A system is described for rapid re-attachment of the rhodophycean alga Gelidium sesquipedale (Clem.) Born. et Thur. on artificial or natural substrata. This method is applicable to industrial cultivation of this species. The function of rhizoidal clusters and the origin of germlings from the apical portion of the thalli are analyzed in relation to the re-attachment process. The role that re-attachment might play in the maintenance and spreading of natural populations and in the observed anomalies of the life-history of this species is discussed.  相似文献

The artificial culture of Macrocystis integrifolia in southeastern Alaskan waters has been initiated as a first step in a project to augment the existing herring roe-on-kelp fishery in the state. The growing of Macrocystis in Prince William Sound has the potential of considerably enhancing this fishery by eliminating the costly importation of this kelp from southeast Alaska. In an ongoing feasibility study, Macrocystis has been cultured under laboratory conditions through the embryonic sporophyte stage and outplanted in waters near Sitka, Alaska (57° N). Growth of the outplanted kelps has been monitored as a function of the time and depth of the outplanting. Preliminary results suggest that light is limiting for growth in winter and that the optimal depth for outplanting will vary with the season.  相似文献

SYNOPSIS A method is described for the axenic mass cultivation of Paramecium tetraurelia strains 51s and 299s. the ciliate is grown in an enriched axenic medium developed by Soldo, Godoy & van Wagtendonk. Under continuous shaking on a rotary shaker, cultures were grown in one-liter Erlenmeyer flasks with 330 ml medium yield cell densities of 32,000 cell/ml and 20,000 cells/ml for strains 299s and 51s respectively. Doubling time is considerably shorter under these conditions than in the conventional static cultures. A 20-liter airlift bioreactor is described in detail which can be used successfully to otain up to 100 g wet weight of Paramecium in a single run; in this reactor the cell density reaches 38,000 cells/ml for strain 299s. and 23,000 cells/ml for 51s. This technic should facilitate the study of minor protein components of the ciliate.  相似文献

Summary Cultures of embryonic rat brain and liver, and embryonic turkey brain were inoculated with sporozoites ofPlasmodium berghei. Sporozoites succeeded in establishing exoerythrocytic infections in approximately 10% of the cultures. The exoerythrocytic parasites developed to a late schizont stage with some showing early segmentation although free merozoites were not observed. The morphology and rate of development of the exoerythrocytic parasites in culture appear similar to that seen in vivo. SYNOPSIS. Five proteins capable of stimulating [3H]thymidine uptake by Trypanosoma cruzi in vitro were isolated from fetal calf serum by (NH4)2SO4 precipitation and ion exchange column chromatography. The proteins were partially characterized by immunodiffusion, immunoelectrophoresis, polyacrylamide gel disc electrophoresis, and SDS electrophoresis. As estimated by SDS electrophoresis, using 4 standards, the molecular weight of protein 1 was 100,000, that of protein 2 was 76,000. and that of proteins 3–5 was 68,000 daltons.  相似文献

Boletus edulis is a well-known ectomycorrhizal mushroom. Although cultivation has been widely attempted, no artificial fruiting has been achieved owing to difficulties associated with mycorrhizal synthesis and acclimatization in fields. We collected fifteen B. edulis basidiomata samples from locations in Japan and identified them microscopically and by phylogenetic analysis of their nuclear ribosomal internal transcribed spacer (ITS) regions. Pure culture isolates of B. edulis were established efficiently on malt extract agar medium, and one isolate, EN-63, was inoculated to axenic Pinus densiflora seedlings in vitro. Brownish ectomycorrhizal tips were observed on the pine lateral roots within four months of inoculation. Ten pine seedlings that formed ectomycorrhizae were acclimatized under laboratory and greenhouse conditions. At four months after transplant, mycorrhizal colonization by B. edulis was observed on newly grown root tips under laboratory conditions, but no B. edulis ectomycorrhiza survived under greenhouse conditions. These results suggest that B. edulis ectomycorrhizae synthesized in vitro with P. densiflora requires additional steps for acclimatization to greenhouse conditions.  相似文献

Twenty-four Basidiomycetes strains were evaluated to determine their free-radical scavenging capacity in submerged cultivation. The scavenging capacity of the extracts varied from 1 to 85% depending on the mushroom species, solvent used, and concentration. A calculation of EC50 of extracts from several wood-rotting Basidiomycetes showed high scavenging abilities at low effective concentration.  相似文献

以不同灵芝品种、不同栽培基质、不同管理方式、不同生长时期获得的灵芝子实体为原料,用95%乙醇超声提取,对提取物进行了含量测定、三萜组成分析和体外抗肿瘤实验。结果表明,不同子实体醇提物中三萜和甾醇类物质的含量在4.60-6.20mg/g之间;高效液相分析发现10种三萜化合物的种类和含量在样品间存在明显差异。所有灵芝子实体醇提物对肿瘤细胞L1210的增殖均有一定的抑制作用,开伞期的灵芝子实体醇提物抑制肿瘤增殖的活性优于其他生长时期。  相似文献

以采自甘肃省祁连山国家级自然保护区菌物保育区的野生侧耳作为试验材料,通过形态学及系统发育分析方法将其鉴定为冷杉侧耳Pleurotus abieticola。对该菌株生物学特性及栽培条件进行初步研究,结果表明:菌丝体最适生长温度为25℃;最适pH为7.0;最适碳源为玉米粉;最适氮源为豆粉;在以棉籽壳、木屑和麸皮为栽培料时,可获得子实体。  相似文献

采自湖南洞庭湖大通湖区芦苇湿地的野生菌株,经鉴定为田头菇Agrocybe praecox。通过驯化实现人工栽培,栽培原料以芦苇屑为主,使用液体菌种和覆土栽培方式,初潮菇平均生物转化率为45.6%。营养成分分析表明:以芦苇屑为栽培培养基,子实体中矿物元素含量分别为K(2 190mg/kg)、Fe(16.2mg/kg)、Mg(59.7mg/kg)、Ca(33.4mg/kg),必需氨基酸含量占比高达44.85%,谷氨酸、蛋氨酸、天冬氨酸含量高,是一种高蛋白、低脂肪、高钾低钠的食用菌。  相似文献
