The rationale and a strategy for the estimation of length-weight relationships (LWR) using preserved specimens of less common fish species in museums is presented, along with preliminary results pertaining to 56 specimens and 31 species of fish from the Australian Museum, Sydney, Australia, the Marine Biological Specimen Museum of Chinese Academy of Sciences in Qingdao, China, and the Beaty Biodiversity Museum of the University of British Columbia, Vancouver, Canada. These results are discussed in the context of the effects of seasonality (in fresh specimens) and preservation (in museum specimens) on the estimation of LWR parameters.  相似文献

Invasive species are known to cause environmental and economic damage, requiring management by control agencies worldwide. These species often become well established in new environments long before their detection, resulting in a lack of knowledge regarding their history and dynamics. When new invasions are discovered, information regarding the source and pathway of the invasion, and the degree of connectivity with other populations can greatly benefit management strategies. Here we use invasive common starling ( Sturnus vulgaris ) populations from Australia to demonstrate that genetic techniques can provide this information to aid management, even when applied to highly vagile species over continental scales. Analysis of data from 11 microsatellites in 662 individuals sampled at 17 localities across their introduced range in Australia revealed four populations. One population consisted of all sampling sites from the expansion front in Western Australia, where control efforts are focused. Despite evidence of genetic exchange over both contemporary and historical timescales, gene flow is low between this population and all three more easterly populations. This suggests that localized control of starlings on the expansion front may be an achievable goal and the long-standing practice of targeting select proximal eastern source populations may be ineffective on its own. However, even with low levels of gene flow, successful control of starlings on the expansion front will require vigilance, and genetic monitoring of this population can provide essential information to managers. The techniques used here are broadly applicable to invasive populations worldwide.  相似文献

The white-headed duck is a globally threatened species native to the Palaearctic with a range extending from Spain in the west to the western edge of China in the east. Its populations have become fragmented and undergone major declines in recent decades. To study genetic differences between populations across the range and change in genetic diversity over time, we sequenced a portion of the mitochondrial DNA control region from 67 museum specimens (years 1861–1976) as well as 39 contemporary samples from Spain and seven from Greece (years 1992–2003). In the historical sample, we found a lack of significant genetic structure between populations in different areas. We found evidence that the species experienced a rapid expansion in the past, perhaps from glacial refugia centred around the Mediterranean following the last ice age. In Spain, the population went through a dramatic bottleneck in the 1970s and early 1980s, when only a few dozens individuals remained in the wild. Although population size has since recovered to a few thousand individuals, we found a highly significant loss of mitochondrial haplotype diversity between the historical and contemporary samples. Given ongoing declines in other areas, losses in genetic diversity that may reduce the adaptive potential of white-headed ducks in the future are a continuing concern throughout the geographic range of this species.  相似文献

We describe the upper portion of the bill sheath (rhinotheca) of the kākāpō (Strigops habroptilus) from three adult female specimens. The external buccal surface of the rhinotheca is deeply concave with a prominent palatal stop and hardened chevrons creating a ‘milling apparatus’ that the kākāpō uses to grind food. The palatal stop presents a working face of 40–50?mm2. The internal surface of the rhinotheca mirrors the overlying premaxilla and provides a distinct thickened abutment consistent with resistance against the increased workload of the mandibles (gnathotheca) due to the kākāpō’s fibrous diet and chewing style. Along the midline, the rhinotheca at the abutment is up to 5.6?mm thick, compared with as thin as 2.1?mm elsewhere on the midline. The closely related Nestor parrots have less developed palatal stops, chevrons and abutments on their rhinothecas consistent with their lower preference for fibrous plant material. The form of the rhinotheca agrees with the kākāpō’s feeding ecology as a generalist herbivore that grinds locally available fibrous material to assist digestion.  相似文献

Natural history museums play a significant role in educating the general public about evolution. This article describes Explore Evolution, one of the largest evolution education projects funded by the National Science Foundation. A group of regional museums from the Midwestern United States worked with leading evolutionary scientists to create multiple permanent exhibit galleries and a curriculum book for youth. This program invites the public to experience current evolutionary research on organisms that range in size from HIV to whales. Learning research is being conducted on museum visitors to understand how they reason about evolution and to determine what influences the process of conceptual change.  相似文献

The genetic variability and distribution of Amazonian fish species have likely been influenced by major disturbance events in recent geological times. Alternatively, the great diversity of aquatic habitat in the Amazon is likely to shape ongoing gene flow and genetic diversity. In this context, complex patterns of genetic structure originating from a joint influence of historical and contemporary gene flow are to be expected. We explored the relative influence of Pleistocene climatic fluctuations and current water chemistry on the genetic structure of a piranha, Serrasalmus rhombeus, in the Upper Amazon by the simultaneous analysis of intron length polymorphism and mitochondrial DNA sequences. The Madeira river is well suited for that purpose as it is characterized by a great diversity of water types, the presence of one of the largest floodplain of the Amazon and the potential occurrence of two Pleistocene refuges. We found evidence of genetic structure even at a small geographical scale (less than 10 km), indicating that the floodplain is not a homogenizing factor promoting interdrainage dispersal in S. rhombeus. Likewise, the hierarchical genetic structure inferred was correlated to geographical distance instead of habitat characteristic. Our results also support the hypothesis that the area underwent population expansion during the last 800 000 years. In addition, a higher level of genetic diversity was found in the samples from the putative Aripuanã refuge. The present findings suggest that Pleistocene refuges contributed significantly to the colonization of the lowlands in the Upper Amazon valley during the Pleistocene.  相似文献

ABSTRACT

Museums are our memory banks. They tell us where we have come from. They also allow us to imagine where we are heading. Which is why it should trouble us that there has never been a truly Australian museum. Each of our state and federal museums has been built on Aboriginal collections, and each has been built on distinctly Western or European concepts, values, categories and practices. Some of these are unavoidable, but are they all? Are we too far down the path to restump the foundations of our institutions and the narratives they perpetuate in public life? The South Australian Museum holds one of the most important collections of Aboriginal material culture in the world. It is, therefore, given the story it can tell about ancient and enduring cultures, one of the most important collections of human heritage on our planet. What we do with such collections, and what we don’t, defines us. This is the great challenge of contemporary custodianship. These collections are calling us out. In this lecture I examine the South Australian Museum’s response to this challenge. Over the past two years our Museum has undertaken a comprehensive rethink of our policies and practices and the politics of both. We are also transforming the way we work with Aboriginal communities and custodians. This is not simply a question of how collections are displayed or exhibitions are developed. We are rethinking the terms of our custodianship, and the kind of truly Australian Museum that could evolve around those new foundations.  相似文献

It would not be an exaggeration to say that among molecular technologies, it is PCR (polymerase chain reaction) that underpins the discipline of molecular ecology as we know it today. With PCR, it has been possible to target the amplification of particular fragments of DNA, which can then be analysed in a multitude of ways. The capability of PCR to amplify DNA from a mere handful of copies further means that conservationists and ecologists are able to sample DNA unobtrusively and with minimal disturbance to the environment and the organisms of interest. However, a key disadvantage of PCR‐based methods has been the necessity for a generally non‐portable, laboratory setting to undertake the time‐consuming thermocycling protocols. LAMP (loop‐mediated isothermal amplification) offers a logistically simpler protocol: a relatively rapid DNA amplification reaction occurs at one temperature, and the products are visualized with a colour change within the reaction tubes. In the first field application of LAMP for an ecological study, Centeno‐Cuadros et al. ( 2016 ) demonstrates how LAMP can be used to determine the sex of three raptor species. By enabling DNA amplification in situ and in ‘real‐time’, LAMP promises to revolutionize how molecular ecology is practised in the field.  相似文献

Despite advances that allow DNA sequencing of old museum specimens, sequencing small‐bodied, historical specimens can be challenging and unreliable as many contain only small amounts of fragmented DNA. Dependable methods to sequence such specimens are especially critical if the specimens are unique. We attempt to sequence small‐bodied (3–6 mm) historical specimens (including nomenclatural types) of beetles that have been housed, dried, in museums for 58–159 years, and for which few or no suitable replacement specimens exist. To better understand ideal approaches of sample preparation and produce preparation guidelines, we compared different library preparation protocols using low amounts of input DNA (1–10 ng). We also explored low‐cost optimizations designed to improve library preparation efficiency and sequencing success of historical specimens with minimal DNA, such as enzymatic repair of DNA. We report successful sample preparation and sequencing for all historical specimens despite our low‐input DNA approach. We provide a list of guidelines related to DNA repair, bead handling, reducing adapter dimers and library amplification. We present these guidelines to facilitate more economical use of valuable DNA and enable more consistent results in projects that aim to sequence challenging, irreplaceable historical specimens.  相似文献

Although least chipmunks (Neotamias minimus) are a widely distributed North American species of least concern, the southernmost population, N. m. atristriatus (Peñasco least chipmunk), is imperiled and a candidate for federal listing as a subspecies. We conducted a phylogeographic analysis across the N. minimus range to assess genomic differentiation and distinctiveness of the N. m. atristriatus population. Additionally, we leveraged the historical component of sampling to conduct a temporal analysis of N. minimus genetic diversity and also considered climate change effects on range persistence probability by projecting a species distribution model into the IPCC5 RCP 2.6 and 8.5 scenarios. We identified three geographically structured groups (West, North, and South) that were supported by both mitochondrial and nuclear data. N. m. atristriatus grouped within a unique South subclade but were not reciprocally monophyletic from N. m. operarius, and nuclear genome analyses did not separate N. m. atristriatus, N. m. caryi, and N. m. operarius. Thus, while least chipmunks in the Southwest represent an evolutionary significant unit, subspecies distinctions were not supported and listing of the Peñasco population as a Distinct Population Segment of N. m. operarius may be warranted. Our results also support consideration of populations with North and West mitogenomes as two additional evolutionary significant units. We found that N. minimus genetic diversity declined by ~87% over the last century, and our models predicted substantial future habitat contraction, including the loss of the full contemporary ranges of N. m. atristriatus, N. m. arizonensis, and N. m. chuskaensis.  相似文献
