The lateral lens eye of adult Craterostigmus tasmanianus Pocock, 1902 (a centipede from Australia and New Zealand) was examined by light and electron microscopy. An elliptical, bipartite eye is located frontolaterally on either side of the head. The nearly circular posterior part of the eye is characterized by a plano-convex cornea, whereas no corneal elevation is visible in the crescentic anterior part. The so-called lateral ocellus appears cup-shaped in longitudinal section and includes a flattened corneal lens comprising a homogeneous and pigmentless epithelium of cornea-secreting cells. The retinula consists of two kinds of photoreceptive cells. The distribution of the distal retinula cells is highly irregular. Variable numbers of cells are grouped together in multilayered, thread-like unions extending from the ventral and dorsal margins into the center of the eye. Around their knob-like or bilobed apices the distal retinula cells give rise to fused polymorphic rhabdomeres. Both everse and inverse cells occur in the distal retinula. Smaller, club-shaped proximal retinula cells are present in the second (limited to the peripheral region) and proximal third of the eye, where they are arranged in dual cell units. In its apical region each unit produces a small, unidirectional rhabdom of interdigitating microvilli. All retinula cells are surrounded by numerous sheath cells. A thin basal lamina covers the whole eye cup, which, together with the distal part of the optic nerve, is wrapped by external pigment cells filled with granules of varying osmiophily. The eye of C. tasmanianus seemingly displays very high complexity compared to many other hitherto studied euarthropod eyes. Besides the complex arrangement of the entire retinula, the presence of a bipartite eye cup, intraocellar exocrine glands, inverse retinula cells, distal retinula cells with bilobed apices, separated pairs of proximal retinula cells, medio-retinal axon bundles, and the formation of a vertically partitioned, antler-like distal rhabdom represent apomorphies of the craterostigmomorph eye. These characters therefore collectively underline the separate position of the Craterostigmomorpha among pleurostigmophoran centipedes. The remaining retinal features of C. tasmanianus agree with those known from other chilopod eyes and, thus, may be considered plesiomorphies. Characters like the unicorneal eye cup, sheath cells, and proximal rhabdomeres with interdigitating microvilli were already present in the ground pattern of the Pleurostigmophora. Other retinal features were developed in the ancestral lineage of the Phylactometria (e.g., large elliptical eyes, external pigment cells, polygonal sculpturations on the corneal surface). Starting from readily available protected 6-tosylates of D-glucose and D-mannose in both their pyranoside and furanoside forms as well as 6-tosylates of alpha-D-galactopyranose, the corresponding primary succinimido derivatives were obtained in good yield by nucleophilic displacement with potassium succinimide. These imido sugars were photochemically transformed into hexahydroazepindione derivatives such as by means of a Norrish type II reaction. As expected, the intramolecular alkylation proceeded via an 1,6 H-abstraction leading to a stabilized diradical. The regiochemistry of the photoreaction was controlled by configurational, conformational and electronic features and was sometimes influenced by the protecting groups. A trimeric protein phosphatase 2A (PP2A(T55)) composed of the catalytic (PP2Ac), structural (PR65/A), and regulatory (PR55/B) subunits was isolated from rabbit skeletal muscle by thiophosphorylase affinity chromatography, and contained two additional proteins of 54 and 55 kDa, respectively. The 54 kDa protein was identified as eukaryotic translation termination factor 1 (eRF1) and as a PP2A interacting protein. The 55 kDa protein is now identified as nucleoredoxin (NRX). The formation of a complex between GST-NRX, PP2A(C) and PP2A(D) was demonstrated by pull-down experiments with purified forms of PP2A, and by immunoprecipitation of HA-tagged NRX expressed in HEK293 cells complexed endogenous PP2A subunits. Analysis of PP2A activity in the presence of GST-NRX showed that NRX competed with polycations for both stimulatory and inhibitory effects on different forms of PP2A. 相似文献

We show that the five subunits of a gamma-aminobutyric acid type A receptor (GABA(A) receptor) can be concatenated to yield a functional receptor. This concatenated receptor alpha(1)-beta(2)-alpha(1)-gamma(2)-beta(2) has the advantage of a known subunit arrangement. Most of its functional properties are not significantly different from a receptor formed by individual subunits. Extent of expression amounted to about 40% of that of non-concatenated receptors in Xenopus oocytes, after injection of oocytes with comparable amounts of cRNA coding for concatenated and non-concatenated receptors. Receptor-type protein tyrosine phosphatases (RPTPs) are considered to transduce extracellular signals across the membrane through changes in their PTP activity, however, our understanding of the regulatory mechanism is still limited. Here, we show that pleiotrophin (PTN), a natural ligand for protein tyrosine phosphatase receptor type Z (Ptprz) (also called PTPzeta/RPTPbeta), inactivates Ptprz through oligomerization and increases the tyrosine phosphorylation of substrates for Ptprz, G protein-coupled receptor kinase-interactor 1 (Git1) and membrane associated guanylate kinase, WW and PDZ domain containing 1 (Magi1). Oligomerization of Ptprz by an artificial dimerizer or polyclonal antibodies against its extracellular region also leads to inactivation, indicating that Ptprz is active in the monomeric form and inactivated by ligand-induced oligomerization. 相似文献

Several biochemical reactions and processes of cell biology are compartmentalized in the endoplasmic reticulum (ER). The view that the ER membrane is basically a scaffold for ER proteins, which is permeable to small molecules, is inconsistent with recent findings. The luminal micro-environment is characteristically different from the cytosol; its protein and glutathione thiols are remarkably more oxidized, and it contains a separate pyridine nucleotide pool. The substrate specificity and activity of certain luminal enzymes are dependent on selective transport of possible substrates and co-factors from the cytosol. ZntA is a P-type ATPase which transports Zn2+, Pb2+ and Cd2+ out of the cell. Two cysteine-containing motifs, CAAC near the N-terminus and CPC in transmembrane helix 6, are involved in binding of the translocated metal. We have studied these motifs by mutating the cysteines to serines. The roles of two other possible metal-binding residues, K693 and D714, in transmembrane helices 7 and 8, were also addressed. The mutation CAAC → SAAS reduces the ATPase activity by 50%. The SAAS mutant is phosphorylated with ATP almost as efficiently as the wild type. However, its phosphorylation with Pi is poorer than that of the wild type and its dephosphorylation rate is faster than that of the wild type ATPase. The CPC → SPS mutant is inactive but residual phosphorylation with ATP could still be observed. The most important findings of this work deal with the prospective metal-binding residues K693 and D714: the substitution K693N eliminates the Zn2+-stimulated ATPase activity completely, although significant Zn2+-dependent phosphorylation by ATP remains. The K693N ATPase is hyperphosphorylated by Pi. ZntA carrying the change D714M has strong metal-independent ATPase activity and is very weakly phosphorylated both by ATP and Pi. In conclusion, K693 and D714 are functionally essential and appear to contribute to the metal specificity of ZntA, most probably by being parts of the metal-binding site made up by the CPC motif. 相似文献

The activity and localization of fructose-1,6-bisphosphatase (FBPase; EC 3.1.3.11) in blood leukocytes of patients with type 1 diabetes mellitus and healthy adults were investigated immunocytochemically. The amount of polymorphonuclear (PMN) and mononuclear (MN) cells with positive FBPase immunocytochemical reaction was 57% and 68%, respectively, in pathological, and 38% and 42%, respectively, in healthy donors. Results of light microscopic investigations were confirmed by measurements of FBPase activity following lysis of PMN and MN cells. The enzyme activity of PMN and MN leukocytes was higher in diabetes mellitus than in healthy adults, by 30% and 127%, respectively. Using immunocytochemistry together with electron microscopy, FBPase was detected not only in the cytoplasm but also in the nucleus of leukocytes of both patients with insulin-dependent diabetes mellitus and healthy donors. 相似文献
