A new taxonomic treatment of the Festuca ovina L. aggregate (Poaceae) in the British Isles. A wide range of morphological, anatomical and cytological characters was collected and analysed by a variety of methods, some taxometric. As a result nine species are recognized in the British Isles, one (F. glauca Vill.) only as a garden plant and two (F. huonii Auquier and F. armoricana Kerguélen) only in the Channel Isles. Two other species sometimes claimed for the British Isles (F. guestfalica Boenn. ex Reichb. and F. indigesta Boiss.) are excluded for various reasons given. One species (F. ovina L.) is divided into three subspecies: the diploid subsp. ovina (commonest in the north); and the tetraploid subsp. hirtula (Hackel ex Travis) M. Wilkinson (the commonest taxon of the aggregate in the British Isles) and subsp. ophioliticola (Kerguélen) M. Summary A major pathogen of potato plants (Solanum tuberosum) is the potato cyst nematode (Globodera spp.), which induces localized redifferentiation of a limited number of host cells to form a specialized feeding-site termed the syncytium. A novel strategy utilizing the polymerase chain reaction (PCR) was employed to construct a cDNA library from dissected potato roots highly enriched in syncytial material. The library was differentially screened with cDNA probes derived from the infected root tissue from a compatible interaction and from healthy root tissue. Characterization of one gene identified by the library screen indicated an expression pattern that correlated with events in the immediate vicinity of the pathogen after syncytial establishment. The strategy for library construction and screening could be applicable to the study of gene expression in any plant-pathogen interaction in which the limited supply of cells at the interface of the two organisms precludes a more traditional approach. 相似文献

With a view to cloning the root-knot nematode resistance gene Mi in tomato by chromosome walking, we have developed a molecular probe for the tightly linked acid phosphatase-1 (Aps-1) locus. The acid phosphatase-1 allozyme (APS-11), encoded by the Aps-1
1 allele originating from Lycopersicon peruvianum, was purified to apparent homogeneity from tomato roots and suspension cells. Microsequencing of CNBr and tryptic peptides generated from APS-11 provided a partial amino acid sequence, which accounted for approximately 23% of the protein and revealed two stretches of homology with soybean proteins KSH3 and VSP27, comprising 22 matches within 26 amino acid residues. The partial amino acid sequence information enabled us to isolate a 2.4 kb genomic Aps-1
1 sequence by means of the polymerase chain reaction (PCR), primed by degenerate pools of oligodeoxyribonucleotides, synthesized on the basis of the amino acid sequences. Synthesis of the 2.4 kb PCR product was specific for genomic templates carrying the L. peruvianum Aps-1
1 allele. Crucial to the priming specificity and the synthesis of the 2.4 kb genomic sequence was the use of degenerate primer pools in which the number of different primer species was limited by incorporating deoxyinosine phosphate residues at three and four base ambiguities. In using cDNA as a template, a 490 bp sequence was obtained, indicating a high proportion of intron sequences in the 2.4 kb genomic Aps-1

Engelm. (a species of claret-up cactus) andE. dasyacanthus
Engelm. (Texas Rainbow Cactus) were successful, resulting in the production of hundreds of seeds with hybrid embryos. The F1 hybrid progeny (i.e., syntheticE. ×lloydii) grew to sexual maturity in about four and one-half years, whereupon successful backcrosses and F2 generation hybrids were also obtained. The known F1 hybrids closely approximated naturalE. ×lloydii. The fertility of these syntheticE. ×lloydii was high, like their natural counterparts. The populations ofE. ×lloydii in Pecos County, Texas are inferred to have originated as the result of natural interspecific hybridization. Plant material ofCalamintha menthifolia was collected from two populations grown in the Vikos-Aoos area (NW. Greece). The structure and ontogeny of the glandular trichomes and the chemical composition of the secreted essential oil were studied. The leaves ofC. menthifolia have numerous glandular trichomes of three morphological distinct types: 1) glandular scales with a unicellular foot and stalk, and a 12-celled head, 2) glandular hairs with a unicellular foot and stalk, and a unicellular pear-shaped head and 3) glandular hairs with a unicellular foot, a bicellular stalk and a unicellular elongated head. Qualitative and quantitative GC-MS analyses of the essential oils revealed piperitone oxide as the main constituent in both populations. In the light of these results the relationships beweenC. menthifolia and other members of theSatureja group as well as its taxonomy are discussed. 相似文献

Hoplonemertines possess a family of pyridine compounds affecting the nervous system (Kem, 1985). Anabaseine, the first pyridine to be isolated, stimulates nicotinic receptors. Two other substances, 2,3-bipyridyl and nemertelline (a tetrapyridyl) were isolated from Amphiporus angulatus. In this study samples of 19 species of hoplonemertines were surveyed for the presence of stable pyridines using thin layer chromatography. Pyridines were selectively detected with the Konig reagent.Pyridines were found to be nearly ubiquitous constituents of this taxonomic group. Nevertheless, individual species often differed in the pyridines present as well as the variety of compounds present. A new Konig-reactive pyridine was found in 11 hoplonemertine species. Only Zygonemertes virescens and Amphiporus lactifloreus contained anabasine. Only A. angulatus contained 2,3-bipyridyl and nemertelline. This initial survey suggests that differences in pyridine compositions between hoplonemertines may be a useful taxonomic character. 相似文献

Summary The polystiliferous hoplonemertean Sagaminemertes nagaiensis (Iwata, 1957) has been redescribed.A distinct pre-cerebral region is wanting. The rhynchocoel opens directly at the tip of the head, whilst the mouth opens seperately below the ventral commissure of the brain which is situated at the anterior end of the head. The dorsal ganglia are well developed and extend posteriorly far behind the ventral ganglia. The cerebral sense organs are situated behind the brain and possess posterior ciliated canals. Cephalic glands are poorly represented, but ocelli are well developed. The sickle-shaped basis of the proboscis bears about 20 central stylets. The caecal appendages of the rhynchocoel are arranged pseudometamerically throughout the body. The foregut is straight, consisting of mouth, oesophagus, stomach and pylorus. Intestinal caeca and lateral diverticula are present. The cephalic lacunae have no anterior anastomosis. A cerebral lacuna is present, there is a rhynchocoel vessel, but transverse vessels are absent. The protonephridia extend throughout the foregut region. Mature spermatogonia are well developed.A systematic discussion of Sagaminemertes nagaiensis suggests that it is taxonomically close to Siboganemertes weberi in the archireptantic family Siboganemertidae. 相似文献

Within the last two decades, syntheses of both light-microscopic and ultrastructural characters have shown that there are three well-defined monophyletic groups within the Platyhelminthes: 1) the Catenulidale, 2) the Nemertodermatida-Acoela, and 3) the Haplopharyngida-Macrostomida-Polycladida-Neoophora (+ parasitic platyhelminth classes). However, the relationships among these three groups are problematic. The possible apomorphies that would unite them are either not true homologues (i.e. frontal organ), are mutually conflicting (i.e. 9+1 axoneme in spermatozoa vs. biflagellate spermatozoa, epidermal ciliary rootlet structure, and protonephridia), or are unrooted with any outgroup and hence untestable or uncertain as apomorphies (protonephridia, mode of epidermal replacement, absence of accessory centrioles on cilia). The chief obstacle to deciphering the relationships of these groups is the lack of information on them; presently available information is insufficient to test potential synapomorphies and insufficient also to allow agreement upon a narrowly defined outgroup for the Turbellaria.A view consistent with the present evidence (and admittedly an unsatisfactory view) is to regard the Turbellaria (and hence the Platyhelminthes) as polyphyletic, consisting of three separate and unrelatable groups. 相似文献

Effects of several population densities ofMeloidogyne incognita on the sweet potato cultivars Centennial (susceptible) and Jasper (moderately resistant) were studied. Field plots were infested with initial levels (Pi) of 0, 10, 100, 1,000, 5,000, and 10,000 eggs and juveniles/500 cm³ soil in 1980 and 0, 100, 1,000, 2,000, 3,000, 4,000, and 5,000 in 1981. M. incognita population development trends were similar on both cultivars; however, at high Pi, more eggs and juveniles were recovered from Centennial than from Jasper. The highest Pi did not result in the highest mid-season (Pm) counts. Pi was negatively correlated with the number of marketable roots and root weight but positively correlated with total cracked roots, percentage of cracked roots, and cracking severity. Jasper tolerated higher Pi with greater yields and better root quality than Centennial. Cracking of fleshy roots occurred with both cultivars at low Pi. 相似文献
