Protein liquid-liquid phase separation drives the dynamic assembly of membraneless organelles for fulfilling different physiological functions. Under diseased condition, protein may undergo liquid-to-solid condensation to form pathological amyloid aggregates closely associated with neurodegenerative diseases. Chemical probe serves as an important chemical tool not only for exploring the basic principle of the dynamic assembly of different protein condensates in vitro and in cell but also for clinical diagnosis and therapeutics of the related diseases. In this review, we first introduce chemical probes to image and regulate protein condensates. Then, we summarized three different categories of chemical probes including general amyloid dye, selective positron emission tomography tracer, and disaggregating binder, which feature distinct interaction pattern and activity upon binding to different pathological amyloid fibrillar aggregates. Next, we discuss the development of chemical probes for tracking protein amorphous aggregates in cells. Finally, we point out future direction in expanding the probes’ chemical space and applications.  相似文献

Summary DNA fingerprints generated by the Jeffreys' probes, 33.6 and 33.15, indicated the presence of minisatellite-like sequences in the red clover genome. The fingerprints generated by probe 33.6 gave less background and fewer but better defined bands than those obtained with probe 33.15. Assay of a regenerative somaclonal variant (F49R) by DNA fingerprinting with probe 33.6 detected mutation that was unlinked to the regenerative trait. The fingerprints obtained under the applied conditions also demonstrated genetic stability of consecutive generations of the regenerants in tissue culture. DNA fingerprints of F1 plants revealed that each polymorphic band was inherited from either one or the other parent. Both probes distinguished individual-specific genotypes in seven cultivars of red clover. Greater variability in DNA fingerprints was detected between (V=0.899) than within (0.417≤V≤0.548) cultivars.  相似文献

Summary A polyphasic taxonomic study was undertaken to establish the genetic and phenotypic relationships among six actinomycetes that produce the immunosuppressant macrolides FK506, FK520/FK523 and rapamycin. Chemotaxonomic studies reveal that all have Type I cell walls. Gas chromatography (GC) of fatty acid methyl esters revealed patterns consistent for strains ofStreptomyces with 160 and 150anteiso predominating. Principal component analysis of GC data revealed distinct profiles for each culture. Reciprocal DNA homology studies atT m -25 showed the rapamycin-producing strain and one FK506-producing strain to have 38–50% homology with the type strain ofStreptomyces hygroscopicus (ATCC 27438). The remaining strains exhibited 6–17% homology. To further explore the relationships among these strains all were probed for the presence of anO-methyltransferase gene specific to this biosynthetic pathway. Among the strains of interest, onlyStreptomyces hygroscopicus subsp.yakushimaensis, the patent strain for FK520/FK523, failed to hybridize with the probes.  相似文献

潘氏闭壳龟(Cuora pani)是中国特有种,国家二级重点保护野生动物。由于非法贸易加之栖息地破坏等,其野外种群数量十分稀少。本研究旨在利用线粒体基因与核基因标记揭示潘氏闭壳龟的遗传多样性和遗传结构,以期为中国潘氏闭壳龟的种群保护和管理提供科学依据。本次共研究14只潘氏闭壳龟个体,其中,6只原产地为四川广元,8只产地未知。结果表明,在14只潘氏闭壳龟样本中鉴定出Cyt b单倍型、ND4单倍型和R35单倍型各2个,且均有1个新单倍型;Cyt b、ND4和R35基因单倍型多样性分别为0.440、0.143和0.154,核苷酸多样性分别为0.000 41、0.000 19和0.000 63;各单倍型的平均遗传距离(p)均小于0.01。基于三个基因联合数据集的系统发育分析表明,潘氏闭壳龟与金头闭壳龟(C.aurocapitata)互为单系进化支,且中性检测和核苷酸错配分析显示,潘氏闭壳龟群体近期历史上可能未经历群体扩张事件。潘氏闭壳龟群体遗传多样性低,种内变异小,群体应对环境变化的能力低,建议应加强对中国潘氏闭壳龟野外种群的基础研究和保护力度,同时规范人工繁殖,以避免近亲繁殖和种群衰退。  相似文献

气囊(gas vesicles,GVs)是一种存在于蓝藻及古菌等微生物中调节浮力的类细胞器纳米结构,由蛋白质外壳包裹气体组成。近年来的研究表明,气囊具有作为超声分子影像探针的潜力。然而,气囊的充放气机制并不明确,限制了生物合成超声分子影像探针的保存和气体更换。本研究发现环境pH值是调节气囊充放气的一个重要因素。其不仅可以调节藻细胞内的气囊充放气进而使微囊藻呈现不同的漂浮状态,还可对提纯的气囊充放气进行体外调节,且该调节过程可逆。该机制的阐明为生物合成超声分子影像探针的大规模生产和保存,特别对气囊中的气体进行更换以满足不同的诊疗需求提供了技术支持,助力生物合成超声造影剂在疾病诊疗中的应用。  相似文献

Efforts to detect effects of cytoplasmic genes are confounded by the problem of partitioning nuclear and cytoplasmic effects. In this study we test for effects of mtDNA haplotype on development in hybrid populations of cutthroat trout (Oncorhynchus clarki) with randomly associated nuclear and mtDNA genotypes. We have previously described several intraspecific hybrid swarms formed by interbreeding of westslope cutthroat trout (O. c. lewisi) and Yellowstone cutthroat trout (O. c. bouvieri). Genetic distance between these subspecies is high (Nei's D = 0.30; mtDNA P = 0.02), and diagnostic alleles at multiple nuclear loci and two distinct mtDNA haplotypes are present in the hybrids. Historical associations between alleles at nuclear loci and between cytotypes and nuclear alleles have largely decayed. We test for differences in meristic characters between fish with alternate mtDNA haplotypes. Counts and fluctuating bilateral asymmetry for these characters have been previously shown to be sensitive indicators of genetic differences that affect development. No differences were found between mtDNA types in meristic counts or fluctuating asymmetry. Therefore, the alternate mtDNA haplotypes had no detectable effect on development as measured by meristic counts in these hybrid populations. However, diagnostic alleles at one nuclear allozyme locus (CK-CI) were associated with several fin ray counts.  相似文献

The emergence and acceptance of the concept of sustainable development warrants that the scope of environmental biotechnology be enlarged to address issues like environmental monitoring, restoration of environmental quality, resource/residue/waste-recovery/utilization/treatment, and substitution of the non-renewable resource base with renewable resources. Variation in the mitochondrial cytochrome b gene (nucleotide and amino acid sequences) is evaluated for 9 genera and 15 species of American opossums in the family Didelphidae, using the American caenolestid rat opossumLestoros and the New Guinean peroryctid bandicootEchimypera as outgroups. Phylogenetic analyses (parsimony and distance) strongly support the monophyly of the Didelphidae and delineate two major clades; (1)Didelphis andPhilander are strongly aligned sister taxa, withMetachirus weakly but consistently associated with them, and (2)Marmosa plusMicoureus, withMonodelphis falling outside that pair. The generaMarmosops, Caluromys, andGlironia exhibit varied relationships, depending upon the method of analysis and data (DNA or amino acid sequences) used, but generally are placed individually or in combinations near or at the base of the didelphid radiation. Some aspects of these relationships are consistent with current taxonomic views, but others are in marked contrast. Specifically, a clade comprised of the mouse opossumsMarmosa, Micoureus, andMarmosops is strongly rejected by log-likelihood analysis, contrary to expectations from some current classifications. Also, the woolly opossumsCaluromys andGlironia also do not form a sister-taxon relationship, as suggested by their placement in a subfamily separate from the remaining didelphids examined. However, such a relationship cannot be rejected from log-likelihood analyses. The relationships suggested fromcyt-b sequences are strongly concordant with those based on DNA-DNA hybridization analyses. In addition to systematic and phylogenetic properties, molecular evolution of the didelphid cytochrome b gene sequence is characterized according to nucleotide bias and rate differentials at each codon position and across the entire sequence.To whom correspondence should be addressed.  相似文献

本文研究了果蝇D.virilis种群D.virilis线粒体DNA(mitochondrialDNA,mtDNA)的多态性。用9种限制性内切酶XbaⅠ,EcoRⅠ,PstⅠ,HindⅢ,BglⅡ,SacⅠ,ScaⅠ,EcoRV和PuvⅡ,对青岛、南京、上海、宁波与泉州5个D.virilis群体的mtDNA进行了限制性片段长度多态性(restrictionfragmentslengthpolymorphism,RFLP)的研究。在5群体中,发现5种不同的酶切图谱,它们彼此之间的遗传差异π为0.46%-1.76%,群体内遗传差异πij为0.00%-0.33%,群体间的差异dxy,为0.00%-0.82%。分布于中国大陆的D.virilis的群体间遗传差异在总遗传差异中所占比例γst值为24.62%。我们发现,D.virilis的栖息环境对mtDNA的遗传变异有十分明显的影响,而不同地理纬度的群体之间其遗传距离并无倾群(cline)表现。  相似文献

In the bivalve mollusc Pecten maximus, the size of the mitochondrial DNA molecules ranges from 20 to 25.8 kbp. This variability is mainly correlated with the occurrence of a variable domain composed with two to five 1.6-kbp repeated units tandemly arrayed in the genome. DNA fragments spanning the 1,586-base-pair-long repeated element and the nearest flanking gene have been cloned and sequenced. This sequence was analyzed regarding its base composition and potential secondary structures. The repeated unit domain was positioned and oriented with regard to the known flanking gene. It ends 2 base pairs upstream relative to the beginning of the tRNAgly gene. The peculiar properties of the repeated unit were compared with those of the 1,442-bp repeated element found in the mitochondrial genome of the deep sea scallop Placopecten magellanicus. This comparison provided evidence for the absence of nucleotide conservation, except for a small sequence engaged in a secondary structure, but argued for a strong pressure maintaining domains with specific nucleotide content.
