目的研究c-myc基因在人完全性葡萄胎中的表达及其意义。方法取人完全性葡萄胎30例,正常早孕流产标本10例,用SABC免疫组织化学染色方法,检测c-myc基因在两种组织中的表达情况,并采用图像分析技术,对正常早孕绒毛组和完全性葡萄胎组c-myc的表达情况进行对比分析。结果与正常绒毛相比,c-myc基因在完全性葡萄胎组织中的表达量和表达的空间特异性有明显不同。结论 c-myc基因可能与完全性葡萄胎的发生密切相关。 相似文献

Despite the acknowledged importance of the locomotory and respiratory functions associated with hypaxial musculature in salamanders, variation in gross morphology of this musculature has not been documented or evaluated within a phylogenetic or ecological context. In this study, we characterize and quantify the morphological variation of lateral hypaxial muscles using phylogenetically and ecologically diverse salamander species from eight families: Ambystomatidae (Ambystoma tigrinum), Amphiumidae (Amphiuma tridactylum), Cryptobranchidae (Cryptobranchus alleganiensis), Dicamptodontidae (Dicamptodon sp.), Plethodontidae (Gyrinophilus porphyriticus), Proteidae (Necturus maculosus), Salamandridae (Pachytriton sp.), and Sirenidae (Siren lacertina). For the lateral hypaxial musculature, we document 1) the presence or absence of muscle layers, 2) the muscle fiber angles of layers at mid‐trunk, and 3) the relative dorsoventral positions and cross‐sectional areas of muscle layers. Combinations of two, three, or four layers are observed. However, all species retain at least two layers with opposing fiber angles. The number of layers and the presence or absence of layers vary within species (Necturus maculosus and Siren lacertina), within genera (e.g., Triturus), and within families. No phylogenetic pattern in the number of layers can be detected with a family‐level phylogeny. Fiber angle variation of hypaxial muscles is considerable: fiber angles of the M. obliquus externus range from 20–80°; M. obliquus internus, 14–34°; M. transversus abdominis, 58–80° (acute angles measured relative to the horizontal septum). Hypaxial musculature comprises 17–37% of total trunk cross‐sectional area. Across taxa, individuals vary in how far they disperse, with most individuals staying close to their origin and fewer dispersing long distances. Costs associated with dispersal (e.g., energy, risk) are widely believed to trade off with benefits (e.g., reduced competition, increased reproductive success) to influence dispersal propensity. However, this framework has not been applied to understand variation in dispersal distance, which is instead generally attributed to extrinsic environmental factors. We alternatively hypothesized that variation in dispersal distances results from trade‐offs associated with other aspects of locomotor performance. We tested this hypothesis in the stream salamander Gyrinophilus porphyriticus and found that salamanders that dispersed farther in the field had longer forelimbs but swam at slower velocities under experimental conditions. The reduced swimming performance of long‐distance dispersers likely results from drag imposed by longer forelimbs. Longer forelimbs may facilitate moving longer distances, but the proximate costs associated with reduced swimming performance may help to explain the rarity of long‐distance dispersal. The historical focus on environmental drivers of dispersal distances misses the importance of individual traits and associated trade‐offs among traits affecting locomotion. 相似文献

Background
Although the reference human genome sequence was declared finished in 2003, some regions of the genome remain incomplete due to their complex architecture. One such region, 1q21.1-q21.2, is of increasing interest due to its relevance to human disease and evolution. Elucidation of the exact variants behind these associations has been hampered by the repetitive nature of the region and its incomplete assembly. This region also contains 238 of the 270 human DUF1220 protein domains, which are implicated in human brain evolution and neurodevelopment. Additionally, examinations of this protein domain have been challenging due to the incomplete 1q21 build. To address these problems, a single-haplotype hydatidiform mole BAC library (CHORI-17) was used to produce the first complete sequence of the 1q21.1-q21.2 region.Results
We found and addressed several inaccuracies in the GRCh37sequence of the 1q21 region on large and small scales, including genomic rearrangements and inversions, and incorrect gene copy number estimates and assemblies. The DUF1220-encoding NBPF genes required the most corrections, with 3 genes removed, 2 genes reassigned to the 1p11.2 region, 8 genes requiring assembly corrections for DUF1220 domains (~91 DUF1220 domains were misassigned), and multiple instances of nucleotide changes that reassigned the domain to a different DUF1220 subtype. These corrections resulted in an overall increase in DUF1220 copy number, yielding a haploid total of 289 copies. Approximately 20 of these new DUF1220 copies were the result of a segmental duplication from 1q21.2 to 1p11.2 that included two NBPF genes. Interestingly, this duplication may have been the catalyst for the evolutionarily important human lineage-specific chromosome 1 pericentric inversion.Conclusions
Through the hydatidiform mole genome sequencing effort, the 1q21.1-q21.2 region is complete and misassemblies involving inter- and intra-region duplications have been resolved. The availability of this single haploid sequence path will aid in the investigation of many genetic diseases linked to 1q21, including several associated with DUF1220 copy number variations. Finally, the corrected sequence identified a recent segmental duplication that added 20 additional DUF1220 copies to the human genome, and may have facilitated the chromosome 1 pericentric inversion that is among the most notable human-specific genomic landmarks. 相似文献36.

We investigate kin-structured migration and its effects on patterns of genetic variation in cytoplasmic and nuclear genomes. We show that colonization events involving close relatives, such as those that might characterize range expansion and the invasion of new habitats, can change the patterns of nuclear and cytoplasmic genetic variation expected at equilibrium. The difference in the patterns of variation between the nuclear and mitochondrial genomes suggests that it may be possible to estimate the time of the effective kin-structured colonization event. Summary Survivorship and growth of larval tiger salamanders, Ambystoma tigrinum nebulosum, in the White Mountains of east-central Arizona were compared in six microhabitats using field enclosures during summers, 1983–85. Microhabitats were vegetated and nonvegetated shallows, surface, middle, and bottom horizontal layers of limnetic areas, and the vertical limnetic water column. Initial enclosure densities (0.025 larvae per 1) were identical among microhabitats. Three enclosures were placed in each microhabitat in two ponds. Larval survivorship and growth were usually greatest in vegetated shallows in lowest in middle and bottom limnetic enclosures, despite several population dieoffs. Lower fitness, as reflected in survivorship and growth, in these latter enclosures was correlated with lower food levels, temperatures, and oxygen concentrations in deeper limnetic areas. Relative fitness varied greatly between years while food levels, temperatures, and oxygen concentrations within microhabitats remained relatively constant indicating additional factors affected fitness. Disparities in fitness between microhabitats apparently affect habitat use patterns of larvae. 相似文献

Mexico, a rich country in terms of amphibian diversity, hosts about 375 described species. Population declines have been documented
for several species where it is evident that their habitat is being destroyed or modified. However, other species which inhabit
pristine areas are declining as well. It has been suggested that the chytrid fungus Batrachochytrium dendrobatidis
(B.d.) may be one of the causes of the enigmatic declines in Mexico. We surveyed a total of 45 localities, in 12 states across Mexico,
examining a total of 360 specimens representing 14 genera and 30 species. We also examined 91 specimens of Ambystoma mexicanum from a captive population in Mexico City as well as one Pachymedusa dacnicolor obtained in a pet shop. We used a two-tiered technique to detect the pathogen. For wild-caught specimens, we utilized light
microscopy to identify presence of B.d. sporangia in amphibian skin. Then, to verify the infection, we used a quantitative real-time PCR assay on collected skin
sections which is specific for B.d. For captive animals, we used a nonlethal version of the real-time PCR technique. We found evidence of B.d. infection in 111 animals comprising 14 species in 13 localities. A large percentage (84%) of Ambystoma mexicanum from the colony were infected with B.d. The two most highly infected individuals were the endangered Ambystoma mexicanum, from a captive colony, and Pachymedusa dacnicolor, purchased at a pet shop.
