The D526-derived BC1F2 population of hexaploid oat segregates for resistance to crown rust isolate 345. A mapping population consisting of 440 F2 individuals was used to develop a high-resolution RFLP map of the area of the genome where Pc71, the locus conferring this resistance, is located. Twelve RFLP markers have been identified within ca. 6 cM of Pc71, with cosegregating loci Xcdo1502 and Xcdo783 positioned 0.2 cM from the locus. All of the RFLP markers map to the same side of the locus, suggesting either that the Pc71 resistance locus resides at the end of a linkage group, or that there is no detectable heterozygosity on the opposite side. Some degree of microcollinearity between rice and oat is present in this area, as the two markers, Xcdo783 and Xrz69, linked to Pc71 are linked also in rice; however the genetic distance in oat is much less than the genetic distance in rice (6.2 cM and 20 cM, respectively).  相似文献

Torenia fournieri Lind. has a naked embryo sac that protrudes from the micropyle. The precise time course of the entire process of double fertilization and the kinetics of fertilization events were determined in this species by the following methods: (i) without squashing, pollen tubes on the torn stylar canal were observed by fluorescence microscopy after staining with both 4′,6-diamidino-2-phenylindole (DAPI) and aniline blue; and (ii) large numbers of living embryo sacs were observed directly by differential interference microscopy before and after fertilization. The pollen began to germinate 5 min after pollination and extruded pollen tubes which elongated at a constant rate of 2.3 mm · h−1. At 4.0 h after pollination, the mitotic index of the generative cell within the pollen tube reached 88% and the two sperm cells were formed. Pollen tubes began to arrive at ovules 8.9 h after pollination and directly entered one of two synergids in the naked embryo sac. The time required for transport of sperm cells in the degenerated synergid was estimated statistically to be 1.9 ± 1.8 min for transport of the first cell and 7.4 ± 1.6 min for the second. In the nucleus of the fertilized egg cell, the male nucleolus began to emerge 10 h after pollination and the female nucleolus often decreased in size. The two nucleoli fused together prior to elongation of the zygote, which began 28 h after pollination. In the central cell, the secondary nucleus migrated to a region adjacent to the egg apparatus after pollination but prior to the arrival of the pollen tube. The primary endosperm nucleus rapidly returned to the inner region after fertilization. Prior to embryogenesis, the first division of the primary endosperm began about 15 h after pollination, at a defined site, to form the chalazal haustorium. The norbornenodiazetine plant growth regulator tetcyclacis, when applied to roots of Avena sativa, caused a substantial increase in the cholesterol content of the shoots. Amounts of the C-24 alkylated sterols campesterol, stigmasterol and sitosterol all declined. A similar alteration in the sterol profile was observed for a plasma membrane preparation from the shoots. Changes in the sterol composition of root tissue were much less pronounced.  相似文献

Barley yellow dwarf virus (BYDV) causes significant losses in yield and in overwintering ability of winter cereals. Mechanisms by which the physiology of plants is affected by the virus are not clear. To see how carbohydrates in the crown of winter cereals were affected by BYDV, fructan isomers of degree of polymerization (DP) 3–5, fructan DP>6 and the simple sugars, glucose, fructose and sucrose, were measured before and during cold hardening in three oat ( Avena sativa L.) cultivars, 'Wintok', 'Coast Black' and 'Fulghum'. On a fresh weight basis fructan DP>6 decreased by 50% in infected 'Wintok' and 'Coast Black' and by 25% in 'Fulghum'. Two DP3, one DP4 and one DP5 isomer were significantly higher than non-infected controls. The percentages of simple sugars in infected crowns were significantly higher than controls in all three cultivars in every week except the first week of hardening. Crude enzyme extracts from BYDV infected plants incubated with sucrose suggested higher invertase and lower sucrose-sucrosyl transferase activity. When incubated with 1-kestose and neokestin, no significant difference was found in fructose fructosyl transferase or in hydrolase activity. The activity of unidentified enzymes catalysing the synthesis of larger (DP>5) fructan was altered by BYDV. The decrease of carbohydrates in the crown induced indirectly by BYDV may alter the plant's capacity to regenerate tillers in the spring. The ability of plants to prevent or tolerate carbohydrate fluctuations induced by BYDV infection may be an important genetically regulated characteristic for developing virus-resistant cultivars.  相似文献

Segments cut from the next-to-last (peduncular-1) internode of Avena sativa L. cv. Victory (oat) shoots elongate as much as 10-fold in response to gibberellic acid (GA3). The objective of the present investigation was to differentiate the effects of GA3 on growth from its effects on wall synthesis (measured gravimetrically and through the incorporation of [14C]-glucose) by using several cell wall synthesis inhibitors with widely varying mechanisms of action. Four compounds, viz. monensin, cycloheximide, lanthanum, and galactose. caused (1) relatively little inhibition of either cell wall synthesis or elongation in segments without GA3, (2) roughly proportionate, dose-dependent inhibition of elongation and wall synthesis in GA3-treated segments and (3) generally greater inhibition of GA3-promoted uptake of radioactivity than of wall incorporation or elongation. Two other compounds, colchicine and 2,6-dichlorobenzonitrile (DCB). (1) inhibited GA3-induced elongation considerably more than cell wall synthesis and (2) caused swelling (radial expansion). especially of GA3-treated segments. DCB-treated internodal cells apparently compensated for inhibited cellulose synthesis by greater synthesis of matrix polysaccharide (beginning between 3 and 6 h). While normal cellulose synthesis was not required for short-term (up to 6 h) GA3-induced elongation or for long-term hormone-promoted radial expansion, it was required for sustained GA3-induced elongation. These results indicate that GA3-promoted cell wall loosening (manifested as radial expansion) and cell wall synthesis in Avena internodes occur at least partially independently of any hormonal effect on the orientation of microtubules and microfibrils.  相似文献

We conducted a series of gravitropic experiments on Avena coleoptiles in the weightlessness environment of Spacelab. The purpose was to test the threshold stimulus, reciprocity rule and autotropic reactions to a range of g-force stimulations of different intensities and durations The tests avoided the potentially complicating effects of earth's gravity and the interference from clinostat ambiguities. Using slow-speed centrifuges, coleoptiles received transversal accelerations in the hypogravity range between 0.1 and 1.0 g over periods that ranged from 2 to 130 min. All responses that occurred in weightlessness were compared to clinostal experiments on earth using the same apparatus.
Characteristic gravitropistic response patterns of Avena were not substantially different from those observed in ground-based experiments. Gravitropic presentation times were extrapolated. The threshold at 1.0 g was less than 1 min (shortest stimulation time 2 min), in agreement with values obtained on the ground. The least stimulus tested, 0.1 g for 130 min, produced a significant response. Therefore the absolute threshold for a gravitropic response is less than 0.1 g.  相似文献

Soil solutions were collected for speciation analysis of nickel from a pot experiment with oats. Oat plants (Avena sativa L.) were grown on 3 soils differing in total amount and origin of nickel (Ni) (Luvisol, LS with 28 mg kg-1; sludge amended Luvisol, LS+SS with 32 mg kg-1; Cambisol, CS with 95 mg kg-1). Results were compared with those for soil solutions obtained from corresponding unplanted pots. Separation methods were used for characterization of size and charge distribution and stability of the Ni species. In addition, short-term experiments were performed on the uptake rates of Ni by oat plants from the different soil solutions as well as from nutrient solutions with increasing concentrations of a synthetic chelator.The Ni concentrations in the soil solutions of unplanted soils increased in the order: LS5000 g mol-1) was the predominant form, whereas in the other soils the low-molecular-size cationic and chelated Ni species (500–1000 g mol-1) dominated in the soil solution. In the short-term uptake studies, the uptake rates of Ni from the solutions decreased in the order: nutrient solution > soil solutions, and in the latter in the order: LS>LS+SS>CS, which was inversely related to the concentrations of dissolved organic carbon in the soil solutions.The results demonstrate that Ni availability to plants is not only affected by total concentration of Ni in the soil solution and the rate of replenishment from the solid phase, but also by Ni species, which can differ considerably between soil types.  相似文献

Puccinia coronata f. sp. avenae (Pca) causes crown rust disease in cultivated and wild oat (Avena spp.). The significant yield losses inflicted by this pathogen make crown rust the most devastating disease in the oat industry. Pca is a basidiomycete fungus with an obligate biotrophic lifestyle, and is classified as a typical macrocyclic and heteroecious fungus. The asexual phase in the life cycle of Pca occurs in oat, whereas the sexual phase takes place primarily in Rhamnus species as the alternative host. Epidemics of crown rust happens in areas with warm temperatures (20–25 °C) and high humidity. Infection by the pathogen leads to plant lodging and shrivelled grain of poor quality. Disease symptoms : Infection of susceptible oat varieties gives rise to orange–yellow round to oblong uredinia (pustules) containing newly formed urediniospores. Pustules vary in size and can be larger than 5 mm in length. Infection occurs primarily on the surfaces of leaves, although occasional symptoms develop in the oat leaf sheaths and/or floral structures, such as awns. Symptoms in resistant oat varieties vary from flecks to small pustules, typically accompanied by chlorotic halos and/or necrosis. The pycnial and aecial stages are mostly present in the leaves of Rhamnus species, but occasionally symptoms can also be observed in petioles, young stems and floral structures. Aecial structures display a characteristic hypertrophy and can differ in size, occasionally reaching more than 5 mm in diameter. Taxonomy : Pca belongs to the kingdom Fungi, phylum Basidiomycota, class Pucciniomycetes, order Pucciniales and family Pucciniaceae. Host range : Puccinia coronata sensu lato can infect 290 species of grass hosts. Pca is prevalent in all oat‐growing regions and, compared with other cereal rusts, displays a broad telial host range. The most common grass hosts of Pca include cultivated hexaploid oat (Avena sativa) and wild relatives, such as bluejoint grass, perennial ryegrass and fescue. Alternative hosts include several species of Rhamnus, with R. cathartica (common buckthorn) as the most important alternative host in Europe and North America. Control : Most crown rust management strategies involve the use of rust‐resistant crop varieties and the application of fungicides. The attainment of the durability of resistance against Pca is difficult as it is a highly variable pathogen with a great propensity to overcome the genetic resistance of varieties. Thus, adult plant resistance is often exploited in oat breeding programmes to develop new crown rust‐resistant varieties. DNA fragments containing the fibroin H-chain gene from two different strains of Bombyx mori, J-139 and Nd (2) were cloned into phage lambda Charon 4A. Comparison of the restriction sites in these cloned DNAs revealed that in addition to the known polymorphism in the region coding for the repetitive amino acid sequence of the fibroin H-chain [Manning and Gage, J. Biol. Chem. 255 (1980) 9451-9457], at least two other types of polymorphism were present, one around the 5' end of the structural gene, and the other in the far upstream region of the gene. Restriction sites around the 5' end of the gene were well conserved between these strains, but some heterogeneity, suggesting the presence of small insertions, deletions or base changes, was noted. In contrast, DNA sequences of the region 2-4 kb upstream from the 5' end of the gene were markedly different between these two strains, indicating that either a deletion or an insertion of a DNA sequence longer than 2 kb had occurred in this region. Comparison with several other strains suggested that the observed changes in the far-upstream region were unique to the Nd(2) strain.  相似文献
