The stratum corneum (SC), the top layer of the epidermis, is the functional site of the skin barrier and serves to maintain hydration of the body by preventing water loss and thwarting the entrance of pathogens. The naked mole rat (NMR) (Heterocephalus glaber) is a rodent that resides in hypoxic underground tunnels in arid Africa. NMRs are not only hairless; their skin is devoid of glands and pain sensation. To understand how the skin barrier of the NMR is uniquely adapted to this environment, skin samples from the dorsum and ventral abdomen in one adult and one neonate were examined by transmission electron microscopy using both reduced osmium tetroxide to assess overall structure and ruthenium tetroxide post-fixation to assess lipid organization. These findings were compared with that of hairless mice—a well-defined model for skin barrier studies. The plasticity of the skin was evaluated on 10 NMRs from a colony at the Philadelphia Zoo in humid and dry conditions by measuring cutaneous hydration, transepidermal water loss (TEWL), and pH. The epidermal ultrastructure of the NMR differed from hairless mice by having the following features: decreased content of lamellar bodies (LBs), higher LB pleomorphism, periodic presence of abnormal lipid bilayers, and an unusually thick SC. The NMRs developed significant TEWL and a trend toward decreased hydration when subjected to dry conditions. While these features illustrate an imperfect skin barrier in terrestrial mammals, they likely represent adaptations of the poikilothermic NMRs to their unique natural fossorial climate. Prolonged exposure to decreased humidity could possibly lead to adverse health effects in this species. 相似文献
The naked mole-rat (NMR) Heterocephalus glaber is a unique and fascinating mammal exhibiting many unusual adaptations to a subterranean lifestyle. The recent discovery of their resistance to cancer and exceptional longevity has opened up new and important avenues of research. Part of this resistance to cancer has been attributed to the fact that NMRs produce a modified form of hyaluronan—a key constituent of the extracellular matrix—that is thought to confer increased elasticity of the skin as an adaptation for living in narrow tunnels. This so-called high molecular mass hyaluronan (HMM-HA) stems from two apparently unique substitutions in the hyaluronan synthase 2 enzyme (HAS2). To test whether other subterranean mammals with similar selection pressures also show molecular adaptation in their HAS2 gene, we sequenced the HAS2 gene for 11 subterranean mammals and closely related species, and combined these with data from 57 other mammals. Comparative screening revealed that one of the two putatively important HAS2 substitutions in the NMR predicted to have a significant effect on hyaluronan synthase function was uniquely shared by all African mole-rats. Interestingly, we also identified multiple other amino acid substitutions in key domains of the HAS2 molecule, although the biological consequences of these for hyaluronan synthesis remain to be determined. Despite these results, we found evidence of strong purifying selection acting on the HAS2 gene across all mammals, and the NMR remains unique in its particular HAS2 sequence. Our results indicate that more work is needed to determine whether the apparent cancer resistance seen in NMR is shared by other members of the African mole-rat clade. 相似文献

The effect of changed O2- and CO2-concentrations in air on oscillations in the transpiration rate of young oat plants in darkness has been investigated. Lowering the O2-concentration to 5% did not affect the oscillations. When the CO2 in the air was removed, the transpiration rate increased, and the oscillations ceased. When the CO2-concentration was raised to 0.3 or 3% the transpiration rate temporarily decreased, but the period of the oscillations was not changed. Further increase of the CO2-concentration caused, after a temporary decrease, an increased transpiration rate, and the oscillations eventually ceased. The period of the oscillations was influenced by the temperature: a lower temperature gave a longer period. It is concluded that substomatal O2-deficit or high CO2-concentration do not play a crucial role in the origin of these oscillations. 相似文献
One function of the Wingless signaling pathway is to determine the naked, cuticle cell fate choice in the trunk epidermis of Drosophila larvae. The zinc finger protein Teashirt binds to the transactivator domain of Armadillo to modulate Wingless signaling output in the embryonic trunk and contributes to the naked cell fate choice. The Hedgehog pathway is also necessary for the correct specification of larval epidermal cell fate, which signals via the zinc finger protein, Cubitus interruptus. Here, we show that Cubitus interruptus also has a Wingless-independent function, which is required for the specification of the naked cell fate; previously, it had been assumed that Ci induces naked cuticle exclusively by regulation of wg. Wg and Hh signaling pathways may be acting combinatorially in the same, or individually in different, cells for this process, by regulating common sets of target genes. First, the loss of the naked cuticular phenotype in embryos lacking cubitus interruptus activity is very similar to that induced by a late loss of Wingless function. Second, overexpression of Cubitus interruptus causes the suppression of denticles (as Wingless does) in absence of Wingless activity in the anterior trunk. Using epistasis experiments, we conclude that different combinations of the three proteins Teashirt, Cubitus interruptus, and Armadillo are employed for the specification of naked cuticle at distinct positions both along the antero-posterior axis and within individual trunk segments. Finally, biochemical approaches suggest the existence of protein complexes consisting of Teashirt, Cubitus interruptus, and Armadillo. 相似文献
The relationship between root stunting caused by the cereal cyst nematode and levels of two root growth inhibiting hormones, abscisic acid and ethylene, was investigated in aseptically cultured root segments and in intact roots of two oat cultivars differing in tolerance to the nematode. Cultured root segments of oat cultivars New Zealand Cape (tolerant) and Sual (intolerant) were inoculated with sterilized Heterodera avenae second-stage juveniles. Suppressed growth of root axes and emerged laterals following nematode penetration corresponded to an increase in abscisic acid and ethylene in roots of both intolerant and tolerant cultivars. When the experiment was repeated on intact root systems, nematodes retarded root growth of Sual more than New Zealand Cape despite an increase in ABA and ethylene in both cultivars. Abscisic acid and (or) ethylene may be involved in growth inhibition of H. avenae-infected roots but appear to play no direct role in determining tolerance. 相似文献
Since CO2 is liberated in the conversion of ACC to ethylene, the evidence that ethylene production by plant tissues is actually promoted by CO2 calls for an explanation. Accordingly, the formation of ethylene by oat (Avena sativa L. cv. Victory) leaves and by apple (Golden Delicious) and pear (Pyrus communis L. cv. Anjou) tissue in very low levels of CO2 has been studied. The gas chromatograph was modified to measure CO2 and ethylene at the same time, by reducing both to methane. (Response of the gas chromatograph to CO2 concentrations is linear.) The work establishes a clear difference between the endogenous production of ethylene and its production from applied ACC, a difference which holds about equally for leaves and for fruit tissue. The difference is in the CO2 requirement, namely, lowering the CO2 level by 99% or more decreases the production of ethylene from applied ACC by 50–60%, but it does not decrease, or even slightly increases, its production from endogenous precursors. Thus, while the need for CO2 has not been explained, it has at least been delimited. The responses to abscisic acid (ABA) also differ, but in the reverse direction, the endogenous production of ethylene being decreased up to 70% by ABA. while the liberation from ACC is promoted about 20%. ABA also promoted the respiratory CO2 production by 30% or, in presence of 1-aminocyclopropane-1-carboxylic acid (ACC), by over 50%. Inhibition of ethylene production by cobalt or EDTA shows no such differentiation, while inhibition by Na catechol-4,6-disulfonate (Tiron) shows a small difference. It is concluded either that endogenous ethylene is formed by an enzyme system different from that reacting with ACC, or (more likely) that when ethylene arises from endogenous precursors the reaction does not proceed by way of free ACC, but by some activated form of it. 相似文献
