmented prolegs with the lumina continuous with the hemocoel and others degenerate eventually, suggesting that the prolegs are true segmental appendages serially homologous with the thoracic legs in Lepidoptera. Therefore, we conclude that the larval prolegs are likely not evolutionarily homologous between Mecoptera and Lepidoptera. J. Morphol. 277:585–593, 2016.

Ascarid Larva Migrans Syndrome (ascarid LMS) is a clinical syndrome in humans, caused by the migration of animal roundworm larvae such as Toxocara canis, Toxocara cati and Ascaris suum. Humans may acquire infection by ingesting embryonated eggs, or infective larvae of these parasites in contaminated meat and organ meats. To detect these pathogenic contaminations, a novel nested multiplex PCR system was developed. Our novel nested multiplex PCR assay showed specific amplification of T. canis, T. cati and Ascaris spp. Detection limit of the nested multiplex PCR was tested with serial dilution of T. canis, T. cati or A. suum genomic DNA (gDNA) from 100?pg to 100 ag and found to be 10?fg, 1?fg and 100?fg, respectively. When larvae were spiked into chicken liver tissue, DNA of T. canis and A. suum was detected from the liver spiked with a single larva, while the assay required at least 2 larvae of T. cati. Moreover, the ascarid DNA was detected from the liver of mice infected with 100 and 300 eggs of T. canis, T. cati or A. suum. This nested multiplex PCR assay could be useful for the detection of contamination with ascarid larvae in meat and organ meats.

Necrotizing enterocolitis (NEC) is a potentially fatal illness in premature neonates. Tumor necrosis factor-α (TNF-α) and autophagy are associated with the pathogenesis of NEC. This study aimed to explore whether TNF-α might regulate apoptosis in neonatal NEC model cells IEC-6 via regulation of autophagy. NEC rat model was induced by hand feeding and exposure to asphyxia/cold-stress for histologic examination. The NEC in vitro model (IEC-6/NEC cells) was established by stimulating the intestinal epithelial cell line IEC-6 with lipopolysaccharide (LPS, 100 μg/mL) for 3 h to investigate the effects of TNF-α on IEC-6 proliferation and apoptosis. In this study, NEC rats showed decreased proliferating cell nuclear antigen (PCNA) expression, increased TUNEL-positive cells, higher expression of TNF-α, p-ERK1/2, and autophagy-related proteins in rat small intestine compared with their controls. Additionally, the LPS-stimulated IEC-6/NEC cells showed a significantly decreased proliferation and increased apoptosis compared with the control cells. Furthermore, the LPS-stimulated IEC-6/NEC cells exhibited enhanced autophagy level, as evidenced by a dose-dependent increase in Beclin-1 protein expression, LC3II/LC3I ratio and accumulation of MDC-positive autophagic vacuoles. Moreover, inhibition of autophagy by wortmannin or LY294002 significantly abolished the LPS-mediated decreased proliferation and increased apoptosis of IEC-6/NEC cells. Results also showed that inhibition of ERK1/2 pathway using U0126 significantly inhibited TNF-α-induced autophagy. Furthermore, the TNF-α-mediated inhibition of IEC-6 proliferation and promotion of IEC-6 apoptosis was abolished by U0126. Our findings demonstrated that TNF-α might induce autophagy through ERK1/2 pathway to regulate apoptosis in neonatal NEC cells IEC-6. Our study enhances our understanding of neonatal NEC pathogenesis.
