【目的】为了明确熊蜂蜂群发育过程中具有代表性的性状指标及其饲料花粉最优配比。【方法】以山杏Armeniaca sibirica花粉、山柳Salix caprea花粉和油菜Brassica rapa花粉为原料,通过{3,3}混料设计得到7种配比的混合花粉,分析不同混合花粉对地熊蜂Bombus terrestris(L.)无王工蜂群的产卵前时间、幼虫拖出数、幼虫总数量、幼虫总重量、蛹总数量、蛹总重量、雄蜂出房时间、出房雄蜂数量和雄蜂出生重9个蜂群性状指标的影响,使用主成分分析对蜂群性状指标进行综合评价,并通过混料回归模型预测熊蜂蜂群饲养过程中的最优花粉配比。【结果】地熊蜂无王工蜂群发育过程的9个性状指标可以归纳为幼虫发育、蛹发育、成蜂发育、蜂群发育周期4类评价因子,其中幼虫总数量、蛹总重量、雄蜂出生重、产卵前时间和雄蜂出房时间是5个主要性状指标;以蜂群主要性状指标为评价依据,得出蜂群饲养过程中最优花粉配比:当以油菜花粉单独饲喂蜂群时蜂群产卵前时间短、幼虫总数量最多、蛹总重量最大,当山杏花粉、山柳花粉和油菜花粉以1︰1.5︰1.5比例饲喂蜂群时雄蜂出房时间最短,当山柳花粉和油菜花粉以3︰1比例饲喂蜂群时雄蜂出生重最大。【结论】明确了地熊蜂无王工蜂群发育过程中的重要性状指标和饲料花粉最优配比,为进一步研究商品化熊蜂群不同发育阶段的营养需求奠定了基础。  相似文献

Cumulative risk assessments (CRAs) include the examination of risks posed by multiple stressors and include population-specific vulnerabilities and susceptibilities. In this case study, we assess potential hearing impairment hazard due to joint exposure from noise and volatile organic compounds (VOCs) in order to examine the strengths and limitations of using secondary data on exposure and health effects for a CRA. Block group-level noise categories were estimated using modeled street-level data. A quantile regression model of sociodemographic and personal predictors from the 1999–2000 U.S. National Health and Nutrition Examination Survey VOC dataset was used along with block group-level sociodemographic and personal variables to estimate VOC exposures. Hazard indices (HIs) for potential hearing impairment due to joint noise and VOC exposures were calculated. County-averaged HIs for hearing impairment ranged from 0.8 (10th total VOCs percentile and 45–60 dB) to 1.7 (90th total VOCs percentile and 71–75 dB). Limitations of the exposure and health effects data included issues combining heterogeneous data and a lack of established threshold levels for combined low-level exposures; yet, this case study illustrates that screening-level CRAs, including non-chemical stressors, can be accomplished with publicly available data and existing methods.  相似文献

The enantioresolution of racemic alcohols as esters of 2-methoxy-2-(1-naphthyl)propionic acid (MalphaNP acid 1) and the determination of their absolute configurations on the basis of (1)H NMR anisotropy effect are described. The enantiopure MalphaNP acid (S)-(+)-1 was allowed to react with racemic 2-alkanols and 1-octyn-3-ol, yielding diastereomeric mixtures of esters, which were easily separated by HPLC on silica gel. To determine the absolute configurations of the first-eluted diastereomeric esters by the (1)H NMR anisotropy method, the general scheme was proposed. Separated esters were reduced with LiAlH(4) or hydrolyzed with KOH/EtOH to recover enantiopure alcohols.  相似文献

Perennial grass mixtures established on Conservation Reserve Program (CRP) lands can be an important source of feedstock for bioenergy production. This study aimed to evaluate management practices for optimizing the quality of bioenergy feedstock and stand persistence of grass-legume mixtures under diverse environments. A 5-year field study (2008–2012) was conducted to assess the effects of two harvest timings (at anthesis vs after complete senescence) and three nitrogen (N) rates (0, 56, 112 kg N ha−1) on biomass chemical compositions (i.e., cell wall components, ash, volatiles, total carbon, and N contents) and the feedstock energy potential, examined by the theoretical ethanol yield (TEY) and the total TEY (i.e., the product of biomass yield and TEY, L ha−1), of cool-season mixtures in Georgia and Missouri and a warm-season mixture in Kansas. The canonical correlation analysis (CCA) was used to investigate the effect of vegetative species transitions on feedstock quality. Although environmental variations (mainly precipitation) greatly influenced the management effect on chemical compositions, the delayed harvest after senescence generally improved feedstock quality. In particular, the overall cell wall concentrations and TEY of the warm-season mixtures increased by approximately 7%. Additional N supplies improved the total TEY of both mixtures by ~1.6–4.2 L ha−1 per 1.0 kg N ha−1 input but likely lowered the feedstock quality, particularly for the cool-season mixture. The cell wall concentrations of cool-season mixture reduced by approximately 3%–6%. The CCA results indicated that the increased legume compositions (under low N input) likely enhanced lignin but reduced ash concentrations. This field research demonstrated that with proper management, grass-legume mixtures on CRP lands can provide high-quality feedstock for bioenergy productions.  相似文献

A model to calculate bone resorption driven by fluid flow at the bone–soft tissue interface is developed and used as a basis for computer calculations, which are compared to experiments where bone is subjected to fluid flow in a rat model. Previous models for bone remodelling calculations have been based on the state of stress, strain or energy density of the bone tissue as the stimulus for remodelling. We believe that there is experimental support for an additional pathway where an increase in the amount of the cells directly involved in bone removal, the osteoclasts, is caused by fluid pressure, flow velocity or other parameters related to fluid flow at the bone–soft tissue interface, resulting in bone resorption.  相似文献

Summary In vitro pollen germination of cultivated tomato, Lycopersicon esculentum Mill., is inhibited by an ambient temperature of 5°C, more so than pollen from a Peruvian ecotype of Lycopersicon hirsutum Humb. & Bonpl. originating from an altitude of 3200 m. The frequency of L. hirsutum gametes contributing to hybrid zygote formation is more than doubled when controlled fertilizations with pollen mixtures of the two species occurs at 12/6°C as compared to crosses with the same mixtures at 24/19°C. The results suggest that differential selection at the gametophytic level occurs in response to low temperature regimes. To our knowledge this is the first time in higher plants that alteration of an environmental factor has been demonstrated to change selection values of male gametophytes in a fashion predicted by the ecology of the parental sporophytes.  相似文献

1. To assess the impact of metal mixtures on microbial decomposition of leaf litter, we exposed leaves previously immersed in a stream to environmentally realistic concentrations of copper (Cu) and zinc (Zn) (three levels), alone and in all possible combinations. The response of the microbial community was monitored after 10, 25 and 40 days of metal exposure by examining leaf mass loss, fungal and bacterial biomass, fungal reproduction and fungal and bacterial diversity.
2. Analysis of microbial diversity, assessed by denaturing gradient gel electrophoresis and identification of fungal spores, indicated that metal exposure altered the structure of fungal and bacterial communities on decomposing leaves.
3. Exposure to metal mixtures or to the highest Cu concentration significantly reduced leaf decomposition rates and fungal reproduction, but not fungal biomass. Bacterial biomass was strongly inhibited by all metal treatments.
4. The effects of Cu and Zn mixtures on microbial decomposition of leaf litter were mostly additive, because observed effects did not differ from those expected as the sum of single metal effects. However, antagonistic effects on bacterial biomass were found in all metal combinations and on fungal reproduction in metal combinations with the highest Cu concentrations, particularly at longer exposure times.  相似文献

This study describes the design, synthesis, and use of selective peptide substrates for cysteine peptidases of the C1 papain family, important in many biological processes. The structure of the newly synthesized substrates is Glp-Xaa-Ala-Y (where Glp = pyroglutamyl; Xaa = Phe or Val; and Y = pNA [p-nitroanilide], AMC [4-amino-7-methylcoumaride], or AFC [4-amino-7-trifluoromethyl-coumaride]). Substrates were synthesized enzymatically to guarantee selectivity of the reaction and optical purity of the target compounds, simplifying the scheme of synthesis and isolation of products. The hydrolysis of the synthesized substrates was evaluated by C1 cysteine peptidases from different organisms and with different functions, including plant enzymes papain, bromelain, ficin, and mammalian lysosomal cathepsins B and L. The new substrates were selective for C1 cysteine peptidases and were not hydrolyzed by serine, aspartic, or metallo peptidases. We demonstrated an application of the selectivity of the synthesized substrates during the chromatographic separation of a multicomponent set of digestive peptidases from a beetle, Tenebrio molitor. Used in combination with the cysteine peptidase inhibitor E-64, these substrates were able to differentiate cysteine peptidases from peptidases of other classes in midgut extracts from T. molitor larvae and larvae of the genus Tribolium; thus, they are useful in the analysis of complex mixtures containing peptidases from different classes.  相似文献

In the present study, bovine milk xanthine oxidase activity in various aqueous-organic mixtures and the effects of pH, temperature, and lyophilization on the enzyme activity have been investigated. The enzyme was incubated with xan-thine as the substrate in Sorenson’s phosphate buffer (pH 7.0) containing 0.1 mM EDTA, and the activity was determined spectrophotometrically in the absence and presence of different fractions of nine water-miscible organic solvents at 27–50°C and at different pH values ranging from 6 to 9. The organic solvents reduced the enzyme activity to different extents. In spite of these inhibitory effects, the enzyme showed relatively good stability in the aqueous-organic mixtures compared with the aqueous medium. Toxicities of binary mixtures of Cu2+, Cd2+, benzo(a)pyrene [B(a)P] andN-ethylmaleimide (NEM) were screened using thein vitro neutral red (NR) assay to test the hypothesis that combined toxicity is more than or less than additive relative to the influence of each mixture constituent on toxicant uptake and brown cell lysosomal membrane stability. Significant cytotoxicity was observed at 25 mol/L Cu2+, 500 mol/L Cd2+ and 25 mol/L NEM. B(a)P at 12 mol/L exerted no toxicity under the conditions of the assay. Interactions between Cu2+ and NEM, between Cd2+ and NEM, and between Cd2+ and B(a)P significantly influenced brown cell survival. Comparison of observed joint toxicity with estimates made using a model of independent joint action indicates that interactive effects are less than additive in character. The 3-way interaction involving Cu2+, B(a)P, and NEM also affected brown cell survival to a statistically significant degree. However, the interactive cytotoxicity of this mixture is attributable mainly to the combined effect of Cu2+ and NEM.
