叶用芥菜细胞质雄性不育相关基因orf 220的分子特性   总被引:10,自引:0,他引:10  
采用榨菜细胞质雄性系为不育源,通过变种间杂交和回交的方法,转育叶用芥菜细胞质雄性不育种质。根据Brassica nap型细胞质雄性不育相关基因off222设计兼并引物,利用特异引物PCR方法,分别在不育源、F1、BC2以及BC3世代中扩增出一条600~700bp大小的特异条带,而在叶用芥菜保持系中无此条带,具细胞质基因遗传特性。进一步序列分析表明,此特异条带大小为663bp,具有起始密码子和终止密码子,编码220个氨基酸,定名为off220。同时,off220推导的氨基酸序列存在两个跨膜区,氨基端与Oenothera berteriana中的COXⅢ(线粒体细胞色素氧化酶复合体亚基)、萝卜中的ATP8(线粒体ATP酶复合体亚基)以及向日葵中的ORFB(ATP酶复合体α亚基相关)蛋白氨基端高度同源。RT-PCR方法分析表明,off220基因的表达为组成性表达,无器官特异性。  相似文献

An 815 by region of the promoter of the Saccharomyces cerevisiae gene CTR/HNM1, encoding choline permease was sequenced and its regulatory function analysed by deletion studies in an in-frame promoter-lacZ construct. In addition to the TATA box, a 10 by motif (consensus 5-CATGTGAAAT-3) was found to be mandatory for CTR/HNM1 expression. This decamer motif is located between nucleotides –262 and –271 and is identical in 9 of 10 by with the regulatory motif found in the S. cerevisiae INO1 and CHO1 genes. Constructs with the 10 by sequence show high constitutive expression, while elimination or alterations at three nucleotide positions, of the decamer motif in the context of an otherwise unchanged promoter leads to total loss of -galactosidase production. Expression of the CTR/HNM1 gene in wild-type cells is regulated by the phospholipid precursors inositol and choline; no such influence is seen in cells bearing mutations in the phospholipid regulatory genes INO2, INO4, and OPI1. There is no regulation by INO2 and OPI1 in the absence of the decamer motif. However constructs not containing this sequence (promoter intact to positions –213 or –152) are still controlled by INO4. Other substrates of the choline permease, i.e. ethanolamine, nitrogen mustard and nitrogen half mustard do not regulate expression of CTR/HNM1.  相似文献

Two different isolates of Turnip mosaic virus (TuMV: UK 1 and JPN 1) belonging to different virus strains were tested on three different Brassica species, namely turnip (Brassica rapa L.), Indian mustard (Brassica juncea L.) and Ethiopian mustard (Brassica carinata A. Braun). Although all three hosts were readily infected by isolate UK 1, isolate JPN 1 was able to establish a visible systemic infection only in the first two. Ethiopian mustard plants showed no local or systemic symptoms, and no virus antigens could be detected by enzyme‐linked immunosorbent assay (ELISA). Thus, this species looks like a non‐host for JPN 1, an apparent situation of non‐host resistance (NHR). Through an experimental approach involving chimeric viruses made by gene interchange between two infectious clones of both virus isolates, the genomic region encoding the C‐terminal domain of viral protein P3 was found to bear the resistance determinant, excluding any involvement of the viral fusion proteins P3N‐PIPO and P3N‐ALT in the resistance. A further determinant refinement identified two adjacent positions (1099 and 1100 of the viral polyprotein) as the main determinants of resistance. Green fluorescent protein (GFP)‐tagged viruses showed that the resistance of Ethiopian mustard to isolate JPN 1 is only apparent, as virus‐induced fluorescence could be found in discrete areas of both inoculated and non‐inoculated leaves. In comparison with other plant–virus combinations of extreme resistance, we propose that Ethiopian mustard shows an apparent NHR to TuMV JPN 1, but not complete immunity or extreme resistance.  相似文献

Effects of varying intensities of sub-lethal heating were ascertained in improving the efficiency of Brassica amendments and summer irrigation on survival of Macrophomina phaseolina, a dry root rot pathogen. Sub-lethal heating (45–55°C) of M. phaseolina infested dry soil reduced the viable propagules by only 12.8% in a period of 90 days. One summer irrigation without sub-lethal heating caused 33.9% reduction in M. phaseolinapropagules, which improved to 43.3% when it was combined with 60 days of sub-lethal heating. Addition of the Brassicaamendments to irrigated soil resulted in significant reduction (60.4–71.6%) in counts of M. phaseolinabut this reduction improved (89.4–96.1%) when sub-lethal heating was combined with amendments. Mustard oil-cake (0.18% w/w) was found to be the most effective with reduction but a 94% inoculum reduction by mustard pod straw (0.36% w/w) was also achieved at 0–30 cm soil depth under similar conditions. Moderate heat level could not exert detectable weakening effect on M. phaseolinapropagules. These results suggest a practical cultural control of soil-borne pathogens by combining sub-lethal heating, Brassica amendments with one summer irrigation.  相似文献

The invasion of non-indigenous plants is considered one of the primary threats to rare and endangered species as well as to the integrity and function of North American ecosystems. However, many of the suspected negative ecosystem impacts are based on anecdotal evidence. For example, there is almost unanimous agreement among natural resource managers of the detrimental ecological impacts of species such as Lythrum salicaria (purple loosestrife), Phragmites australis (common reed) and Alliaria petiolata (garlic mustard) but convincing documentation is scarce. Experimental and theoretical ecology predicts large ecosystem impacts of the most widespread invasive species. However, it is difficult to prioritize control of species that occur at intermediate densities. Long-term monitoring before and during the invasion as well as before, during and after any control attempts can provide valuable ecological information. In particular, it is important to understand how changes in the abundance of species influence ecosystem properties and processes which, in turn, will help guide management decisions. Ideally, this monitoring has to go beyond 'simple impacts on plant communities, involve cross-disciplinary teams of scientists and should incorporate many different taxa and their interactions. Monitoring design and data collection should be sophisticated enough to allow statistically sound data analysis. The available information will be paramount in (1) developing new political and scientific guidelines in invasive species management, (2) helping resolve potential conflicts of interest and (3) helping change public attitudes regarding growth, sale, and control of non-indigenous species.  相似文献

In our continued effort to develop prodrugs of phosphoramide mustard, conjugates of 4-aminocyclophosphamide (4-NH2-CPA) with three PSA-specific peptides were synthesized and evaluated as substrates of PSA. These include conjugates of cis-(2R,4R)-4-NH2-CPA with a tetrapeptide Succinyl-Ser-Lys-Leu-Gln-OH, a hexapeptide Succinyl-His-Ser-Ser-Lys-Leu-Gln-OH, and a pentapeptide Glutaryl-Hyp-Ala-Ser-Chg-Gln-OH. These conjugates were cleaved by PSA efficiently and exclusively after the expected glutamine residue to release 4-NH2-CPA, the activated prodrug form of phosphoramide mustard. The cleavage was most efficient for the pentapeptide conjugate 3 (Glutaryl-Hyp-Ala-Ser-Chg-Gln-NH-CPA), which showed a half-life of 55 min with PSA, followed by the hexapeptide conjugate 2 (Succinyl-His-Ser-Ser-Lys-Leu-Gln-NH-CPA) and the tertrapeptide conjugate 1 (Succinyl-Ser-Lys-Leu-Gln-NH-CPA) with half-lives of 6.5 and 12 h, respectively. These results indicate a potential of the conjugate 3 as an anticancer prodrug of phosphoramide mustard for selective PSA activation.  相似文献

Plant phenotypic plasticity in response to antagonists can affect other community members such as mutualists, conferring potential ecological costs associated with inducible plant defence. For flowering plants, induction of defences to deal with herbivores can lead to disruption of plant–pollinator interactions. Current knowledge on the full extent of herbivore‐induced changes in flower traits is limited, and we know little about specificity of induction of flower traits and specificity of effect on flower visitors. We exposed flowering Brassica nigra plants to six insect herbivore species and recorded changes in flower traits (flower abundance, morphology, colour, volatile emission, nectar quantity, and pollen quantity and size) and the behaviour of two pollinating insects. Our results show that herbivory can affect multiple flower traits and pollinator behaviour. Most plastic floral traits were flower morphology, colour, the composition of the volatile blend, and nectar production. Herbivore‐induced changes in flower traits resulted in positive, negative, or neutral effects on pollinator behaviour. Effects on flower traits and pollinator behaviour were herbivore species‐specific. Flowers show extensive plasticity in response to antagonist herbivores, with contrasting effects on mutualist pollinators. Antagonists can potentially act as agents of selection on flower traits and plant reproduction via plant‐mediated interactions with mutualists.  相似文献

The inhibitory effects of Cd, Cu, Zn, Pb, and Fe on root elongation, contents of photosynthetic pigments, and metal accumulation in the roots and shoots of Sinapis alba were assessed. On the basis of growth inhibition metals can be arranged in a order Cu > Cd > Fe = Zn > Pb. All the metals, except Fe, were accumulated in significantly higher amount in the roots than in the shoots. Cd, Zn, Cu and Pb reduced chlorophyll a, and especially chlorophyll b content, and Zn and Pb reduced the carotenoid content, but less than that of chlorophyll a+b. The plants contained the highest concentration of Cd, and the lowest concentration of Zn.  相似文献

Abstract Depending on the applied concentration, nitrate can either stimulate or inhibit germination of dormant seeds of Sinapis arvensis L. (wild mustard). Seed NO?3 levels that corresponded to the maximal germination frequencies recorded, ranged from 0.3 to 4.4 nmol seed?1 for applied NO?3 concentrations between 2.5 and 20 mol m?3. Germination was significantly lower in seeds containing more than 5 nmol NO?3. Although the presence of NO?3 within the seed was required to promote germination, seed NO?3 levels were 5 to 15 times less than levels calculated from the volume of solution taken up by the seeds. Seed NO?3 levels also responded in a passive fashion to the external NO3? concentration when the seeds were incubated on filter paper or in soil. In a representative soil containing 26 mg NO?3-N per kg dry weight and 8 to 16% water by weight, the amount of NO?3 taken up by the seeds reached inhibitory levels. Our results suggest that levels of inorganic nitrogen (NO?3, NH+4) in managed soils may play an important role in regulating germination of dormant S. arvensis seeds.  相似文献
