Aims:  Investigating the influence of an added starter culture on the properties of fermented liquid pig feed.
Methods and Results:  Diets of cereal grain blended with wet wheat distillers' grain that were either not inoculated (WWDG), inoculated with a silage starter culture at start (WWDGsc1) or at start and at each backslopping (replacement of 80% the content with fresh mixture, simulating feed outtake, WWDGsc5) were fermented for 5 days, followed by 5 days of daily backslopping. Numbers of undesirable micro-organisms (enterobacteria, moulds) were reduced in all fermentations; particularly enterobacteria in the starter culture inoculated diets. Lactobacillus plantarum present in the starter culture became dominant in diets WWDGsc1 and WWDGsc5. However, Lactobacillus panis that was dominating WWDG was also abundant in WWDGsc1 and WWDGsc5. Yeast populations were not influenced by the starter culture, with Pichia fermentans dominating all fermentations. All diets had similar chemical characteristics with the exception of a significant increase of all tested organic acids in WWDGsc5.
Conclusions:  The addition of a starter culture influences the bacterial population in fermented liquid feed, but there is also a strong impact of the flora already present in the feed ingredients. The yeast population is not influenced by adding a lactic acid bacteria (LAB) starter culture. A consortium of LAB and yeast strains adapted to the fermentation should be used as starter culture.
Significance and Impact of the Study:  The results suggest that it is possible to influence the current unpredictable and spontaneous process of feed fermentation when appropriate starter cultures are used. For this purpose, LAB and yeasts with desirable characteristics should be isolated.  相似文献

目的建立并优化环介导等温扩增(LAMP)技术对解脲脲原体(U.urealyticum)的检测,并应用于临床样本分析。方法针对U.urealyticum的urease基因设计LAMP引物;研究LAMP的最适温度、最佳检测时间及灵敏度和特异度;与传统PCR检测进行方法学比对。结果 LAMP技术检测U.urealyticum的最适温度和最佳时间分别是61℃和60 min,并且具有良好灵敏度和特异度,较普通PCR检测的灵敏度高出1 000倍。临床样本检测中,PCR和LAMP技术达到的灵敏度分别为25.00%和87.50%。两种方法的特异度均为100.00%。结论 LAMP与PCR相比在基层检测和大规模筛查方面有显著的优势和巨大的利用价值。  相似文献

目的:探讨难治性肺炎支原体肺炎(RMPP)患儿血清炎性细胞因子变化的临床意义及其危险因素。方法:将我院从2016年1月~2020年1月收治的72例RMPP患儿纳入研究,记作RMPP组。另取我院同期收治的普通肺炎支原体肺炎(MPP)患儿70例作为对照组。检测所有患儿的血清炎性细胞因子水平,并进行对比。此外,对比两组患儿各项基线资料,并以多因素Logistic回归分析RMPP发生的危险因素。另外,比较两组患儿的影像学特征。结果:RMPP组患儿血清白细胞介素-6(IL-6)以及白细胞介素-10(IL-10)水平均高于对照组(P0.05);而两组肿瘤坏死因子-α(TNF-α)水平对比差异不明显(P0.05)。RMPP组年龄、发热持续时间、发热峰值、住院时间以及发热伴随症状人数占比均高于对照组(P0.05)。经多因素Logistic回归分析发现:发热持续时间、发热峰值、住院时间以及发热伴随症状均是RMPP患儿发病的危险因素(OR=2.773、2.344、1.058、2.515,均P0.05)。RMPP组大叶性肺炎、胸腔积液人数占比均高于对照组,而支气管肺炎人数占比低于对照组(P0.05)。结论:RMPP患儿血清IL-6以及IL-10水平均存在明显高表达,且发热持续时间、发热峰值、住院时间以及发热伴随症状均是RMPP发病的危险因素,临床工作中需要予以重视。  相似文献

Aims:  To investigate the inter-strain variation in (i) substrate utilization and (ii) the restriction fragment length polymorphism (RFLP) pattern based on the distribution of an insertion element (IS 1550 ) in Mycoplasma fermentans strains, and to establish any correlation between subgroups within the species and their source or habitat.
Methods and Results:  Using a sensitive dynamic pH method, the pattern and kinetics of substrate utilization by a panel of 17 M. fermentans strains from various sources was determined. This study correlated the biochemical characteristics of these strains with RFLP patterns based on the distribution of an insertion sequence (IS 1550 ) with the sources of the strains. The test isolates were divided into four major groups according to the pattern of substrates metabolized. Interestingly, two strains isolated from cell lines in RFLP cluster I failed to utilize arginine. Ovine strains showed distinct substrate utilization patterns and produced RFLP patterns not previously encountered.
Conclusions:  All strains utilized glucose, but the ability to utilize arginine, fructose and N -acetyl glucosamine varied. There was also some correlation evident between the metabolic data and the RFLP clusters.
Significance and Impact of the Study:  This study has provided a better understanding of the biochemical and genetic diversity of M. fermentans strains from various sources.  相似文献

The contamination of cell cultures by mycoplasmas remains a major problem in cell culture. Mycoplasmas can produce a virtually unlimited variety of effects in the cultures they infect. These organisms are resistant to most antibiotics commonly employed in cell cultures. Here we provide a concise overview of the current knowledge on: (1) the incidence and sources of mycoplasma contamination in cell cultures, the mycoplasma species most commonly detected in cell cultures, and the effects of mycoplasmas on the function and activities of infected cell cultures; (2) the various techniques available for the detection of mycoplasmas with particular emphasis on the most reliable detection methods; (3) the various methods available for the elimination of mycoplasmas highlighting antibiotic treatment; and (4) the recommended procedures and working protocols for the detection, elimination and prevention of mycoplasma contamination. The availability of accurate, sensitive and reliable detection methods and the application of robust and successful elimination methods provide powerful means for overcoming the problem of mycoplasma contamination in cell cultures. Mycoplasma pneumoniae is the leading cause of bacterial community‐acquired pneumonia among hospitalised children in United States and worldwide. Community‐acquired respiratory distress syndrome (CARDS) toxin is a key virulence determinant of M. pneumoniae. The N‐terminus of CARDS toxin exhibits ADP‐ribosyltransferase (ADPRT) activity, and the C‐terminus possesses binding and vacuolating activities. Thiol‐trapping experiments of wild‐type (WT) and cysteine‐to‐serine‐mutated CARDS toxins with alkylating agents identified disulfide bond formation at the amino terminal cysteine residues C230 and C247. Compared with WT and other mutant toxins, C247S was unstable and unusable for comparative studies. Although there were no significant variations in binding, entry, and retrograde trafficking patterns of WT and mutated toxins, C230S did not elicit vacuole formation in intoxicated cells. In addition, the ADPRT domain of C230S was more sensitive to all tested proteases when compared with WT toxin. Despite its in vitro ADPRT activity, the reduction of C230S CARDS toxin‐mediated ADPRT activity‐associated IL‐1β production in U937 cells and the recovery of vacuolating activity in the protease‐released carboxy region of C230S indicated that the disulfide bond was essential not only to maintain the conformational stability of CARDS toxin but also to properly execute its cytopathic effects.  相似文献

Mycoplasma fermentans has been reported to be pathogenic for man. All fourteen strains tested contain an insertion sequence-like element (ISLE) which may be present in multiple copies. To determine whether ISLE copies are similarly distributed in different strains of M. fermentans, restriction enzyme digest fragments of genomic DNA from 14 isolates, from a variety of sources, were separated by electrophoresis, blotted and hybridized to a biotin labelled polymerase chain reaction (PCR) amplified fragment of ISLE. A range of patterns was observed suggesting that the element has a tendency to undergo rearrangement within the genome. Analysis of ISLE sequences revealed inter- and intra-strain polymorphisms.  相似文献

Attempts were made to detect Mycoplasma fermentans in saliva sampled from 201 subjects (108 males and 93 females) aged from 4 months to 59 years by a polymerase chain reaction-based assay. M. fermentans was detected in saliva from 110 (54.7%) of 201 subjects, and 10 (28.6%) of 35 subjects aged from 4 months to 3 years. Of ten positive subjects, three were aged from 16 to 23 months and five were from 26 to 31 months. The incidence tended to increase with age up to the teens. The incidence was significantly greater in teenagers than in subjects aged from 7 to 12 years, but there was no significant difference in the incidence between the group of teenagers and each of the groups of subjects older than the teenagers. Thus, it was suggested that M. fermentans colonized the mouth at the age of about 16 months up to the age of 19 years.  相似文献
