Outcome-dependent sampling designs have been shown to be a cost-effectiveway to enhance study efficiency. We show that the outcome-dependentsampling design with a continuous outcome can be viewed as anextension of the two-stage case-control designs to the continuous-outcomecase. We further show that the two-stage outcome-dependent samplinghas a natural link with the missing-data and biased-samplingframeworks. Through the use of semiparametric inference andmissing-data techniques, we show that a certain semiparametricmaximum-likelihood estimator is computationally convenient andachieves the semiparametric efficient information bound. Wedemonstrate this both theoretically and through simulation.  相似文献

Post-translational modification of proteins by the small ubiquitin-like modifiers (SUMOs) has emerged as an important regulatory mechanism for alteration of protein activity, stability, and cellular localization. It has been reported that SUMOylation plays an important role in some activities of neuronal cells. However, the link between SUMOylation and activity-dependent stimuli of neurons remains to be elucidated. Here we showed that KCl-induced depolarization increased SUMO conjugation in SHSY5Y cell line in a time-dependent manner. The increase of SUMOylation was largely dependent on calcium influx and intracellular calcium signals. Our study demonstrates the link between the activity-dependent stimuli and global SUMOs conjugation, which may play an important role in activity-dependent signals of neurons.  相似文献

The application of enantioseparation methods alone can only yield up to 50% of the desired chiral product. Thus enantioseparation becomes more attractive when accompanied by the racemization of the counter‐enantiomer. Here we present first results of dynamic kinetic resolution of L ‐asparagine (L ‐Asn) via preferential crystallization and enzymatic racemization from a racemic, supersaturated solution on a 20 mL scale. An enzyme lyophilisate (WT amino acid racemase from P. putida KT2440 (E.C. 5.1.1.10), overexpressed in E. coli BL21(DE3)) was used for in situ racemization (enzyme concentrations varying from 0 to 1 mg/mL). When preferential crystallization was applied without any enzyme, a total of 31 mg of L ‐Asn monohydrate could be crystallized, before crystal formation of d ‐Asn started. Crystallization experiments accompanied by enzymatic racemization led to a significant increase of crystallized L ‐Asn (198 mg L ‐Asn monohydrate; >92%ee) giving the first experimental proof for this new process concept of dynamic kinetic resolution via preferential crystallization and enzymatic racemization. Measurements of the racemase activity before and after the crystallization process showed no significant differences, which would allow for enzyme recovery and recycling. Biotechnol. The mammalian cell-lines used to produce biopharmaceutical products are known to produce endogenous retrovirus-like particles and have the potential to foster adventitious viruses as well. To ensure product safety and regulatory compliance, recovery processes must be capable of removing or inactivating any viral impurities or contaminants which may be present. Anion exchange chromatography (AEX) is a common process in the recovery of monoclonal antibody products and has been shown to be effective for viral removal. To further characterize the robustness of viral clearance by AEX with respect to process variations, we have investigated the ability of an AEX process to remove three model viruses using various combinations of mAb products, feedstock conductivities and compositions, equilibration buffers, and pooling criteria. Our data indicate that AEX provides complete or near-complete removal of all three model viruses over a wide range of process conditions, including those typically used in manufacturing processes. Furthermore, this process provides effective viral clearance for different mAb products, using a variety of feedstocks, equilibration buffers, and different pooling criteria. Viral clearance is observed to decrease when feedstocks with sufficiently high conductivities are used, and the limit at which the decrease occurs is dependent on the salt composition of the feedstock. These data illustrate the robust nature of the AEX recovery process for removal of viruses, and they indicate that proper design of AEX processes can ensure viral safety of mAb products.  相似文献

Large‐scale manufacture of human embryonic stem cells (hESCs) is prerequisite to their widespread use in biomedical applications. However, current hESC culture strategies are labor‐intensive and employ highly variable processes, presenting challenges for scaled production and commercial development. Here we demonstrate that passaging of the hESC lines, HUES7, and NOTT1, with trypsin in feeder‐free conditions, is compatible with complete automation on the CompacT SelecT, a commercially available and industrially relevant robotic platform. Pluripotency was successfully retained, as evidenced by consistent proliferation during serial passage, expression of stem cell markers (OCT4, NANOG, TRA1‐81, and SSEA‐4), stable karyotype, and multi‐germlayer differentiation in vitro, including to pharmacologically responsive cardiomyocytes. It was the aim of this study to identify risk factors of perinatal mortality (PM) on a large dairy farm in Germany. Four hundred and sixty-three cows were observed continuously around calving. Data such as the lactation number, body condition score, pelvic measurements, duration of second stage of labour (SSL), degree of abdominal press (AP), presentation/position/posture of the calf, calving classification, various measurements of the calves and every action of the barn staff were documented. Calves born dead and those that died within 24 h were classified as cases of PM. Logistic regression was performed to identify risk factors for the dependent variables PM, SSL and AP. 9.7% of all calves born (n = 483) were classified as PM. In the logistic regression model, only SSL and presentation/position/posture of the foetus remained as significant effects for PM. The odds ratio for PM was 0.20, when SSL was ≤120 min, and 0.33, when the calves were born in anterior presentation, upper position and normal posture. With regard to SSL, primipara, insufficient AP, assisted calvings and as a trend, work shift changeover, these were risk factors for a prolonged SSL beyond 120 min. The risk for insufficient AP was lower when the calves were born spontaneously, in anterior presentation and as single calves. In conclusion, the duration of SSL and presentation/position/posture of the calf seemed to be key risk factors for PM. Interventions should be considered when SSL lasts longer than 2 h. Insufficient monitoring around parturition had a negative effect on the duration of SSL and thereby, indirectly on PM.  相似文献

Erdos Larus Relictus Nature Reserve is the only wetland of international importance to protect Larus Relictus in China. From the early part of this century, the impact of water use, vegetation construction and the climate of the catchment area increasing on the wetland of this Nature Reserve has brought about severe water shortage. Until now, no eco-hydrological model at home or abroad integrates the wetland ecological and hydrological processes. This paper considers the relationship between the food chains of the Larus Relictus and environmental elements, and establishes the eco-hydrological processes from Larus Relictus to damselfly + Chironomidae and the salinity and the area of Bojiang Lake. It also establishes a relationship among Larus Relictus breeding, the area of islands outcropped in the levels of the lake and hydrological season based on the relationship between Larus Relictus breeding and environmental elements. The eco-hydrological model is then constructed according to these relationships. In virtue of the constraints of the lake salinity, the suitable area of the lake is 9.01 km2, and 4.88 km2 for the smallest area. While in accordance with the area of islands outcropped needed for Larus Relictus breeding, the lake area is 11.02 km2 in breeding season corresponding to critical high level.  相似文献

High pressure is, at least, as effective as cryoprotective agents (CPAs) and are used for decreasing both homogenous nucleation and freezing temperatures. This fact gives rise to a great variety of possible cryopreservation processes under high pressure. They have not been optimized yet, since they are relatively recent and are mainly based on the pressure–temperature phase diagram of pure water. Very few phase diagrams of biological material are available under pressure. This is owing to the lack of suitable equipment and to the difficulties encountered in carrying out the measurements. Different aqueous solutions of salt and CPAs as biological models are studied in the range of 0°C down to ‐35°C, 0.1 up to 250 MPa, and 0–20% w/w total solute concentration. The phase transition curves of glycerol and of sodium chloride with either glycerol or sucrose in aqueous solutions are determined in a high hydrostatic pressure vessel. The experimental phase diagrams of binary solutions were well described by a third‐degree polynomial equation. It was also shown that Robinson and Stokes' equation at high pressure succeeds in predicting the phase diagrams of both binary and ternary solutions. The solute cryoconcentration and the ice content were calculated as a function of temperature and pressure conditions during the freezing of a binary solution. This information should provide a basis upon which high‐pressure cryopreservation processes may be performed and the damages derived from ice formation evaluated. © 2009 American Institute of Chemical Engineers Biotechnol. 解释群落的物种多样性大小是生态学研究的一个重要的理论和实践问题。人们提出了群落物种多样性的多种假说, Zobel等人提出的种库假说(species pool hypothesis)是生物多样性理论研究的重要发展。该假说认为, 一个群落的物种多样性不仅与环境条件和生态过程(ecological process)(如竞争、捕食)有关, 也受区域种库(regional species pool)的限制。区域种库是指一个地区可进入某一群落的潜在物种数量, 它由地史过程(如冰期、地质年代)和区域过程(物种形成、迁移扩散以及消亡)所决定。按照种库假说, 某一生境类型的面积越大, 地质年代越古老, 物种形成的机会也就越多, 因而能适应和分布于该生境的物种也就越多, 实际群落中的物种丰富度也就越高。种库在空间上主要有两个层次: 区域种库和实际种库, 前者指某一生境所拥有的潜在物种数量, 主要由生物地理过程(biogeographic processes)所决定; 后者则为调查的群落中实际出现的物种数量, 主要由竞争等生态过程和区域种库共同决定。本文对种库假说的基本概念、主要内容、种库确定方法等作了介绍, 并阐述了作者对这些问题的理解和认识。  相似文献
