We conducted a series of gravitropic experiments on Avena coleoptiles in the weightlessness environment of Spacelab. The purpose was to test the threshold stimulus, reciprocity rule and autotropic reactions to a range of g-force stimulations of different intensities and durations The tests avoided the potentially complicating effects of earth's gravity and the interference from clinostat ambiguities. Using slow-speed centrifuges, coleoptiles received transversal accelerations in the hypogravity range between 0.1 and 1.0 g over periods that ranged from 2 to 130 min. All responses that occurred in weightlessness were compared to clinostal experiments on earth using the same apparatus.
Characteristic gravitropistic response patterns of Avena were not substantially different from those observed in ground-based experiments. Gravitropic presentation times were extrapolated. The threshold at 1.0 g was less than 1 min (shortest stimulation time 2 min), in agreement with values obtained on the ground. The least stimulus tested, 0.1 g for 130 min, produced a significant response. Therefore the absolute threshold for a gravitropic response is less than 0.1 g. 相似文献

Identification and characterization of a heat-induced isoform of aldolase in oat chloroplast 总被引:1,自引:0,他引:1
An analysis of protein synthesis at elevated temperatures in oat (Avena sativa) leaves revealed a heat-induced 44 kDa polypeptide. A cDNA library of heat-treated leaves was constructed and screened with specific antibodies raised against this 44 kDa polypeptide. A clone encoding the 44 kDa protein was identified as a form of the chloroplast-localized fructose-bisphosphate aldolase (EC 4.1.2.13). Northern and western blot analyses indicated heat-induced accumulation of the chloroplast aldolase isoform at both the RNA and protein level. Heat inducibility was restricted to the chloroplastic form of the enzyme, and was not observed for the cytoplasmic aldolase. The heat-induced isoform co-purified with thykaloid fractions, as confirmed by immunoassay and activity analyses. However, when thylakoid membranes were treated with proteinase K, the aldolase isoform completely disappeared, suggesting that this enzyme is not embedded but rather tends to adhere to the chloroplast membranes. Immunoblot analysis of other plant species revealed similar heat induction of thykaloid-associated aldolase homologues, suggesting the possible existence of a universal control mechanism for this enzyme's heat tolerance 相似文献

Abstract A new species of pest mite, Penthaleus tectus sp. n. (Acari: Penthaleidae) is described from south-eastern Australia, where it attacks several grain crops including wheat, in which it can cause significant damage. It has previously been confused with the blue oat mite Penthaleus major (Dugès). Penthaleus tectus differs from P. major morphologically and genetically, and in geographical distribution, response to pesticides, and the timing of diapause. Previous records of ' P. major ' from Australia and other countries probably include a mixture of several species. Penthaleus minor (Canestrini) is recorded from South Africa for the first time. 相似文献

Polyclonal antibodies, raised against ((1→3), (1→4)-β-D-glucans from oat ( Avena sativa L.) caryopsis, were used to investigate the location and the metabolism of mixed-linked β-D-glucans. The binding of these antibodies to the cell walls of oat coleoptiles was shown by an indirect fluorescence method. Distinct fluorescent regions were observed along the inner layers of the walls of each cell. The preimmune serum or antibodies pretreated with oat caryopsis β-D-glucans did not react with the cell walls. Glucan antibodies were bound to the walls of other Poaceae coleoptiles as well as to those from oat mesocotyls and roots, whereas they were not bound to the walls of some dicotyledons tested. The relative glucan content of the cell walls of oat coleoptiles as determined by β-D-glucanase (EC 3.2.1.73) treatment was maximum between day 3 and 4 after soaking, but it declined during further elongation. A rapid decrease in glucan content was observed in excised coleoptiles when auxin or β-D-glucanase was present. There was a clear correlation between the glucan content expressed on a basis of cell wall polysaccharides and the amount of the antibodies bound to the cell walls. 1 Population growth of four arthropod pests on agricultural crops, the pea aphid, the bird‐cherry oat aphid, the Mediterranean fruit fly, and the two‐spotted spider mite, was modelled using stage matrix models. 2 Temperature was included as a variable in the model, affecting fecundity, mortality and growth rate. Linear temperature dependence was used to describe fecundity and individual growth rate in the matrix model. Life table data collected at various constant experimental temperatures were used to parameterize the model. 3 Sensitivity analysis identified the most influential demographic parameters determining the growth rate of the population at different temperature regimes. Mortality of young adults and immature stages were found to be the most important parameters in determining population growth for all species presented. However, high temperatures increased the relative impact of individual growth rates and fecundity on the growth of the population. 相似文献

The planthopper Delphacodes kuscheli is the main vector of Mal de Río Cuarto virus in Argentina, disease that severely affects maize production. In this study, we investigated the effects of heat stress on fitness traits and on the number of its obligate yeast‐like symbionts (YLS). The exposition of newly‐hatched nymphs to 35°C for 3 days, a well‐known procedure used to reduce the number of YLS in planthoppers, was applied. To compare different fitness components between control and heat‐treated insects, we estimated nymphal instars development time, nymphal survival, adult body length, longevity, fecundity and fertility. Also, correlates of fitness, as proportion of sexes and wing forms of the emerging adults, were evaluated. In heat‐treated group, the nymphal developmental time increased due to an increase in the fifth instar duration, and the nymphal survival, body length of adults and fecundity were reduced when compared to control. There was a significant association between treatments (control and heat‐treated insects) and wing morphs. The heat treatment successfully reduced the number of YLS in third instar nymphs of D. kuscheli. Our results revealed the negative effect of heat stress on development, survival and reproduction of D. kuscheli and on the load of its YLS endosymbionts suggesting that YLS could play a crucial role in the development and reproduction of these planthoppers. 相似文献

Naked mole‐rats are eusocial mammals that live in colonies with a single breeding female and one to three breeding males. All other members of the colony, known as subordinates, are nonreproductive and exhibit few sex differences in behavior or genital anatomy. This raises questions about the degree of sexual differentiation in subordinate naked mole‐rats. The striated perineal muscles associated with the phallus [the bulbocavernosus (BC), ischiocavernosus (IC), and levator ani (LA) muscles], and their innervating motoneurons, are sexually dimorphic in all rodents examined to date. We therefore asked whether perineal muscles and motoneurons were also sexually dimorphic in subordinate naked mole‐rats. Muscles similar to the LA and IC of other rodents were found in naked mole‐rats of both sexes. No clear BC muscle was identified, although a large striated muscle associated with the urethra in male and female naked mole‐rats may be homologous to the BC of other rodents. There were no sex differences in the volumes of the LA, IC, or the urethral muscles. Motoneurons innervating the perineal muscles were identified by retrograde labeling with cholera‐toxin‐conjugated horseradish peroxidase. All perineal motoneurons were found in a single cluster in the ventrolateral lateral horn, in a position similar to that of Onuf's nucleus of carnivores and primates. There was no sex difference in the size or number of motoneurons in Onuf's nucleus of naked mole‐rats. Thus, unlike findings in any other mammal, neither the perineal muscles nor the perineal motoneurons appear to be sexually differentiated in subordinate naked mole‐rats. © 2002 Wiley Periodicals, Inc. The objective of this study was to evaluate the use of oat hulls (OH) to substitute traditional fiber (a mixture of rice hulls and mugwort (RHM) leaf) in the diets of fattening rabbits by examining on its effect on the growth performance, coefficient of total tract apparent digestibility (CTTAD) of nutrients, gastrointestinal tract development, cecum fermentation and carcass traits. A total of 160 mixed sex Hyla commercial meat rabbits (40/treatment) were used to study the effects of including OH in the diet from 30 to 80 days of age. A control diet based on RHM and containing 175.2 g crude fiber and 169.7 g CP/kg was included. Growth performance and CTTAD of nutrients were recorded from day 35 to day 80 and day 74 to day 80, respectively, whereas gastrointestinal tract development, cecum fermentation and carcass traits were determined at day 80. Increasing concentrations of OH in the diet increased average daily feed intake (P=0.0018), but have no effects on average daily gain and feed conversion ratio (P>0.05). Moreover, the 150 g/kg OH diet decreased the relative weight of full cecum and cecal content (P<0.05), but did not affect other gastrointestinal organs. The CTTAD of NDF and gross energy decreased with the inclusion of OH (P<0.05). No effects of CTTAD of ADF, ADL, dry matter, CP and ether extract were observed (P>0.05). The concentrations of total volatile fatty acid, acetic and butyric acids were greater in rabbits fed the lower levels of OH (0 to 100 g/kg) compared with higher rate (150 g/kg; P<0.05). However, the proportions of acetic, propionic and butyric acids were not affected by treatments (P>0.05). Furthermore, no significant effect on the ratios of acetic/(propionic+butyric) was observed (P>0.05). Hot carcass weight, pH (45 min, 24 h), lightness, redness, yellowness, 24-h drip loss of longissimus lumborum muscles were not affected by diet OH (P>0.05). However, dressing out percentage increased with higher (150 g/kg) inclusion of OH. It is concluded that OH can be included in rabbit diets at levels up to 100 g/kg, but negative effect on digestion and performance were observed with the increasing of OH (150 g/kg). 相似文献
