实验中观察到,用MUG培养基对植物药中的大肠杆菌定量时多发生荧光猝灭现象,影响检测结果。本文对此现象产生的原因与克服方法进行了系统的考察,发现以一种简便的转接方法可排除植物药介质对菌检的干扰。该方法由两组检验系列构成,当怀疑正常稀释系列(第一系列)40h培养液的荧光结果可能因猝灭现象呈假阴性时,立即分别将该系列的1—3号管培养液以0.5ml的接种量转接入新鲜的MUG培养基(第二系列),重新培养24h,荧光猝灭现象即可克服。综合两系列的荧光、产气和吲哚三项生化特征得出检品中大肠杆菌含量。实际应用表明,此法能显著提高使用该培养基时菌检结果的可靠性。 相似文献

The effects of pulsed microwaves (2.45 GHz, 10 μs, 100 pps, SAR: 81.5 kW/kg peak, 81.5 W/kg average) on membrane input resistance and action potential (AP) interval statistics were studied in spontaneously active ganglion neurons of land snails (Helix aspersa), at strictly constant temperature (20.8±.07°C worst case). Statistical comparison with sham-irradiated neurons revealed a significant increase in the mean input resistance of neurons exposed to pulsed microwaves (P ? .05 ). Pulsed microwaves had no visible effect on mean AP firing rate; this observation was confirmed by analysis of interspike intervals (ISIs). Using an integrator model for spontaneously active neurons, we found the net input current to be more variable in neurons exposed to pulsed microwaves. The mean input current was not affected. The standard deviation of ISIs and the autocorrelation of the input current were marginally affected, but these changes were not consistent across neurons. Although the observed effects were less obvious than those reported in other studies, they represent evidence of a direct interaction between neurons and pulsed microwaves, in the absence of macroscopic temperature changes. Continuous culture experiments with identical experimental designs were run with a mixed microbial community of activated sludge origin and an axenic bacterial culture derived from it. Each culture received 2-chlorophenol (2-CP) at a concentration of 160 mg/L as COD and L-lysine at a concentration of 65 mg/L as COD. A factorial experimental design was employed with dilution rate and media composition as the two controlled variables. Three dilution rates were studied: 0.015, 0.0325, and 0.05 h–1. Media composition was changed by adding four biogenic compounds (butyric acid, thymine, glutamic acid and lactose) in equal COD proportions at total concentrations of 0, 34, 225, and 1462 mg/L as COD. The measured variables were the effluent concentrations of 2-CP as measured by the 4-aminoantipyrene test and lysine as measured by the o-diacetylbenzene procedure. The results suggest that community structure and substrate composition play important roles in the response of a microbial community to mixed substrates. The addition of more biogenic substrates to the axenic culture had a deleterious effect on the removal of both lysine and 2-CP, although the effect was much larger on lysine removal. In contrast, additional substrates had a positive effect on the removal of 2-CP by the mixed community and much less of a negative effect on the removal of lysine. The dilution rate at which the cultures were growing had relatively little impact on the responses to the additional substrates.Abbreviations COD

In a phytotron experiment four rice varieties (Pokkali, IR 28, IR 50, IR 31785-58-1-2-3-3) grown in individual pots were subjected to low (40/55% day/night) and high (75/90%) air humidity (RH), while soil salinity was gradually increased by injecting 0, 30, 60 or 120 mM NaCl solutions every two days. Bulk root and stem base water potential (SWP), abscisic acid (ABA) content of the xylem sap and stomatal resistance (rs) of the youngest fully expanded leaf were determined two days after each salt application. The SWP decreased and xylem ABA and rs increased throughout the 8 days of treatment. The effects were amplified by low RH. A chain of physiological events was hypothesized in which high soil electric conductivity (EC) reduces SWP, followed by release of root-borne ABA to the xylem and eventually resulting in stomatal closure. To explain varietal differences in stomatal reaction, supposed cause and effect variables were compared by linear regression. This revealed strong differences in physiological reactions to the RH and salt treatments among the test varieties. Under salt stress roots of IR 31785-58-1-2-3-3 produced much ABA under low RH, but no additional effect of low RH on rs could be found. By contrast, Pokkali produced little ABA, but rs was strongly affected by RH. RH did not affect the relationships EC vs. SWP and SWP vs. ABA in Pokkali, IR 28, and IR 50, but the relationship ABA vs. rs was strongly affected by RH. In IR 31785-58-1-2-3-3 RH strongly affected the relationship SWP vs. ABA, but had no effect on ABA vs. rs and EC vs. rs. The metabolism of [3H]formate has been examined in etiolated and greening leaves of barley (Hordeum vulgare), dwarf bean (Phaseolus vulgarls), broad bean (Vicia faba) and corn (Zea mays). Tritium was extensively incorporated by primary leaves incubated for 20-min periods in light or dark. The organic acids and free amino acids were the principal products of formate metabolism but these and other products were more heavily labelled in green tissues. Time course experiments with barley leaves revealed a rapid labelling of serine, accompanied by increasing amounts of 3H in glycine and aspartate as the feeding period was extended. These amino acid products were formed throughout a 4-day greening period with an approximate doubling in total incorporation being due to large accumulations of tritiated glycine and aspartate. The involvement of tetrahydrofolate-dependent reactions in formate metabolism was indicated by inhibition of [14C] and [3H]formate incorporation by the folate antagonist, aminopterin. Labelling of glycine and serine was also strongly inhibited (up to 90%) when the leaves were incubated with increasing concentrations of isonicotinylhydrazide. 相似文献
